The Thirteenth Washington International Spring Symposium was organized with the intention of bringing together scientists working on different aspects of the cell cycle. The goal of this gathering was to help formulate a more comprehensive and integrated picture of events driving and being driven by the cell cycle, as well as to evaluate the possibilities for clinical application of this knowledge. This symposium, held in Washington, D.C. from May 10-14, 1993, was attended by more than 400 scientists from 20 countries, including many of the scientific leaders in this field. This volume contains most of the papers presented at the seven plenary sessions in addition to selected contributions from a total of nine special oral and poster sessions.

Book excerpt: For more than three decades the methods for assessing cell pro liferation have been largely the preserve of experimental biologists, and in their hands such techniques have contributed greatly to our understanding of the dynamic organisation of normal and patho logical tissues. In recent years, with the advent of novel method ologies, there has been increased interest among both pathologists and clinicians, particularly oncologists and others interested in neoplasia, in assessing cell proliferation. This interest has been stimulated by the possibility that indices of cell proliferation may have direct clinical relevance, for example in being useful predictors of outcome in patients with certain forms of malignancy. In addi tion, interest in assessing cell proliferation has been fuelled by the tremendous advances in our understanding of the mechanisms of cell proliferation and their deregulation in pathological processes. Consequently, the time is ripe for a monograph critically reviewing the available methods for assessing cell proliferation, their potential and their problems. We have been particularly concerned to present a balanced view of the advantages and disadvantages of different methods currently available for assessing cell proliferation. The assessment of cell proliferation often requires some familiarity with mathematical methods, but in this book we have attempted to keep detailed mathematical analyses to a minimum. Book excerpt: More than 10 years ago, the discovery of cyclin-dependent ki nases (Cdks) ushered in a new era in the understanding of cell proliferation and its control. Not only were both of the known cell cycle transitions, from G 1 to S phase and G2 to M phase, found to be dependent on these protein kinases, but the reg ulatory assumption intrinsic to cyclin-dependent kinases, a stable inactive catalytic subunit (the Cdk) and an unstable requisite positive regulatory activating subunit (the cyclin), led to a simple model for cell cycle control. Modulation of cyclin accumulation, and thereby Cdk activation, was proposed to be the overarching principle governing the passage through cell cycle phases. An other reality to emerge from the discovery of Cdks was the ex ceptional degree of evolutionary conservation maintained in the machinery and organization of proliferation control. Not only were Cdks shown to be structurally conserved between yeast and man, but mammalian Cdks could substitute functionally for the endogenous enzymes in a yeast cell. The problem of cell cycle control was thought to have been virtually solved. The ensuing years have provided a much more complex view of cell cycle control and the role and regulation of Cdks. The uncritical enthusiasm with which many of the ideas were em braced has required tempering. For example, although Cdks appear to be highly conserved phylogenetically, cyclins are much less so.
