Book excerpt: Although upstaged by the tragic appearance of the human immunodeficiency virus, herpes simplex viruses (HSV) types 1 and 2 continue to be major human pathogens against which we lack acceptable vaccines or other means of immunological control. The virus is large and complex, coding for 70 or more proteins. Although many mysteries remain to be unraveled, our knowledge base regarding genomic organization, gene expression and regulation, pathogenesis, and immune recog nition of component parts is quite considerable. Indeed, meet ings devoted entirely to herpesviruses are conspicuous by their frequency and excellent, yet sometimes exclusive, attendance. The purpose of this volume is to compile in a single book a series of reviews by leading investigators that deal with various aspects of virus-host interactions and which hopefully will pro vide clues as to how to best manage HSV from an immunobio logical perspective. Ultimately, one anticipates that a full under standing of virus-host interaction will lead to strategies useful for the prevention and control of HSV. The state of current progress with conventional vaccines is presented, as is a chapter on intracellular immunization. This latter novel approach to virus infections comes at approximately the bicentenary of Jenner's introduction of a successful conventional immunization strategy.

The objective of much of the current research on herpesviruses is directed toward an understanding of the molecular mechanisms involved in initiation of infection, establish ment and termination of latent state, virus multiplication, and the destruction of cells which ultimately is the basis of the diseases caused by these viruses. At no time during the past 80 years, since members of the herpesvirus family were first discovered, has there been so much progress in our understanding of the biology of these viruses as in the past few years. Along with the development of a greater understanding of the molecular biol ogy of the well-known herpesviruses we have witnessed the isolation of new human herpes viruses.

We sensed that the fields of microbiology and im munolgy, which had diverged as each was focusing on its individual search, were coming together. An inten sive examination of the multitude of primitive or more recently developed host defense mechanisms to determine their relative contribution to man's resistance to a given infectious agent appeared to us to be of crucial basic· and practical interest. Many immune mechanisms studied in animals were being explored in humans and it appeared timely to focus particularly on what was known about man's resistance to infectious agents, correlating this information with lessons learned from relevant experiments in animal models.

It is certainly not the size of their genomes or the individual features of their reproductive cycle, although in toto striking features that are exclusive to the herpesviruses abound. Unquestionably, the pre-eminent feature is the relationship of herpes viruses with their natural hosts. As described in preceding volumes, all herpesviruses seem to be able to colonize and to remain in a latent, nonproductive form for life of their hosts. Once established in the host, the relationship is best described as that of an armed truce. What happens when this truce breaks down or when the host encounters the virus for the first time is the subject of this volume.
