The size andshape of the surface features could be controlled based on the number of nanoparticledepositions. Characterization of the substrates was performed using W-Visspectroscopy and atomic force microscopy. The multilayer nanoparticle substrates wereexamined for their suitability in surface-enhanced Raman scattering (SERS) by obtainingthe SERS spectrum of oxazine-720 on the substrates. A dramatic increase in the SERSsignal is noted with increasing nanoparticle layers and reaches a maximum for 11nanoparticle-layer depositions. The enhancement is attributed to the underlying surfacemorphology: Surface features on the order of 40 nm in size yield the greatest SERSenhancement due to surface plasmon (SP) excitation. The non-linear optical properties of colloidal nanostructures were examined using secondharmonic generation (SHG). A pronounced second harmonic emission was noted for 13nanoparticle layers and was attributed to SP excitation to yield surface-enhanced SHG. The self-assembly of gold nanorods to form 3-dimensional nanostructures was examined. While the self-assembly was not successful, the aggregated gold nanorods exhibited alarge SERS enhancement. This suggests that the incorporation of gold nanorods intoSERS substrates is a viable avenue for future research.

Book excerpt: Water drops that nucleate and grow over an evaporating polymer solution exposed to a current of moist air remain noncoalescent and self-assemble into close packed arrays. The hexagonally close packed, nearly monodisperse drops, eventually evaporate away, leaving a polymer film, with ordered array of pores. Meanwhile, typical breath figures or dew that form when moist air contacts cold surfaces involve coalescence-assisted growth of highly polydisperse, disordered array of water drops. : This dissertation provides the first quantitative attempt aimed at the elucidation of the mechanism of the breath figure templated assembly of the ordered arrays of pores in polymer films. The creation and evolution of a population of close packed drops occur in response to the heat and mass fluxes involved in water droplet condensation and solvent evaporation. The dynamics of drop nucleation, growth, noncoalescence and self-assembly are modeled by accounting for various transport and thermodynamic processes. The theoretical results for the rate and extent of evaporative cooling and growth are compared with experiments. Further, the dissertation describes a rich array of experimental observations about water droplet growth, noncoalescence, assembly and drying that have not been reported in the published literature so far. The theoretical framework developed in this study allows one to rationalize and predict the structure and size of pores formed in different polymer-solvent systems under given air flow conditions.

Currently, only a few gold nanoparticle-based anticancer drug delivery systems have been reported, compared to the polymer-based delivery systems. Additionally, there is still a lack of understanding for the behavior and fate of the gold-drug conjugate in the body that further attention is required. The overall goal of this thesis is to investigate the in vivo behavior of colloidal gold nanoparticle and its therapeutic efficacy in an animal model, especially in a drug delivery application. To achieve this goal, we investigated the feasibility of using colloidal gold nanoparticle as an anticancer agent delivery vehicle for treatment of cancer. Then, long-term clearance, toxicity, and biodistribution of colloidal gold nanoparticle were studied to further aid in understanding of using colloidal gold nanoparticle as a drug delivery platform. In particular, two representative sizes of gold nanoparticles, 5nm and 60nm, were investigated for the size effect on the therapeutic efficacy, toxicity, biodistribution, and clearance in cancer nanotherapy. We believe that nanoparticle size plays a critical role in not only delivering the drug to the target site but also determining the in vivo behavior such as biodistribution and clearance. By choosing an appropriate size scale for the system, we successfully used the small-sized gold nanoparticles for drug delivery applications, which also displayed no apparent toxicity with desirable clearance from the biological system. This work is significant by providing an insight on a potential ideal candidate for drug delivery carrier for cancer nanotherapy.
