Book excerpt: This research presents an investigation of synthetic model complexes with relevance to the active site of Ni(II) acireductone dioxygenase (Ni-ARD) and Ni(II) superoxide dismutase (Ni-SOD). Acireductone dioxygenases (ARDs) are a unique set of enzymes found in the methionine salvage pathway that catalyze the oxidation reaction of acireductone (1, 2-dihydroxy-3-oxo-5-(methylthio)pent-1-ene). These enzymes share a common polypeptide sequence but bind different metal ions, Ni 2+ or Fe2+, at the active site. The Ni-ARD enzyme is responsible for the off pathway shunt in the pathway. Using the tridentate nitrogen donor ligands hydrotris(3,5-dimethyl-1-pyrazolyl)borate (Tp*) and the newly developed tris(1, 2-dimethyl-4-imadozyl)carbinol, (4-TIC Me, Me) several reactions involving the acireductone analog 2-hydroxy-1, 3-diphenylpropan-1, 3-dione and O2 were investigated for similarities to the Ni-ARD active site. Superoxide dismutases (SODs) play a key role in protecting cells against oxidative damage by regulating the cellular concentration of the superoxide radical (O2.- ) which is an unwanted byproduct of cellular metabolism. This process is accomplished by converting the superoxide radicals to hydrogen peroxide and molecular oxygen. Several small-molecule complexes were synthesized and characterized in an effort to model the reduced state of the Ni-SOD using the Tp* ligand. The structures for these complexes have been determined using X-Ray Crystallography.

Book excerpt: The research reported herein involves synthetic metal complexes of relevance to dioxygenase enzymes (Ni(II)-containing acireductone dioxygenase (Ni(II)-ARD) and quercentinase (2,4-flavonol dioxygenase) that promote oxidative carbon-carbon bond cleavage and CO release. The experiments focus on the elucidation of structure-reactivity relationships and evaluation of the conditions under which CO is generated. It had been proposed that hydrogen bond donors in the secondary environment of the active site metal center in Ni(II)-ARD influence the coordination of the acireductone substrate on the nickel center. To evaluate this proposal, we investigated the Ni(II) coordination chemistry of an acireductone-type enolate anion using a supporting chelate ligand having two internal hydrogen bond-donors. The resulting complex exhibited differences in terms of the organic product distribution in a CO release reaction resulting from oxidative C-C bond cleavage of the enolate ligand relative to this reported for the hydrophobic, 6-Ph2TPA-supported (6-Ph2TPA = N, N-bis((6-phenyl-2-pyridyl)methyl)-N-((2-pyridyl)methyl)amine) analogue. In another study, we found that changes in the supporting chelate ligand or metal center influenced the coordination chemistry of the acireductone-type enolate anion. This chemistry highlighted the propensity of the enolate to undergo isomerization without CO release in the presence of water. Rigorously excluding water enabled the isolation of a Ni(II) enolate complex of the 6-PhTPA ligand and examination of its oxidative CO release chemistry, as well as the spectroscopic characterization of the first Co(II) complex of an acireductone-type enolate. To elucidate factors influencing the CO-release reactivity of metal-flavonoid complexes, some of which have relevance to quercentinase enzymes, we synthesized and characterized the first series of structurally-related metal-flavonolate complexes [(6-Ph2TPA)M(3-Hfl)]X (M = Mn(II), Co(II), Ni(II), Cu(II), Zn(II), Cd(II), Hg(II); X = OTfor ClO4 - ). Exposure of these complexes to UV light initiated photooxidative carboncarbon bond cleavage and CO release in a metal-dependent manner, with closed-shell d10 metals giving rise to the highest rate of CO release. These studies suggest that metal flavonolate species may be useful as a new type of photo-induced CO release molecules (CORMs). Such species are of current interest for possible therapeutic applications.

Book excerpt: The chemistry of nickel in biological systems has been intensely investigated since the discovery of the essential role played by this transition metal in the enzyme urease, ca. 1975. Since then, several nickel-dependent enzymes have been discovered and characterized at the molecular level using structural, spectroscopic, and kinetic methods, and insight into reaction mechanisms has been elaborated using synthetic and computational models. The dual role of nickel as both an essential nutrient and as a toxin has prompted efforts to understand the molecular mechanisms of nickel toxicology and to uncover the means by which cells select nickel from among a pool of different and more readily available metal ions and thus regulate the intracellular chemistry of nickel. This latter effort highlights the importance of proteins involved in the extra- and intra-cellular sensing of nickel, the roles of nickel-selective proteins for import and export, and nickel-responsive transcription factors, all of which are important for regulating nickel homeostasis. In this Special Issue, the contributing authors have covered recent advances in many of these aspects of nickel biochemistry, including toxicology, bacterial pathogenesis, carcinogenesis, computational and synthetic models, nickel trafficking proteins, and enzymology.

Their Ni(II) complexes, and some Zn(II), Cu(II), and Fe(III) complexes, have been structurally and spectroscopically characterized. Deprotonation of the oxime groups gives Ni(II)--Polyoximate complexes that form bis(N-O) oximate bridged dimers with significantly low oxidation potentials for Ni(II). These Ni(II)-polyoximate complexes react with dioxygen, consuming multiple equivalents. This is unusual chemistry for Ni(II), especially in the absence of ligand oxidation. The Ni(II) / O 2 reaction system promotes oxidation of triphenyl phosphine, with oxygen atom transfer, benzyl alcohol, and 3,5-di-tert-butylcatechol. Reaction of the Ni(II)-polyoximate complexes with o-quinones causes the formation of high-valent nickel complexes of the reduced dioxolenes, semiquinones and catecholates.

Book excerpt: Metalloenzymes are able to perform a wide variety of difficult chemical transformations in a highly selective and efficient manner. The ability of metal complexes to perform a particular chemical reaction is dependant on the properties of the primary and secondary coordination environments. Development of transition metal complexes that catalyze the formation of organic compounds in a selective and highly efficient manner is of interest to synthetic chemists. Advancement in this area of research requires studies aimed at developing ligands that will give desirable properties upon binding to a metal ion. Using interactions present in biological systems, the ligands in this study have been developed to investigate the effects of the primary coordination sphere as well as hydrogen bonding interactions on complex formation as well as reactivity. Chapter 2 discusses the synthesis of terminal monomeric NiII-OH complexes with H41R. The hydroxo ligand is stabilized by two intramolecular hydrogen bonding interactions and the source of the hydroxo ligand was confirmed to be water. Attempts to deprotonate the NiII-OH complex to form a NiII-oxo complex were unsuccessful. Treating the NiII-OH complexes with KOtBu afforded the alkoxide substituted product. NiII-siloxide complexes were isolated upon reaction of NiII-OH with K[N(TMS)2]. Chapter 3 features the synthesis of a series of Ni(II) complexes with H2pmb, which contains two carboxyamido units appended from pyridine rings. Four Ni(II) complexes were synthesized and structurally characterized. It was determined that the carboxyamido groups either provided intramolecular H-bonds or the carbonyl oxygen can bind directly to the metal center. Chapter 4 explores the electrochemistry of the Ni(II) complexes characterized in Chapter 3. Based upon the observed reduction potentials, the reduction chemistry of [NiIIH2pmb(Cl)2] was explored. Further reactivity of the reduced species was also explored with dioxygen. Preliminary reactivity studies will also be discussed.
