Book Fluorescence Fluctuation Spectroscopy and Microscopy

Download or read book Fluorescence Fluctuation Spectroscopy and Microscopy written by Syuan-Ming Guo and published by . This book was released on 2016 with total page 139 pages. Available in PDF, EPUB and Kindle. Book excerpt: Fluorescence fluctuation spectroscopy and microscopy have been powerful tools for studying molecular organization and dynamics in cells. Fluorescence Correlation Spectroscopy (FCS) has been widely applied to probing molecular dynamics in live cells with single molecule sensitivity and the ability to resolve local molecular concentrations, aggregation states, and transport mechanisms. Despite the broad utility of FCS, interpretation of cellular FCS data is often confounded by the heterogeneity in the underlying biological process and the low signal-to-noise ratio. Systematic data evaluation and interpretation become even more challenging for imaging FCS, where hundreds to thousands of FCS curves are generated in a single measurement. This thesis presents an objective Bayesian inference procedure for testing multiple competing FCS models. Bayesian inference determines model probabilities by considering the probability distributions over the full range of parameter values, thereby naturally penalizes model complexity and prevents over-fitting. We applied this procedure to imaging FCS data in order to resolve hIAPP-induced microdomain spatial organization and temporal dynamics in the cell membrane. Our analysis resolved the temporal evolution of multiple diffusing species in the spatially heterogeneous cell membrane, lending support to the "carpet model" for the association mode of hIAPP aggregates with the plasma membrane. Finally, we presented a fluctuation-based microscopy approach, Points Accumulation for Imaging in Nanoscale Topography (PAINT), that enables highly multiplexed super-resolution imaging of synaptic proteins. We employed DNA-PAINT to resolve nano-scale organization of seven targets simultaneously including synaptic proteins and cytoskeletal markers. These approaches demonstrated the broad applicability of fluorescence fluctuation spectroscopy and microscopy in resolving molecular dynamics and organization in cells.

Book Fluorescence Fluctuation Spectroscopy FFS

Download or read book Fluorescence Fluctuation Spectroscopy FFS written by Sergey Y. Tetin and published by . This book was released on 2013 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: Fluorescence Fluctuation Spectroscopy (FFS) is a growing family of methods for studying molecular motions and interactions. Introduced as Fluorescence Correlation Spectroscopy (FCS) 40 years ago, this elegant technique has evolved into a large group of powerful tools of modern fluorescence spectroscopy. FFS has found numerous applications in biophysical and biochemical studies of proteins, nucleic acids, and membranes. In combination with fluorescence imaging microscopy, FFS provides unique opportunities for characterizing dynamic processes in living cells and has been increasingly used by cell and systems biologists. Significant progress in the technology was achieved in the past decade, and the number of new FFS methods is still growing. The attraction of FFS relates to fact that it is based on statistical analysis of the fluctuations occurring at the molecular level. Therefore, these methods can resolve molecular populations and provide additional information about biological systems. The chapters in this two-volume publication are written by the authors and first-hand users of the methods under discussion. Part A is dedicated predominately to FFS methods. It contains well-established and also recently introduced experimental techniques and data analysis algorithms. Part B continues with FFS methods and also includes various FFS applications in vivo and in vitro. Here the reader will find specific examples and detailed experimental protocols developed for several biological systems. The purpose of these chapters is to show the power and potentials of the technology in biological research
