The book also introduces the discussion on mitochondrial calcium transport protein, polypeptide and amine hormone regulation of calcium fluxes, mechanism of secretion by cultured pituitary cells, and regulation of the tubular transport of phosphate. A chapter that assesses the effects of vitamin D and insulin on phosphate transport in the differentiating chick small intestine concludes the book. A short account of the history of the effort led to the Phosphate Workshops is appro priate and can be of interest to the reader. The idea for Phosphate Workshops was born in the early days of November, 1974. One of us (S. G. M. ) suggested the thought to a group of scientists gathered for a luncheon in one of the attrac tive small restaurants in Weisbaden, Germany. The purpose of the workshop was to bring together interested scientists to discuss the newer developments and the recent advances in the field of phosphate metabolism and the other related minerals. An Organizing Committee made of Shaul G. Massry (USA), Louis V. Avioli (USA), Philippe Bordier (France), Herbert Fleisch (Switzerland), and Eduardo Slatopolsky (USA) was formed. The First Workshop was held in Paris during June 5-6, 1975 and was hosted by Dr. Philippe Bordier. Its proceeding was already published. The Second Workshop took place in Heidelberg during June 28-30, 1976 and was hosted by Dr. Eberhard Ritz. Both of these workshops were extremely successful scientific endeavors, and the need for them was demonstrated by the great interest they generated among the scientific community. The Or ganizing Committee, therefore, decided to continue with the tradi tion to hold additional Workshops annually or every other year.

The American Society for Pharmacology and Experimental Therapeutics had the primary responsibility for organizing the scientific program with the assistance of other member societies, including the American Physiology Society, American Association of Pathologists, and American Institute of Nutrition. The purpose of the Calcium Theme was to review progress in the diverse areas of investigation bearing on the ubiquitous role of calcium in biological systems. In addition to contributions from those participating in the Theme, this volume also includes a number of invited papers that were added to fill certain voids in topics covered. The authors were selected because they are investigators active in the mainstream of their particular research area, possessing the acumen to analyze cogently not only their own recent findings but also to relate these findings to their respective area. New information as well as reviews of current concepts generally highlight the individual contributions. Undoubtedly, some readers may argue with the emphasis made and/or the conclusions reached on individual topics. In such cases, other volumes will hopefully provide a forum for alternative points of view. Due to the broad scope of subjects covered and the large number of contributions, the papers have been arranged in three sections.

Book excerpt: The endothelium, a monolayer of endothelial cells, constitutes the inner cellular lining of the blood vessels (arteries, veins and capillaries) and the lymphatic system, and therefore is in direct contact with the blood/lymph and the circulating cells. The endothelium is a major player in the control of blood fluidity, platelet aggregation and vascular tone, a major actor in the regulation of immunology, inflammation and angiogenesis, and an important metabolizing and an endocrine organ. Endothelial cells controls vascular tone, and thereby blood flow, by synthesizing and releasing relaxing and contracting factors such as nitric oxide, metabolites of arachidonic acid via the cyclooxygenases, lipoxygenases and cytochrome P450 pathways, various peptides (endothelin, urotensin, CNP, adrenomedullin, etc.), adenosine, purines, reactive oxygen species and so on. Additionally, endothelial ectoenzymes are required steps in the generation of vasoactive hormones such as angiotensin II. An endothelial dysfunction linked to an imbalance in the synthesis and/or the release of these various endothelial factors may explain the initiation of cardiovascular pathologies (from hypertension to atherosclerosis) or their development and perpetuation. Both structurally and functionally, there are many peculiarities that differentiate the brain from all other organs. The brain is our connection to the world around us and by governing nervous system and higher function, any disturbance induces severe neurological and psychiatric disorders that can have a devastating effect on quality of life. Our understanding of the physiology and biochemistry of the brain has improved dramatically in the last two decades. In particular, the critical role of cations, including magnesium, has become evident, even if incompletely understood at a mechanistic level. The exact role and regulation of magnesium, in particular, remains elusive, largely because intracellular levels are so difficult to routinely quantify. Nonetheless, the importance of magnesium to normal central nervous system activity is self-evident given the complicated homeostatic mechanisms that maintain the concentration of this cation within strict limits essential for normal physiology and metabolism. There is also considerable accumulating evidence to suggest alterations to some brain functions in both normal and pathological conditions may be linked to alterations in local magnesium concentration. This book, containing chapters written by some of the foremost experts in the field of magnesium research, brings together the latest in experimental and clinical magnesium research as it relates to the central nervous system. It offers a complete and updated view of magnesiums involvement in central nervous system function and in so doing, brings together two main pillars of contemporary neuroscience research, namely providing an explanation for the molecular mechanisms involved in brain function, and emphasizing the connections between the molecular changes and behavior. Several metabolic pathways are compartmentalised within the ER lumen, for example hydrolysis of glucose 6-phosphate, glucuronidation of endo- xenobiotics, posttranslational modification of proteins including redox reactions required for oxidative folding, oxidoreduction of steroid hormones, synthesis of ascorbate. Therefore, enzyme activities of these pathways depend on the special luminal microenvironment, on access to substrates and on release of products. However, in spite of great efforts, the molecular mechanism for the generation and maintenance of this special microenvironment still remains to be elucidated. Beside the well-known functions of the endoplasmic reticulum, such as calcium signaling and the synthesis of secretory proteins, recent findings underlined the importance of the intraluminal redox biochemistry and the role of membrane transporters. The field is currently undergoing extensive reappraisal, new transporters have been identified either molecular or functional level.The local synthesis and the membrane transport of redox active compounds (pro- and antioxidants) seem to be important not only in the disulfide bond formation, but also in the generation of intracellular proliferative/apoptotic signals. The different points of views in this publication help highlight the potential importance of several recently described phenomena, whose significance needs elucidation.
