Book excerpt: It is only relatively recently that neurochemists and neurobiologists have shown appreciable interest in the class of macromolecules now generally re ferred to as complex carbohydrates, although gangliosides were, of course, first identified and studied in brain. The glycosaminoglycans fell chiefly within the province of connective-tissue biochemists, and earlier informa tion concerning the structure and metabolism of glycoproteins was largely limited to the more accessible glycoproteins and oligosaccharides (such as those found in plasma, milk, and urine), or ones which are relatively simple to prepare in a soluble and manageable form. Techniques were later devised for the isolation and purification of tightly bound membrane glycoproteins, where initial studies concentrated mainly on the erythrocyte, for which large amounts of a single cell population are available. Because of the structural complexity of nervous tissue and the large numbers, low concentrations, and membrane-bound form of many of its complex carbohydrates, progress has occurred more slowly in this area.

Book excerpt: The purposes of this senes were discussed in the preface to Volume I: to present "a range of methods . . . from the physical to the physiological . . . in sufficient detail for the reader to use them in his laboratory" and also to describe "the theoretical backgrounds of the methods and their limita tions in membrane biology" so that the reader will be enabled "to evaluate more critically and to understand more fully data obtained by methods foreign to [his] usual experiences. " The chapter by Lee, Birdsall, and Metcalfe with which Volume 2 begins accomplishes these twin goals with a thorough description of the application of nuclear magnetic relaxation measurements to membrane biology together with a lucid and succinct integration of the results of such studies into present concepts of the organi zation of membrane lipids. This then permits speculation on the physical basis of membrane permeability. The powerful tool of NMR spectroscopy will have even fuller application with the development of techniques, al ready partially exploited, for l3C-Iabeling of specific carbon atoms in lipid molecules and with extension of the observations to membrane proteins. The following two chapters, by Glick and by Laine, Stellner, and Hako mori, describe the isolation and characterization of membrane glycoproteins and membrane glycolipids, respectively.

One paper investigates the stereochemical assignment in some 2-thio sugar derivatives and the stereochemical lability of 2-thioaldoses in basic media. Another paper examines the synthetic chemistry of oligosaccharides, particularly, the synthesis of trisaccharides, one of which is a repeating unit of O-antigenic polysaccharide from Salmonella anatum. One paper notes that plant acidic polysaccharides should be classified in terms of their basal core structures to enable groupings and comparisons of glycuronoglycans isolated from various sources. Another paper shows that the structure of the carbohydrate moiety of glycoproteins points to the biological role of carbohydrate moieties as possible cell recognition signals. This collection can prove valuable for bio-chemists, cellular biologists, micro-biologists, and developmental biologists.

Book excerpt: A comprehensive review of the Golgi apparatus and its functioning would require a multi-volume publication and not a monograph and it would be so repetitious as to discourage the reader. The requirement at this stage is for a reinterpretation of the character and functioning of this organelle since the last major interpretations have concentrated on its role in secretion and it has now been shown to be a component of essentially all cells whether or not they have been traditionally emphasized as secreting cells. As a conse quence the efforts have been placed on the common characteristics of the organelle, a postulate concerning its functioning in cells generally, and the details of variations where these seem important. The major acknowledgment of assistance in compiling the material must go to the investigators whose contributions, sometimes positive and sometimes of a character to spur additional investigations, allowed the development of this postulate. The paper has been prepared with the detailed assistance of Dr. MARIANNE DAUWALDER who, by her own studies and her insight into the significance of other studies, has been a working partner of many years in the development of a general hypothesis and whose knowledge of investiga tions of the Golgi apparatus is great enough to let her call attention to instances of support and contention with the general functional hypothesis that has been involved.
