Book excerpt: Cellulases are important in industrial enzyme which is a main part in industrial process such as in bio-bleaching and bioethanol production. Nowadays, our country encourages the production of bioethanol from the biomass material such as oil palm empty fruit bunch fiber in order to enhance the resources of ethanol production and reduce the cost.In this project, oil palm empty fruit bunch fiber was used as a substrate for the production of cellulase enzyme by Aspergillus terreus SUK-1. In order to enhance the cellulase production, pretreatment on substrate are performed with different type of methods. Comparison results of cellulase enzyme production are determined between the substrate which is pretreated with acid and alkaline pretreatment. The effect of steam explosion on the substrate also was identified in shaking flask fermentation. From the experimental work, it was found that the highest xylanase and carboxymethylcellulase activity was in fermentation of acid treated substrate than alkali treated substrate and untreated substrate It also found that by autoclaved the substrate with chemicals produced the highest cellulase enzyme with value 0.167836U/ml for xylanase and 0.3534U/ml for carboxymethylcellulase.

Book excerpt: The study was conducted for the production of cellulase enzyme through liquid state bioconversion of palm oil mill effluent (POME) by filamentous fungi and characterization of the produced enzyme. Several experiments such as selection of filamentous fungi, optimization of media and process parameters, development of the separation/purification conditions for ultra-filtration, characterization and evaluation of purified cellulase enzyme through hydrolysis of oil palm empty fruit bunches (EFB) were carried out to achieve the entire objectives of the study. The results on isolation indicated that among the strains isolated, the strains IBPC108R, IBEC302B and IBEC305B gave the highest FPAse activity (0.3 U/ml) after 2 days of fermentation and IBEC302B showed the highest CMCase activity (0.42 U/ml) after 3 days of fermentation. Among the laboratory stocks, Aspergillus IBO-102MNB, gave the highest FPAse activity (0.28 U/ml) and Aspergillus IBO-103MNB gave the highest CMCase activity (0.62 U/ml) after 2 days of fermentation. Additionally, cellulase contributes about 20% of the total enzyme market globally because of its massive demand in various industries such as in biofuel production, pulp, paper, textile, food, and beverages, as well as in detergent industries. Among these, the demand of cellulase may become frequently selected in the commercial production of biofuels in the future and thus will further increase demand of cellulase in the biofuel industry. Because biofuel production is still not realized in a cost-effective, practical implementation due to its high cost (the higher cost of biofuels is due to higher production costs of enzymes), there is a need to introduce these types of approaches, which will help to lower the cost of enzyme production for developing overall economic biofuel production.

Book excerpt: Hydrolysis of EFB was studied using crude enzyme from Aspergillus terreus SUL-1 as well as commercial enzyme from Trichoderma reesai and Aspergillus niger. Hydrolysis was performed at 50 C but sugar yield dropped significantly when commercial enzymes of T. reesei was used and the hydrolysis time extended beyonf 24 hrs. Low sugar yield was obtained from the hydrolysis of EFB. Glucose concentrations of 0.25 % and 0.5 % caused significant inhibition on the crude enzyme. Glucose was found to be a competitive inhibitor, with a Ki value of 3.33 mM when x-cellulose was used as a substrate.Contrary to the results with crude enzyme, glucose inhibition was found to have little effect on the commercial enzymes. In the present study, optimization for cellulase production by Aspergillus niger was investigated by using paddy straw as substrate.Cellulase enzyme was produced by the fungus extracellularly. Optimization of cellulase production was done by using various physical (Temperature, pH, Salinity and Incubation time) and chemical parameters (Carbon sources and nitrogen sources) which could influence the enzyme activity. Cellulase production was maximum at the temperature 20°C and minimum at 40ºC. The optimal pH for the cellulase production was observed maximum in 6.0 and minimum in 7.0. Cellulase production was maximum at 48 hours and minimum at 24 hours. Cellulase production was maximum with when fructose was used as a carbon source and minimum with sucrose. Cellulase production was maximum when Malt extract was used as a nitrogen source and minimum with yeast extract. The cellulase enzyme extract was partially purified and its protein fraction was stained with its molecular weight determination by using SDS-PAGE. It revealed two protein bands with the molecular weights of about 83 kD and 50kD.

In the book, the author reported on production optimization and characterization of cellulolytic enzymes by thermophilic bacteria. Various adaptation strategies adapted by thermophilic bacteria are also discussed along with. The thermophilic bacterial strain produced cellulase in the CMC broth, pH 7 containing 0.5% peptone, 0.5% malt extract, 0.2% ammonium sulphate, 0.2% ammonium nitrate and 0.2% NaCl at 50 ̊C. Interestingly, the bacteria could not grow at 37 ̊C, confirming its thermophilic nature. Further, the cellulase was characterized after getting partially purified by ammonium sulphate precipitation method. The partially purified cellulase may be employed to hydrolyze agricultural wastes to produce bioethanol, i.e. biofuels. Thus, the present research may help solve issues of crisis of renewable energy as well as environmental pollution.
