Book excerpt: In mice infected with Listeria monocytogenes, a fish oil diet, rich in n-3 polyunsaturated fatty acids (n-3 PUFA), is associated with reduced survival and clearance of the bacteria from host tissues. The objective of this investigation was to determine to what extent dietary n-3 PUFA affect the development of adaptive immune responses against L. monocytogenes . Weanling female BALB/c mice were fed one of two experimental diets that differed only in that one was devoid of n-3 PUFA (i.e., lard) and the other was high in n-3 PUFA (i.e., menhaden fish oil). Mice were maintained on experimental diets for 28 d prior to immune challenge. We observed no impairment in the generation of immunological memory associated with n-3 PUFA feeding, even though bacterial clearance and survival were significantly reduced during the primary response. Further, the clonal expansion of Ag-specific CD8 + T cells was not altered by dietary n-3 PUFA. The absolute number of Listeria -specific CD4 + and CD8 + effector and memory/effector cells in the spleen were not affected, but the frequency of memory/effector cells was reduced by 50% in the n-3 PUFA-fed mice. Dietary n-3 PUFA did not significantly impair the ability of Ag-experienced splenocytes to adoptively transfer immunity to naive recipients on a low n-3 PUFA diet. Collectively, these results demonstrate that despite profoundly impairing host resistance during a primary infection with L. monocytogenes, the generation of adaptive immune responses does not appear to be compromised by n-3 PUFA.

Severe malnutrition can cause significant alteration in immune response, but even subclinical deficits may be associated with an impaired immune response, and an increased risk of infection. Infectious diseases have accounted for more off-duty days during major wars than combat wounds or nonbattle injuries. Combined stressors may reduce the normal ability of soldiers to resist pathogens, increase their susceptibility to biological warfare agents, and reduce the effectiveness of vaccines intended to protect them. There is also a concern with the inappropriate use of dietary supplements. This book, one of a series, examines the impact of various types of stressors and the role of specific dietary nutrients in maintaining immune function of military personnel in the field. It reviews the impact of compromised nutrition status on immune function; the interaction of health, exercise, and stress (both physical and psychological) in immune function; and the role of nutritional supplements and newer biotechnology methods reported to enhance immune function. The first part of the book contains the committee's workshop summary and evaluation of ongoing research by Army scientists on immune status in special forces troops, responses to the Army's questions, conclusions, and recommendations. The rest of the book contains papers contributed by workshop speakers, grouped under such broad topics as an introduction to what is known about immune function, the assessment of immune function, the effect of nutrition, and the relation between the many and varied stresses encountered by military personnel and their effect on health.

Many parts of the world in which common infectious diseases are endemic also have the highest prevalence of trace metal deficiencies or rising rates of trace metal pollution. Infectious diseases can increase human susceptibility to adverse effects of metal exposure (at suboptimal or toxic levels), and metal excess or deficiency can increase the incidence or severity of infectious diseases. The co-clustering of major infectious diseases with trace metal deficiency or toxicity has created a complex web of interactions with serious but poorly understood health repercussions, yet has been largely overlooked in animal and human studies. This book focuses on the distribution, trafficking, fate, and effects of trace metals in biological systems. Its goal is to enhance our understanding of the relationships between homeostatic mechanisms of trace metals and the pathogenesis of infectious diseases. Drawing on expertise from a range of fields, the book offers a comprehensive review of current knowledge on vertebrate metal-withholding mechanisms and the strategies employed by different microbes to avoid starvation (or poisoning). This report fulfills the final reporting requirement of the grant, and presents a summary of activities for the grant period from December 1, 1994 through May 31, 1999. During this grant period, the CMNR has met from three to six times each year in response to issues that are brought to the committee through the Military Nutrition and Biochemistry Division of the U.S. Army Research Institute of Environmental Medicine at Natick, Massachusetts, and the Military Operational Medicine Program of USAMRMC at Fort Detrick, Maryland. The CMNR has submitted five workshop reports (plus two preliminary reports), including one that is a joint project with the Subcommittee on Body Composition, Nutrition, and Health of Military Women; three letter reports, and one brief report, all with recommendations, to the Commander, U.S. Army Medical Research and Materiel Command, since September 1995 and has a brief report currently in preparation. These reports are summarized in the following activity report with synopses of additional topics for which reports were deferred pending completion of military research in progress. This activity report includes as appendixes the conclusions and recommendations from the nine reports and has been prepared in a fashion to allow rapid access to committee recommendations on the topics covered over the time period.
