Book excerpt: The annual research conference for 1996 of the American Institute for Cancer Re search was again held at the Loews L'Enfant Plaza Hotel in Washington, DC, August 29 and 30. The topic for this, the seventh in the series, was "Dietary Fat and Cancer: Genetic and Molecular Mechanisms. " Two separate presentations were given as the conference overview. "Fat and Cancer: The Epidemiologic Evidence in Perspective" noted that die tary fat can be saturated, largely from animal or dairy sources, or mono- or polyunsatu rated, mostly from plant sources. Unlike animal fats, fish contain relatively high levels of protective omega-3 fatty acids. Although the hypothesis that dietary fat is associated with cancer is plausible, the mechanisms involved are reasonable, and many animal studies support the hypothesis, there are many obstacles in any direct extrapolation to humans, in cluding imprecise measures of dietary fat intake, variability in individual diets, and spe cies variations. Despite these limitations, there is a weak positive correlation between colon cancer and dietary fat intake, but with substantial differences for various ethnic groups. In the case of breast cancer, there is substantial variation among countries and eth nic groups, but the overall evidence indicated an association with fat in the diet. Epidemiologic studies of dietary fat and prostate cancer are more consistent and most show a positive relationship. However, it was not clear which types of dietary fat were im plicated in the effect.

Book excerpt: First attempts to isolate plant genes were for those genes that are abun dantly expressed in a particular plant organ at a specific stage of devel opment. However, many important gene products are produced in a very minute quantity and in specialized cell types. Such genes can now be isolated using a variety of approaches, some of which are described in this volume. The rapid progress during the last decade in regeneration of a number of crop plants and the availability of molecular tools to introduce foreign genes in plants is allowing the engineering of specific traits of agri cultural importance. These genes must, however, be regulated in a spatial and temporal manner in order to have desired effects on plant devel opment and productivity. The habitat of plants necessitate adaptive responses with respect to the environmental changes. Starting from germination of the seed, the plant begins to sense environmental cues such as moisture, light, temperature and the presence of pathogens, and begins to respond to them. Little is known about various signal transduction pathways that lead to biochemical and morphogenetic responses, in particular, transition from vegetative to reproductive phase. With the availability of tools to generate specific mutations via transposon tagging, identification and isolation of genes affecting these processes may be facilitated. Transfer of these genes into heterologous environments will allow understanding of the complex processes that control plant development.

With the advent and impact of molecular biology, the small weed Arabidopsis is now the object of rapidly growing scientific attention. Since it is reasonable to assume that the general molecular mechanisms that are responsible for the physiological, cellular and biochemical properties of plants will be essentially conserved in all plants, it follows that these mechanisms should also operate in Arabidopsis and hence that its genome should contain most of the genes that we need to know about if we want to understand the genetic determination of the life processes in plants.Arabidopsis has a small genome and well documented genetic studies are available. It is easy to grow in large numbers and mutants defining important genetically controlled mechanisms are either available, or can readily be obtained. Various methods to introduce and express isolated homologous or heterologous genes are available. It is therefore realistic and desirable to aim at exploring the genome of this plant in very great detail. As will be illustrated in this book all the elements for such a grand strategy are in place.More and more scientists are therefore willing to accept the obvious and very real practical disadvantages resulting from its small size when experiments call for the isolation of proteins, membranes, subcellular fractions etc, in order to benefit from its extraordinary experimental advantages as a model system in molecular genetics. One can safely predict that in the next decade studies with Arabidopsis will provide major breakthroughs in our understanding of most aspects of plant physiology and developmental biology. The importance of this knowledge for plant breeding and therefore for a sustainable highly productive agriculture cannot be overestimated. We therefore expect that this book will provide valuable guidelines to all those who are planning experiments aimed at understanding various aspects of plant growth, productivity and interactions with the environment. The book offers a wealth of methodical and theoretical information as well as valuable references.
