Available in PDF, EPUB and Kindle. Book excerpt: Reoviruses are ether-resistant icosahedral viruses 60 to 75 mp in diameter which contain ribonucleic acid. They have been recovered from man and lower animals and are ubiquitous in their geographic distribution. At present, the importance of these viruses as a cause of human or animal disease is still largely unknown. As a result of having a number of unusual characteristics, reoviruses have attracted the attention of many workers in the relatively short time since they were first recognized. For example, investigators interested in the molecular aspects of virology have been attracted by the unusual double-helical ribonucleic acid of high molecular weight which reoviruses possess, while those interested in epidemiology have been attracted by the occurrence of apparently identical viruses in both man and an unusually wide variety of lower animals. This com pilation is based on information available to the author as of October 31, 1966. II. History The term "reqvirus" was proposed in 1959 (SABIN, 1959) as a group name for a number of viruses then classified (SABIN, 1956; RAMOS-ALvAREz and SABIN, 1958) as being identical with, or related to, ECHO type lO virus.

Book excerpt: It is now just 20 years since Gomatos and his co-workers at the Rocke feller University showed that the nucleic acid in reovirus particles is double-stranded RNA (dsRNA). This discovery created great excitement, for dsRNA was at that time under intense investigation as the replicative form of viral genomes consisting of single-stranded RNA. An equally interesting and important finding followed soon after: it was found that the reovirus genome consists, not of a single nucleic acid molecule, but of 10 discrete "segments," each with its specific sequence content and each transcribed into its own messenger RNA. It is clear now that these segments are genes. Not surprisingly, the availability of a viral genome 10 unlinked genes has permitted some unique lines of in consisting of vestigation in molecular biology. Mammalian and avian reoviruses proved to be but the first of several viruses recognized as sharing Similarity in size and morphology and ge nomes consisting of 10, II, or 12 separate genes. These viruses are dis tributed throughout living organisms; among the natural hosts of mem bers of this virus family are vertebrates, Insects, and plants. Members of the Reoviridae family differ widely in the virulence that they exhibit toward their hosts . . For example, the first discovered mam malian reovirus literally is, as the name signifies, a "respiratory enteric orphan" virus, that is, a virus unassociated with disease.

The development of methods in environmental virology became a focus of growing interest about two decades ago. Progress has been significant since that time in pure experimental systems, where there are no interferences, consistent high recoveries of viruses from environmental waters has been achievable for some time. In the natural environment, however, in relatively clean waters, substances such as humic and fulvic acids interfere with viral recoveries and average recovery rates probably do not reach 20%. With sewage sludges and shellfish, recoveries are undoubtedly much lower. Yet, even relatively low viral recovery rates have made possible the detection of viral hazards in drinking waters. The hazards that exist are undoubtedly much greater than those demonstrated with the relatively inefficient methods inefficient methods developed thus far. Improving methods, as they are developed in the years to come, will undoubtedly bring the true extent of the hazards into better perspective.
