Book excerpt: An in vitro rumen experimental study was conducted on the effects of dietary supplementation of Brevibacillus laterosporus (LATER) and Bacillus coagulans (COAG) or both on diet fermentation. In vitro ruminal fermentation and fibre digestibility with respect to time were determined with nylon bag technique using two cannulated cows. Two separate in vitro ruminal batch cultures were performed on different days using the following treatments: Control (Basal diet: CON), Basal diet plus Monensin(positive control: MON), Basal diet plus Brevibacillus/aterosporus (1.0 x 109 cfu: LATER), Basal diet plus Bacillus coagulans (1.0 x 109 cfu: COAG) and Basal diet plus LATER+COAG. Rumen fluid was collected from a cannulated lactating Holstein cow and mixed with Menke's buffer (4:1) under anaerobic conditions. The diet (0.5 g) was added to 100 ml buffered rumen fluid and incubated in triplicate in 250 ml glass serum bottle in a shaking water bath at 39 °c for 24, 48 and 72 h. The pH was measured after each incubation period, and the in vitro dry matter digestibility (IVDMD), neutral detergent fibre (NDF) and acid detergent fibre (ADF) digestibility were determined. There was no difference in IVDMD after 24 h of treatment supplementation, but at 48 and 72 h, the lVDMD was higher with COAG, LATER and their combination compared to the control and Monensin treatments. The additives did not have effect on pH. Ammonia nitrogen production was reduced at 48 h compared to CON. Total VFA’s was higher for COAG+LATER than the rest of treatments and higher for MON and LATER than CON and COAG. Propionate was higher for LATER and COAG+LATER compared to CON. All the additives and MON decreased butyrate. These results show that COAG and LATER can be used as an alternative to antibiotics. The improved IVDMD was more than Monensin, which has been widely used as antibiotic. The results of volatile fatty acids and reduction in ammonia production suggest that the additives have potential to increase growth and cellulotic activity of rumen bacterial and improve nutrient digestibility.
