Book excerpt: Rhoptry proteins participate in the invasion of red blood cells by merozoites during the malaria parasite's asexual-stage cycle. Interference with the rhoptry protein function has been shown to prevent invasion, and three rhoptry proteins have been suggested as potential components of a vaccine against malaria. Rhoptry associated membrane antigen (RAMA) is a 170-kDa protein of Plasmodium falciparum which is processed to a 60-kDa mature form in the rhoptries. p60/RAMA is discharged from rhoptries of free merozoites and binds to the red-cell membrane before being internalized to form part of the parasitophorous vacuole of the newly developing ring. We examined the range of anti-RAMA responses in individuals living in an area of endemicity for malaria and determined its association with clinical immunity. RAMA is immunogenic during infections, and at least three epitopes within RAMA are recognized by hyperimmune sera in immunoblots. Sera from individuals living in a region of Vietnam where malaria is endemic possessed strong antibody responses toward two C-terminal regions of RAMA. Cytophilic antibody isotypes (immunoglobulin G1 [IgG1] and IgG3) predominated in humoral responses to both C-terminal epitopes. Acute episodes of P. falciparum infection result in significant boosting of levels of antibody to an epitope at the extreme C terminus of RAMA that harbors the red-cell-binding domain. Immunity to P. falciparum infection was linked to elevated levels of IgG3 responses to this functional domain of RAMA, suggesting that the region may contain a protective epitope useful for inclusion in a multiepitope vaccine against malaria.

This group of human-pathogenic Plasmodium species are usually referred to as malaria parasites. This book presents leading-edge new research in this field.

The genome sequences were a result of the Plasmodium falciparum Genome Project. This book covers in detail the biology of malaria parasites and the mosquitoes that transmit the disease, how the Genome Project came into being, the people who created it, and the cadre of scientists who are attempting to see the promise of the Project realized. The promise was: a more complete understanding of the genes of the parasite (and its vector) would provide a rational basis for the development of antimalarial drugs and vaccines, allow a better understanding of the regulation of the complex life cycle in the red blood and liver cells of the human, identify the genes the parasite uses to thwart the host immune response and the ways in which the parasite evades cure by drug treatments, as well as leading to more effective measures of control transmission. The hope was that cracking the genetic code of Plasmodium and Anopheles would reveal the biochemical Achilles heel of the parasite and its vector, leading to the development of novel drugs and better methods of control, and by finding the targets of protective immunity could result in the manufacture of effective vaccines. Book excerpt: Malaria, caused by infection with protozoan parasites belonging to the genus Plasmodium, is a highly prevalent and lethal infectious disease, responsible for 435,000 deaths in 2017. Optimism that malaria was gradually being controlled and eliminated has been tempered by recent evidence that malaria control measures are beginning to stall and that Plasmodium parasites are developing resistance to front-line anti-malarial drugs. An important milestone has been the recent development of a malaria vaccine (Mosquirix) for use in humans, the very first against a parasitic infection. Unfortunately, this vaccine has modest and short-lived efficacy, with vaccinated individuals possibly being at increased risk of severe malarial disease when protection wanes. Thus, to define new ways to combat malaria, there remains an urgent requirement to identify the immune mechanisms that promote resistance to malarial disease and to understand why these so often fail. The review and primary research articles in this Research Topic illustrate the breadth of research performed worldwide aimed to understand the biology of the Plasmodium parasite, the roles of the various cell types that act within the immune response against the parasite, and the parasitological and immunological basis of severe malarial disease. The articles in section 1 exemplify the different vaccination strategies being developed and tested by the research community in the fight against malaria. The articles in section 2 review important overarching aspects of malaria immunology and the use of models to study human malaria. The articles in section 3 describe the ways through which the Plasmodium parasite is initially recognised by the immune system during infection, how the parasite can directly impact this critical event to restrict anti-Plasmodial immunity, and resolve the roles of key innate cell populations, such as dendritic cells, in coordinating malarial immunity. The articles in sections 4-6 outline the roles T and B cell populations play during malaria, highlighting the activation, diversification and regulation of the crucial cell types during malaria, and discuss some of the reasons adaptive immunity to malaria is often considered so poor compared with other diseases. The articles in section 7 provide up to date information on the pathogenesis of cerebral malaria, bridging our understanding of the syndrome in humans with information learned from animal models. Overall, the articles in this research, many of which are published by leaders in the malaria field, emphasize the imagination and technical advances being employed by researchers against malaria. Based on the work of a number of research groups, we know that complement plays an important role in these processes. The development of a vaccine against the human malaria parasite Plasrnodium falciparum requires first, the identification of the parasitic antigens involved in protection, and second, the use of appropriate animal models to study protective immune mechanisms and to optimize immunization protocols intended to induce them. A subset of P. falciparum preerythrocytic antigens (LSA l, LSA3, SALSA and STARP) have been identified and characterized. Among them, LSA3 displayed promising antigenic, immunogenic and protective efficacy in différent animal models. The current thesis describes the immune responses induced by these antigens, particularly the immunogenicity and protective potential of LSA3 in mice and Aotus monkeys. Aotus lemurinus griseirnembra is a small New World monkey. We confirmed that the species A./.griseimembra is susceptible to infection by P. falciparum sporozoites and we were able to maintain the parasite by cyclical passages through Aotns and mosquitoes. When Aotus lemurinus were immunized with either, a mixture of lipopeptides from 4 P. falciparum preerythrocytic antigens or LSA3 recombinant proteins, the monkeys were able to induce high cellular and humoral immune responses, indicating that it is a suitable animal model for evaluating immunogenicity of pre-erythrocytic antigens. Finally, for the first time we report evidence of protection against challenge with P. falciparum sporozoites in Aotus monkeys.Taking advantage of the recent advances in peptide synthesis, we used 17 Long Synthetic Peptides (LSP) covering all the LSA3 protein to confirm the antigenicity of LSA3 antigen in individuals highly exposed to natural P. falciparum infections. In addition, these LSP were found to be strongly immunogenic in mice, eliciting T-helper, CTL responses and specific antibody production. On the other hands, LSA3-DNA-based immunization in mice induced high immune responses against different regions of the molecule and preferentially generated responses Th 1-type. Moreover, this immunization protocol was able to induce protection against the challenge with P. yoelii sporozoites. These findings confirm previous results about the vaccine potential of LSA3 and allow to validate Aolus monkeys as model for preerythrocytic malaria vaccines research. They open the way to a great number of future studies on P. falciparum pre-erythrocytic stages.
