Book Quantitation of Amino Acids and Amines by Chromatography

Download or read book Quantitation of Amino Acids and Amines by Chromatography written by Ibolya Molnár-Perl and published by Elsevier. This book was released on 2005-06-27 with total page 669 pages. Available in PDF, EPUB and Kindle. Book excerpt: Quantitation of Amino Acids and Amines by Chromatography: Methods and Protocols is intended to serve as a ready-to-use guide for the identification and quantification of amino acids and amines in various matrices, providing an overview on the theory and protocol of available methods. It presents chromatograms with exact elution programs enabling visual analysis and compares the advantages-disadvantages of various chromatographic techniques. In accordance with the chronological order of the development of chromatographic methods, different techniques are discussed: The possibilities of gas chromatography (GC), followed by those of the high performance liquid chromatography (HPLC) and the most recent techniques capillary electrophoresis (CE), capillary, electrochromatography (CEC).The characteristics of the given chromatographic procedure, relating to the topic in question, are classified according to the preliminary preparation/derivatization process(es), which means the simple methods, suitable for the analysis of the selected compound(s) in natural form, are followed by various derivatization proposals. Detailed protocols provide the reader with guidance in beginning tasks and on how to improve current methods. This book appeals to a wide audience and is recommended for those looking towards the wider reaches of identification and quantification of amino acids and amines.* Provides a systematic, and comprehensive summary of chromatographic techniques and derivatization processes* Compares advantages/disadvantages of various chromatographic techniques* Readers can undertake practical tasks using detailed protocols given in the book

Book Methods of Protein Microcharacterization

Download or read book Methods of Protein Microcharacterization written by John E. Shively and published by Springer Science & Business Media. This book was released on 2008-02-22 with total page 458 pages. Available in PDF, EPUB and Kindle. Book excerpt: Milestones in the techniques and methodology of polypeptide structure determination include the determination of the sequence of insulin by Sanger in 1951 (I) and the introduction of the repeti tive degradation of proteins with phenylisothiocyanate by Edman in 1959 (2). The automation of Edman chemistry (3) played a major role in the determination of polypeptide structures. Important modifications of Edman chemistry include the solid-phase approach by Laursen in 1971 (4) and the use of modified Edman reagents such as 4-N, N-dimethylaminoazobenzene-4'-isothiocy- ate (DABITC) for manual sequencing by Chang et al. (5) in 1976. A second major breakthrough in the analysis of polypeptides was automated amino acid analysis described by Spackman et al. in 1958 (6). However, during the period from 1975 to 1980, it became increasingly clear that the amount of material required for struc tural analysis was more than could be easily isolated for the vast majority of proteins. The field was criticized for its lack of sensitive techniques for the analysis of growth factors, immune modulators, membrane receptors, and peptide hormones. In addition, very little had been done to modernize and improve the original instruments introduced in the mid-1960s. The first indications of improved instrumentation for Edman chemistry came from Wittmann-Liebold's laboratory (7), followed by the introduction of a "micro" sequencer by Hunkapiller and Hood in 1978 (8). The movement toward improved instrumentation culminated in the "gas"--Phase sequencer of Hewick et al. (9) in 1981

Book Thin Layer Chromatography

Download or read book Thin Layer Chromatography written by Hans Rudolf Bolliger and published by Springer. This book was released on 1965 with total page 584 pages. Available in PDF, EPUB and Kindle. Book excerpt: Thin-layer chromatography has become so widely known in the space of a few years that it has proved necessary to gather into book form and thus make generally accessible the experimental material previously only available in isolated publications. As thin-layer chromatography can be used both for organic and inorganic matter as well as on quantities ranging from the nanogram to the microgram, it is impossible for anyone individual to possess sufficient laboratory experience or overall knowledge to produce a practical handbook that will be of real assistance to be ginner and specialist alike. For this reason, an international group was formed, who made it their task to produce the best possible treatise. In view of the present stage of development reached by thin-Iaycr chromatography, it seems specially apt that the authors should include yet unpublished work of their own. As thin-layer chromatography is used in many different fields in natural science and medicine, the kind of brief description of materials intelligible only to the expert has been avoided. The short guides to the chemical properties of the groups to be separated, their names, and relevant bibliographic details should facilitate introductory studies arid make possible a close acquaintance with the material in hand. It also seemed advisable to give brief details of the analytical classification of material, which is so often necessary. Although the classification used may appear unusual, it is in fact pre-eminently suitable to thin-layer chromatography.
