Book Autoradiography at the Cellular Level

Download or read book Autoradiography at the Cellular Level written by Brigitte Schultze and published by Academic Press. This book was released on 2013-10-22 with total page 313 pages. Available in PDF, EPUB and Kindle. Book excerpt: Physical Techniques in Biological Research, Second Edition, Volume III, Part B: Autoradiography at the Cellular Level focuses on high resolution autoradiography with tritium. This book reviews the different autoradiographic techniques and difficulties in theoretical principles needed for the interpretation of autoradiograms. The topics include the characteristics of the different types of radiation; autoradiographic resolution; techniques for applying photographic emulsions; and autoradiography of water-soluble substances. The application of different isotopes; general aspects of the use of thymidine; physiological RNA synthesis; and RNA synthesis under pathological conditions are also elaborated. This text likewise covers the protein synthesis under normal conditions; electron microscopic autoradiography; application of electron microscopic autoradiography; and autoradiographic methods in cytochemistry. This publication is a good reference for students, researchers, and clinicians conducting work on the use of tritiated compounds in high resolution autoradiography.

Book The Use Of Radioautography in Investigating Protein Synthesis

Download or read book The Use Of Radioautography in Investigating Protein Synthesis written by C Leblond and published by Elsevier. This book was released on 2012-12-02 with total page 361 pages. Available in PDF, EPUB and Kindle. Book excerpt: The Use of Radioautography in Investigating Protein Synthesis represents the 1965 Symposium on The Use of Radioautography in Investigating Protein Synthesis, supported by the International Society for Cell Biology. This book is composed of four parts encompassing 18 chapters. The opening part deals first with the basic aspects and mode of operation of the radioautography technique. This part also examines the advantages and potentials of this technique compared to other cytological techniques. The next parts are devoted to the experimental methods of protein synthesis in nucleus and in cells of various types, as revealed by the electron microscope radioautography. The concluding part discusses the secretion and synthesis of protein enzymes by the acinar cells in rat pancreas and fibroblasts in wound healing. This part also considers the application of radioautography to determining the sites of synthesis of proteins. This book is of great value to cell biologists, biochemists, cytologists, histologists, and pathologists.

Book Microautoradiography and Electron Probe Analysis

Download or read book Microautoradiography and Electron Probe Analysis written by U. Lüttge and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 245 pages. Available in PDF, EPUB and Kindle. Book excerpt: In biological literature, several definitions of quantitative autoradio graphy are given. The term is defined as either the determination and com parison of the density of silver grains above various structures or under varying conditions, or the determination of absolute quantities of radio activity. In both these cases, photometric measurement serves for more rapid and more exact evaluation of grain densities than would be possible by visual counting of the grains. The equipment generally used for the photometric measurement of silver grains consists of a microscope, a photocell, an electronic amplifier system and a display unit. Grains can be made accessible to photometric evaluation by various kinds of microscopic illumination: 1. Substage bright-field illumination. 2. Substage dark-field illumination. 3. Incident dark-field illumination. 4. Vertical bright-field illumination. With all these types of illumination, the relationship between the luminous flux I absorbed by the film, scattered into the objective and reflected or diffracted, and the flux 10 which is not affected by the film is used as a measure of grain density. Since these are differential measurements, the light beam I transmitted by the film is in itself a measure of grain density if the luminous flux 10 incident on the grains is kept constant. This approach has been used in a large number of measuring setups.

Book Autoradiography and Immunocytochemistry

Download or read book Autoradiography and Immunocytochemistry written by M.A. Williams and published by Elsevier. This book was released on 2013-10-22 with total page 238 pages. Available in PDF, EPUB and Kindle. Book excerpt: Autoradiography and Immunocytochemistry sets out in detail the preparation of autoradiographs and methods of preparing and use of immunological staining reagents. This book begins with an introduction to the demand for autoradiography and immunocytochemistry and their most advantageous use, followed by a discussion on the immunocytochemistry at the electron microscopical (EM) level in relation to the preparation of labeled antibodies. Other topics include the general remarks on “staining with antibodies; applying Fer-Ab conjugates to ultrathin sections; and controls in immunoferritin staining. The multistep antibody staining methods with unmodified proteins; control observations in staining experiments using lectins; and pattern analysis in EM immunocytochemistry are also covered. This text concludes with a description of the preparation of electron microscope autoradiographs, including the radioisotopes in EM autoradiography, preparation of radioactively-labeled tissues and cells, and methods of applying nuclear emulsions. This publication serves as a detailed laboratory guide for researchers and workers to successfully conduct electron microscope techniques.
