Book excerpt: Phage biology is one of the most significant and fundamental aspects of biological research and is often used as a platform for model studies relating to more complex biological entities. For this reason, phage biology has enjoyed focused attention and significant advances have been made in the areas of phage genomics, transcriptomics and the development and characterisation of phage-resistance mechanisms. In recent years, considerable research has been performed to increase our understanding of the interactions of these phages with their hosts using genomic, biochemical and structural approaches. Such multidisciplinary approaches are core to developing a full understanding of the processes that govern phage infection, information that may be harnessed to develop anti-phage strategies that may be applied in food fermentations or applied in a positive sense in phage therapy applications. The co-evolutionary processes of these phages and their hosts have also been a considerable focus of research in recent years. Such data has promoted a deeper understanding of the means by which these phages attach to and infect their hosts and permitted the development of effective anti-phage strategies. Furthermore, the presence and activity of host-encoded phage-resistance systems that operate at various stages of the phage cycle and the potential for the application of such systems consolidates the value of research in this area. Conversely, phages and their components have been applied as therapeutic agents against a number of pathogens including, among others, Clostridium difficile, Lactococcus garviae, Mycobacterium spp., Listeria spp. and the possibilities and limitations of these systems will be explored in this topic. Additionally, phage therapeutic approaches have been applied to the prevention of development of food spoilage organisms in the brewing and beverage sectors and exhonorate the positive applications of phages in the industrial setting. This research topic is aimed to address the most current issues as well as the most recent advances in the research of phages infecting Gram-positive bacteria covering areas such as phages in food fermentations, their impact in industry, phage ecology, genomics, evolution, structural analysis, phage-host interactions and the application of phages and components thereof as therapeutic agents against human and animal pathogens.

The diagnosis, antibiotherapy, and prevention of the streptococcal diseases have improved considerably. However, the reemergence of severe streptococcal and enterococcal diseases constitutes a growing public health con cern, which remains open to scientific and medical debate. The XIII'h Lancefield International Symposium on Streptococci and Streptococcal Diseases, held at Institut Pasteur, Paris, France, September 16---2el, 1996, attracted 505 par ticipants from 43 countries. Twenty-two percent of the participants were students, a clear sign of the intense interest in this field. Of the 390 presentations made at the symposium, 260 were submitted as manuscripts for the Proceedings; we have included 249 of these in this volume. This symposium provided a forum for the presentation of the most recent findings and approaches to understanding several important fields, such as new aspects of infec tion, bacteria~host interactions, epidemiology, and molecular genetics of streptococci and enterococci. Over the last three years, the study of these subjects has expanded as increas ingly sophisticated methods of molecular analysis have been applied to investigate the bi ology of pathogenic streptococci and enterococci. Virulence, vaccine strategies, genetics, antibiotic resistance, epidemiology, and immunology are now being examined through the lens of molecular biology. The application of recently developed techniques to this field will continue to yield insight into the mechanism by which these organisms cause disease.

Book excerpt: Lactic acid bacteria (LAB) are gram-positive, acid tolerant rods or cocci which produce lactic acid as the major end product of fermentation (Axelsson, 1993). This group comprises the genera Lactococcus, Lactobacillus, Leuconostoc, Streptococcus, Pediococcus and Carnobacterium, many of which play important roles in the food fermentation industry (McKay and Baldwin, 1990). LAB are used as starter cultures for the production of a wide range of fermented dairy foods (i.e. cheeses, yogurts, and fermented milks) as well as a diversity of other products such as fermented meats, sour dough bread and silage (Mercenier et al., 1994). In addition to their capacity to rapidly acidify the fermentable food product, thereby inhibiting growth of pathogens and spoilage microorganisims, they also provide a range of flavour and texture characteristic to the food. Bacteriophage (phage) are obligete intracellular, bacterial parasites and are found ubiquitously in nature. Phages infecting LAB are of considerable economic importance because they repreesent one of the main causes of fermentation failure. The dairy industry has implemented a number of technological strategies to alleviate the problem of phage infection including air filtration, separation of bulk starter tanks from the production area, direct vat inoculation, the use of closed vats, rotation of starter strains and the application of phage-resistant starter cultures (Neve, 1996). However, these measures have not eliminated phage as a serious problem in industrial practice. As an alternative strategy a significant amount of research has been aimed at understanding phage-host interactions as well as phage biology in the expectation that a fundamental understanding of the LAB phages may assist in providing an efficient and long-term solution to phage infection. Due to these investigations, a significant body of knowledge is now available on the primary genome structure and organisation of many phages infecting LAB thus giving an insight into their evolutionary history while at the same time providing the information for the development of "intelligent" phage resistance strategies. This review will summarise the accumulated information available on LAB phage biology and genomics with occasional emphasis on phages infecting S. thermophilus.

To communicate and stimulate the research on lactic acid bacteria and their applications, a series of tri-annual symposia on lactic acid bacteria was started in 1983 under the auspices of the Netherlands Society for Microbiology (NVVM), which was later also supported by the Federation of European Microbiological Societies (FEMS). The aim of these state-of-the-art symposia is to offer a unique platform for universities, institutes, and industry in this area of biotechnology, to present recent work, to obtain information on new developments, and to exchange views with colleagues from all over the world on scientific progress and applications. The growing number of participants at these symposia has been a clear demonstration of the interest of the international industrial and scientific community in this area of research. The 7th Symposium is based on a number of plenary lectures that review the scientific progress of the last years in the different areas of research on lactic acid bacteria, and which are documented in this special issue of Antonie van Leeuwenhoek.

A striking feature of the virus is that it consists essentially of two components: a protein envelope and the nucleic acid con tained within it. In contrast to the elementary structural unit of the living or ganism, the cell, which contains two types of nucleic acid (DNA and RNA), the virus particle contains only one type of nucleic acid - either DNA or RNA. It is perhaps this which is responsible for the imperfection of the virus as a living organism.
