Book excerpt: Achieving direct and selective functionalization of carbon-hydrogen (C-H) bonds to give carbon-carbon (C-C) or carbon-heteroatom (C-Y) bonds is a significant and long-standing goal in chemistry. C-H bonds are attractive reaction partners because they are ubiquitous in organic molecules. Thus, C-H functionalization methods could potentially expedite the synthesis of target molecules by providing new disconnections in retrosynthetic analysis. Among the numerous methods to affect this transformation, palladium-catalyzed C-H functionalization is one of most promising methods to construct C-C and C-Y bonds in term of versatile reactivity. The major challenges of palladium-catalyzed C-H functionalization are developing reactions that work with common and useful structural motifs and discovering new transformation, such as C-N or C-F bond formation. This thesis explores palladium-catalyzed C-H bond functionalization with substrates containing simple functional groups such as carboxylic acids and triflamides, which direct C-H cleavage through weak coordination with the metal catalyst. Chapter one introduces different types of C-H bond functionalization and focuses on Pd(II)/Pd(IV) catalysis. Chapter two covers Pd-catalyzed C-H iodination of arene carboxylic acids enabled by the discovery of coutercation-promoted C-H activation. Weak coordination has also been found to enable versatile reactivity of simple arene carboxylic acids. Chapter three focuses on the development of practical and useful C-H fluorination using triflamide as a weakly coordinating directing group that can be easily manipulated to a wide range of useful and common aryl halides. Chapter four describes applications of bystanding F+ oxidants to promote selective C-N reductive elimination in Pd(II)/Pd(IV) catalysis.

Book excerpt: In 1972, a very powerful catalytic cycle for carbon-carbon bond formation was 2 first discovered by the coupling reaction of Grignard reagents at the sp -carbon. Over the past 30 years, the protocol has been substantially improved and expanded to other coupling reactions of Li,B,N,O,Al,Si,P,S,Cu,Mn,Zn,In,Sn, and Hg compounds. These reactions provided an indispensable and simple methodology for preparative organic chemists. Due to the simplicity and rel- bility in the carbon-carbon, carbon-heteroatom, and carbon-metalloid bo- formations,as well as high efficiency of the catalytic process,the reactions have been widely employed by organic chemists in various fields. Application of the protocol ranges from various syntheses of complex natural products to the preparation of biologically relevant molecules including drugs, and of sup- molecules, and to functional materials. The reactions on solid surfaces allow robot synthesis and combinatorial synthesis. Now, many organic chemists do not hesitate to use transition metal complexes for the transformation of org- ic molecules. Indeed, innumerable organic syntheses have been realized by the catalyzed reactions of transition metal complexes that are not achievable by t- ditional synthetic methods. Among these, the metal-catalyzed cross-coupling reactions have undoubtedly contributed greatly to the development of such a new area of “metal-catalyzed organic syntheses”. An excellent monograph for the cross-coupling reactions and other met- catalyzed C-C bond-forming reactions recently appeared in Metal-catalyzed Cross-coupling Reactions (Wiley-VCH,1998).

Part I describes the Pd-catalyzed activation/functionalization of inert alkyl C(sp3 )-H bonds through the development of highly reactive directing groups and identification of compatible ligands that cooperatively promote C(sp3 )-H functionalization. Prior to this dissertation research, methods to selectively functionalize methyl and methylene C(sp 3 )-H bonds at the [beta]-position of carbonyl groups were underdeveloped. Inspired by the indispensable roles of electronically and sterically optimized phosphine (PR3 ) and N- heterocyclic carbene (NHC) ligands in traditional cross-coupling reactions, we have invented a class of highly optimized N- arylamide directing groups that weakly coordinate to Pd, yet promote cleavage of [beta] -C(sp3 )-H bonds and facilitate subsequent conversion into an array of C-C and C-heteroatom bonds. A unique feature of Pd-catalyzed C(sp3 )-H activation reactions using N- arylamide directing groups is that a range of coordinating moieties that are known to compete for the coordination sites around Pd (e.g. , PR3 , NHC, pyridine, CO, acrylates, and DMF) can be accommodated, while maintaining robust C(sp 3 )-H activation. By exploiting the N- arylamide directing group, we have developed the following unprecedented transformations, which will be summarized in five discrete chapters: 1. Pd(0)/PR3 -catalyzed intermolecular arylation of [beta]-C(sp3 )-H bonds. 2. Pd(II)-catalyzed olefination of [beta] -C(sp3 )-H bonds. 3. Pd(II)-catalyzed carbonylation of [beta]-C(sp3 )-H bonds. 4. Pd(II)/amino acid-catalyzed enantioselective activation of cyclopropyl C-H bonds. 5. Pd(II)/quinoline-catalyzed arylation of [beta]-methylene C(sp3 )-H Bonds. Part II of this dissertation addresses applications of this same class of amide directing groups for C(sp2 )-H activation reactions in developing previously unprecedented C(sp2 )-H functionalization reactions. In particular, we have focused our efforts on the following two topics: 1. Pd(II)-catalyzed synthesis of lactams via intramolecular C(sp 2 )-H amination. 2. Pd(0)/PR3 -catalyzed N- arylamide-directed regioselective C-H arylation of pyridines. The versatility and practicality of these reactions in their current forms are evaluated with respect to the efficiency of catalysis and substrate scopes. Key problems and potential solutions are also discussed.
