The Electrophoretic technique is almost free from these vitiating factors and results are fairly reliable. Its further extension that these studies can be used to reduced the level of these metal ions in biological system. These studies may also be used for the resolution of racemic mixture of penicillamine and other amino acids.

Book excerpt: Chapter 1 introduces the theory and principles behind capillary electrophoresis separations and the instrumental requirements. An overview of the speciation of metal complexes by capillary electrophoresis is given, detailing the various situations reported in the literature. A summary of the recent advances in the area of microchip electrophoresis is also included, focusing in particular on the challenge of the metal complexes detection in miniaturised systems. Finally, the concept of equilibrium in electrophoretic systems is highlighted, discussing the determination of liabile complexes during electrophoretic processes. Chapter 2 reports one of the first examples of separation by microchip electrophoresis of individual species in equilibrium within a single metal-ligand system. Initial work showed that the effective separation of the three stoichiometries of nickel-bathophenanthroline disulfonic acid (NiBPS) complexes, i.e. [Ni(BPS)1, Ni(BPS)2 and Ni(BPS)3] could be achieved by capillary electrophoresis in less than 5 min. In chapter 3, the research focused on the study of labile copper-histidine equilibrium. A CE method was developed for the separation of copper-histidine complexes and free histidine. Preliminary work on solid phase extraction and molecular imprinted polymer of the metal complex is also presented. Chapter 4 outlines the use of microchip electrophoresis as an efficient tool for the analysis of labile complexes. A 2.5 cm channel was used for the separation of copper-histidine complex and free histidine, where no decomplexation was observed and therefore the exact distribution could be assessed. However, the labile character of copper comples prevented the separation of the two stoichiometries of copper histidine systems. In chapter 5, capillary electrophoresis was explored as a man for the determination of pKa values of drugs, outlining its advantages over traditional techniques. A linear model for the analysis of monoprotic molecules was applied to two acidic (nicotinic acid, naproxen), two basic (adenine and probranolol) and two amphotheric drugs (labetalol, pyridoxine) and compared to values reported previously in literature in order to confirm its validity. This model was then successfully applied to labile drugs, pyrilamine and procaine. Since capillary electrophoresis allows the use of minimal amounts of analytes, the utility of this method was extended to the analysis of synthetically limited non-natural amino acids and novel Sulindac analogues.

Book excerpt: Although not the only volume in this series in which lipids are discussed, the present volume is devoted entirely to methods for the study of membrane lipids. Even now, when membrane proteins are properly receiving so much attention, this emphasis on membrane lipids is appropriate. Essentially all of the phospholipids and sterols of cells are in membranes. Moreover, although membrane proteins are certainly of utmost importance, the more we learn about the functional properties of membrane proteins, the more we appreciate the unique features of phospholipids, without which biological membranes would be impossible. The hydrophobic-hydrophilic duality of phospholipids allows, indeed requires, their association, in an aqueous environment, into an essentially two-dimensional membrane-only molec ularly thick in one dimension but relatively infinite in the other two; a structure composed of small molecules, not covalently linked, and therefore, infinitely mobile and variable, but yet a structure with great stability and one largely impermeable to most biomolecules. These membrane-forming properties are shared by many amphipathic polar lipids-phospholipids, glycolipids, and sphingolipids-that differ significantly from each other in the nature of their polar head groups and their fatty acids. These variations in structure allow a range of specific interactions among membrane lipids and between lipids and proteins and also provide for membranes of variable, but controlled, fluidity. In this way, phospholipids provide an appropriate milieu for functional membrane proteins and also significantly modulate their catalytic activities.

These questions have been asked for over one hundred years about simple and complex chemical systems, and the answers constitute the macroscopic reaction mechanism. In Determination of Complex Reaction Mechanisms authors John Ross, Igor Schreiber, and Marcel Vlad present several systematic approaches for obtaining information on the causal connectivity of chemical species, on correlations of chemical species, on the reaction pathway, and on the reaction mechanism.Basic pulse theory is demonstrated and tested in an experiment on glycolysis. In a second approach, measurements on time series of concentrations are used to construct correlation functions and a theory is developed which shows that from these functions information may be inferred on the reaction pathway, the reaction mechanism, and the centers of control in that mechanism.
