Book excerpt: Chromatographic & Electrophoretic Techniques, Fourth Edition, Volume II: Zone Electrophoresis presents the methods of zone electrophoresis. This book discusses all the techniques that can be applied routinely. Organized into 17 chapters, this volume begins with an overview of the fundamental principle of zone electrophoresis. This text then examines the apparatus in common use for paper electrophoresis and discusses paper electrophoresis at low voltages. Other chapters describe the apparatus that combines several advantages resulting from high voltage paper electrophoresis and thin-layer chromatography. This book discusses as well the advantages of cellulose acetate membrane filter paper as a supporting medium for zone electrophoresis. The final chapter deals with preparative acrylamide gel electrophoresis on a small scale, which may involve either continuous electrophoretic elution or extraction of the RNA from zones cut out of the gel. This book is a valuable resource for biochemists, clinical chemists, clinicians, technicians, biophysicists, pathologists, and medical researchers.

Book Applications of Infrared Spectroscopy in Biochemistry Biology and Medicine

Download or read book Applications of Infrared Spectroscopy in Biochemistry Biology and Medicine written by Frank Parker and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 612 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book is not intended to be a basic text in infrared spectroscopy. Many such books exist and I have referred to them in the text. Rather, I have tried to find applications that would be interesting to a variety of people: advanced undergraduate chemistry students, graduate students and research workers in several disciplines, spectros copists, and physicians active in research or in the practice of medicine. With this aim in mind there was no intent to have exhaustive coverage of the literature. I should like to acknowledge my use of several books and reviews, which were invaluable in my search for material: G. H. Beaven, E. A. Johnson, H. A. Willis and R. G. 1. Miller, Molecular Spec troscopy, Heywood and Company, Ltd., London, 1961. J. A. Schell man and Charlotte Schellman, 'The Conformation of Polypeptide Chains in Proteins," in The Proteins, Vol. II, 2nd Ed. (H. Neurath, ed.), Academic Press, New York, 1964. R. T. Book Physical Principles and Techniques of Protein Chemistry Part A

Download or read book Physical Principles and Techniques of Protein Chemistry Part A written by Sydney Leach and published by Elsevier. This book was released on 2012-12-02 with total page 545 pages. Available in PDF, EPUB and Kindle. Book excerpt: Physical Principles and Techniques of Protein Chemistry, Part A deals with the principles and application of selected physical methods in protein chemistry evaluation. This book is organized into nine chapters that cover microscopic, crystallographic, and electrophoretic techniques for protein conformational perturbations evaluation. This text first presents a general account of electron microscopy, its specimen preparation, optimum conditions for high resolution, measurement of electron micrographs, and illustrative examples of protein study. This book then examines the different types of maps from X-ray methods and the diffraction data from fibrous proteins. The subsequent chapters cover discussions on UV spectroscopy of proteins; luminescence properties of proteins and related compounds; and perturbation and flow methods for evaluation of proteins' dynamic properties and rate constants. Other chapters deal with the evaluation of proteins' dielectric properties using dielectric relaxation, electric birefringence, and dichroism techniques. The concluding chapters outline the theoretical and experimental advances of the electrophoretic and gel filtration methods for the study of protein structure and molecular weight. This book is of great value to chemists, biologists, and researchers who have great appreciation of protein chemistry.

Book Physical and Chemical Methods

Download or read book Physical and Chemical Methods written by Curtis A. Williams and published by Academic Press. This book was released on 2014-05-12 with total page 480 pages. Available in PDF, EPUB and Kindle. Book excerpt: Methods in Immunology: Volume II, Physical and Chemical Methods is a collection of papers dealing with electrophoresis, analytical ultracentrifugation, dialysis, ultrafiltration, cellulose ion exchangers, and chromatographic separation of macromolecules on porous gels. Some papers explain the applications of radioisotopes, optical analysis, and chemical analysis of proteins, carbohydrates, lipids, and nucleic acid. One paper describes the theory of electro-migration. Factors such as electrical charge or frictional coefficients govern the rate of migration of charged particles in an electric field. The differences found in their velocities can be used to separate substances or analyze them. Mobility is a characteristic property of molecules and can also be influenced by the composition of the medium or solution. Dialysis separates solvents too large to diffuse through a barrier from smaller solutes; ultrafiltration (reverse osmosis) forces solvent and solutes up to a certain critical size through the barrier by a high pressure on one side. The book notes that the membrane never becomes plugged in dialysis because of some opposite movement of the solvent. Another paper points out that the significance of radioactive tracers in immunochemistry employed to identify and label macromolecules functioning as antigens and antibodies. The collection can prove valuable to bio-chemists, cellular biologists, micro-biologists, developmental biologists, and scientists involved in immunological research.

The next chapter describes the general experimental methods for the separation of abnormal hemoglobins and surveys the application of paper electrophoresis to the final identification of a hemoglobin variant. The remaining chapters deal with the principles, apparatus, reagents, and applications of other zone electrophoretic techniques, including cellulose acetate; agar gel, starch block, starch gel, and sponge rubber; high voltage paper; and continuous electrophoresis. This book will prove useful to analytical chemists and biologists.
