Book excerpt: Une enquête ethnobotanique effectuée dans les îles de la Grande-comore et d’Anjouan, a permis de recenser 44 plantes utilisées en médecine traditionnelle. L’intérêt de ce travail s’est porté plus particulièrement sur les plantes utilisées dans le traitement du paludisme. Après évaluation sur des cultures in vitro de Plasmodium falciparum, trois plantes ont été sélectionnées pour une étude phytochimique et pharmacologique. Des feuilles de Flueggea virosa ont été isolés huit composés, dont cinq décrits pour la 1e fois dans la plante. Quatre composés phénoliques : la rutine, l’acide gallique, le gallate de méthyle, la bergénine ; et quatre alcaloïdes : deux de type sécurinine, un alcaloïde indolique de type β-carboline, et un alcaloïde indolomonoterpénique. Le gallate de méthyle est le composé le plus actif de Flueggea virosa (CI50 = 2,6 μg/ml). Suite à ce résultat, une synthèse d’acides phénols et d’esters correspondants a été réalisée. Les esters de l’acide gallique, l’ester méthylique de l’acide caféïque et les esters de l’acide protocatéchique présentent des activités intéressantes. L’extrait chlorométhylénique des parties aériennes de Piper capense, nous a permis d’isoler cinq composés, parmi lesquels 2 composés nouveaux : les Kaousines A et B. La décoction des parties aériennes de Flacourtia indica a permis d’isoler 3 composés, l’homaloside D, le poliothrysoside et le pyrocatéchol, décrits pour la 1e fois dans la plante. Les trois composés montrent des activités antiplasmodiales intéressantes (CI50 = 2 et 3 μg/ml). Les extraits de Flacourtia obtenus après biotransformations montrent une modification des profils chromatographiques, mais pas de gain d’activité antiplasmodiale par rapport à la décoction.
