Book excerpt: Amino acids (AAs) are not only building blocks of protein, but are also signalling molecules as well as regulators of gene expression and the protein phosphorylation cascade. Additionally, AAs are key precursors for syntheses of hormones and low-molecular weight nitrogenous substances with each having enormous biological importance. For example, physiological concentrations of AA metabolites (e.g., nitric oxide, polyamines, glutathione, taurine, thyroid hormones, and serotonin) are required for cell functions. Growing evidence shows that humans and animals have dietary requirements for all proteinogenic AAs. Mammals, birds and fish also have species- and age-dependent needs for some AA-related substances. However, elevated levels of other products (e.g., ammonia, homocysteine, H2S, and asymmetric dimethylarginine) are pathogenic factors for neurological disorders, oxidative stress, and cardiovascular disease. Thus, optimal amounts of AAs and their ratios in diets and circulation are crucial for whole body homeostasis and health. Adequate provision of one or a mixture of functional AAs or metabolites may be beneficial for ameliorating health problems at various stages of the life cycle (e.g., fetal growth restriction, neonatal morbidity and mortality, weaning-associated intestinal dysfunction and wasting syndrome, obesity, diabetes, cardiovascular disease, the metabolic syndrome, and infertility). Dietary supplementation of these nutrients can also optimize the efficiency of metabolic transformations to enhance muscle growth, milk production, and athletic performance, while preventing excess fat deposition and reducing adiposity. Therefore, functional AAs hold great promise in improving the growth, health and well-being of individuals. They can support the biosynthesis of a myriad of natural products including nonprotein amino acids, cyanogenic glycosides, phar macologically active alkaloids, certain phenols, purines and pyrimidines, nucleic acids, condensed tannins, lignins and other metabolites. The approximately twenty or so amino (and imino) acids that comprise proteins are well known; less familiar are what is now approaching 600 nonprotein amino acids that have been isolated and characterized from plant, fungal or animal sources. Investigations of the protein amino acids have proven of outstanding value in enhancing our understanding of a variety of physiological and neurological topics that affect human health and well being. Book excerpt: The Joint FAO/IAEA Division of Nuclear Techniques in Food and Agriculture has a long history of coordinating isotope aided research projects for improving animal productivity in developing countries. These projects have focused on enhancing the efficiency of animal production through better use of locally available feed resources and those that do not compete with human food. As a part of these efforts, the Joint Division has been actively involved in developing and simplifying approaches for measuring microbial protein supply to ruminant livestock. The contribution of microbial protein in meeting the overall requirement for protein in ruminants is significant, but understanding the contribution of microbial protein to the nutrition of ruminant livestock has been hampered by the lack of simple and accurate methods for measuring microbial protein production in vivo. A Consultants’ Meeting was held in 1995 in Vienna to explore the feasibility of using nuclear and related techniques to develop and validate new approaches for measuring microbial protein supply in ruminant animals. Based on the available information, the consultants considered that the purine derivative excretion method, which is non-invasive and both simple to use and inexpensive compared with other available methods, had the potential for use in developing countries. However, development of models for various animal species and breeds and their validation were required before this technique could be advocated for use.
