Book excerpt: Intake of polyunsaturated fatty acids (PUFA) has changed drastically in the past century in the American diet has received attention due to potential effects on chronic inflammation-related metabolic diseases. In this project, the effects of dietary PUFA content and the n-6 to n-3 ratio on inflammatory responses in the acute and chronic inflammation models were evaluated. The PUFA content was modified on a Western diet background to deliver both n-6 and n-3 intakes at the 10th and 90th percentile of the population in the United States, and models of acute and chronic inflammation were tested in mice model. The experimental PUFA diets had a modest effect on the response to lipopolysaccharide (LPS) in the acute inflammation model. A high n-6 to n-3 ratio promoted lipogenesis. In addition, high n-6 PUFA enhanced the inflammatory responses via the NF-ÎðB pathway. In chronic inflammation model, low-grade inflammatory stress was induced by osmotic mini-pump delivery of LPS. A high n-6 intake increased glucose intolerance. On the other hand, high n-6 intake promotes cholesterol ester accumulation in both acute and chronic inflammation models.

Book excerpt: Enteral nutrition with eicosapentaenoic acid (EPA; 20:5 n-3) and [gamma]-linolenic acid (GLA; 18:3 n-6) decreased pulmonary inflammation by reducing neutrophil counts and chemotactic factors in bronchoalveolar lavage fluid during acute respiratory distress syndrome (ARDS). We hypothesize that the antiinflammatory effects of EPA and GLA may be due, in part, to induction of neutrophil apoptosis. Neutrophil apoptosis is an important physiological process in the resolution of pulmonary inflammation. Furthermore, pro-inflammatory eicosanoids formed from arachidonic acid (AA) by lipoxygenase (LO) and cyclooxygenase (COX) pathways have been shown to inhibit apoptosis in certain cell types. The purpose of this dissertation is to determine whether EPA and GLA, alone or in combination, trigger apoptotic cell death in the human promyelocytic leukemia HL-60 cell line. This dissertation will also determine whether inhibition of LO and COX increases apoptosis with AA, the combination of EPA and GLA, and EPA in HL-60 cells, in vitro. Finally, this dissertation will determine the apoptotic gene profile of EPA/GLA-treated HL-60 cells. Study 1, HL-60 cells were incubated with 10, 20, 50, and 100 [mu] M EPA, GLA or various combinations of EPA and GLA for 2, 4, 8, 12 and 24 hours. Oleic acid (18:1 n-9) was used as a fatty acid control. Flow cytometry using dualstaining with propidium iodide and annexin V-FITC assessed apoptosis, necrosis and viability. Apoptosis was verified by DNA fragmentation as assessed by agarose gel electrophoresis. EPA, GLA, and various combinations of EPA and GLA significantly induced apoptosis and reduced cell viability in HL-60 cells. Viability was significantly reduced to the same extent with the combination of 50 [mu] M EPA\20 [mu] M GLA as compared with 100 [mu] M EPA. These data indicate that EPA and GLA, alone or in combination, reduce cell survival by induction of apoptosis. Thus, induction of apoptosis by select dietary n-3 (EPA) and n-6(GLA) polyunsaturated fatty acids may be the mechanism of reduces pulmonary inflammation in ARDS.

The conference had two objectives: (1) to review the research data on the health effects of polyunsaturated fatty acids in seafoods in terms of the impact of omega-3 fatty acids on eicosanoid formation, thrombosis and inflammation, and the role of docosahexaenoic acid in membrane function and metabolism, and (2) to develop a research agenda to determine the spectrum of the health effects of polyunsaturated fatty acids of seafood origin in the American diet. The 1985 conference established the fact that omega-3 fatty acids of marine origin - eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) - play important roles in prostaglandin metabolism, thrombosis and atherosc1erosis, immunology and inflammation, and membrane function. In response to the conference recommendations, the Congress of the United States provided special funding for the establishment of a "test materials 1aboratory" within the US Department of Commerce to produce under documented quality contr01 the types and quanti ties of omega-3 te~3t materials required by biomedica1 researchers. The forms of test materials to be produced include refined fish oil, polyunsaturated fatty acid enhanced triglycerides, concentrates of esters of fatty acids, purified omega-3 fatty acids, and omega-3 mono-, di- and tri~lyceride mixtures.

Book excerpt: There are two essential fatty acids for humans, a-linolenic acid (ALA) and linoleic acid (LA), which should be taken from foods to maintain health. Once incorporated into cells, ALA and LA, which are omega-3 poly unsaturated fatty acid (n-3 PUFA) and n-6 PUFA respectively, undergo elongation and desaturation to generate longer and more unsaturated fatty acids influencing inflammation and immunological responses. Numerous studies showed a dietary reduction of n-6/n-3 PUFA ratio improves cardiovascular health, inflammation, and insulin resistance. Fish oil, the main resource for n-3 PUFA, is shown to increase these health benefits. In our lab, we investigated the efficacy of ALA-enriched butter (n3Bu) as an alternative to fish oil for n-3 PUFA. n3Bufed mice showed increased bioconversion of ALA to long-chain n-3 PUFA (LC n-3 PUFA) and attenuated high fat (HF) diet-induced insulin resistance and inflammation. Besides, these health benefits, n-3 PUFA is shown to improve obesity and its related diseases by regulating lipid metabolism in both white adipose tissue (WAT) and brown adipose tissue (BAT). Fish oil abundant with n-3 PUFA promotes BAT formation and increases its thermogenic activity in cold acclimation. However, the impact of ALAenriched agricultural products on the BAT function is unknown. In this study, we investigated the effect of ALA-biofortified butter (n3Bu) on lipid metabolism and thermogenic functions in BAT. Intake of n3Bu significantly reduced the whitening of BAT and increased the thermogenesis in response to acute-cold treatment. Consumption of n3Bu promoted bioconversion of LC n-3 PUFA, fatty acid elongation and desaturation, and mitochondrial biogenesis. Taken together, our results support that ALA-biofortified butter is a novel source of n-3 PUFA that potentiates the BAT thermogenic function.
