Book excerpt: Prolonged exposure of G-protein coupled receptors to their agonists often leads to a rapid loss of signaling ability. The molecular basis for desensitization of G-protein coupled receptors was studied using the m2 subtype of muscarinic acetylcholine receptor (m2 mAChR) as a model. Previous studies have demonstrated that the m2 mAChR is rapidly phosphorylated following agonist exposure, and that this phosphorylation correlates with the ability of the m2 mAChR to bind arrestin proteins, and an inability of the receptor to interact with G-proteins. Internalization of the m2 mAChR from the plasma membrane is also associated with receptor desensitization following agonist exposure. In this dissertation, the molecular events underlying m2 mAChR desensitization were studied. These experiments demonstrated a requirement for G-protein couples receptor kinase activity in agonist-dependent phosphorylation of the m2 mAChR, and in the subsequent desensitization of the receptor. Structure-function analysis identified a cluster of serine and threonine residues which are the sites for agonist-dependent phosphorylation and desensitization of the m2 mAChR. The structural requirements for m2 mAChR desensitization correlated with the determinants of arrestin binding. Finally, the role of receptor phosphorylation and arrestin binding in m2 mAChR internalization was investigated, and novel structural requirements were observed for internalization versus desensitization of the receptor.
