Book excerpt: Our aim is to identify epitopes on the surface of prostate cells that are largely or exclusively tissue- or tumor- specific. This is done by expansion of T cells reactive for HLA-restricted, established tumor lines from the peripheral blood cells and tumor-infiltrating lymphocytes of cancer patients. Towards this end, we established a reagent bank of patient material that includes tissue, cells, and serum. We successfully and stably gene-modified the established prostate cell line, LNCaP, so that it expresses HLA-A2 and HLA-Al. We successfully transfected two other cell lines with these genes, transiently but sufficiently, for our in vitro stimulation technique. Several potentially HLA-restricted, tumor-specific T cells have been identified and are currently being cloned for retesting. In order to study tissue distribution, primary normal and tumor prostate tissue are being cultured by methods developed in our laboratory. In addition, we are performing an antibody-based method of antigen discovery that utilizes patient serum. Using this approach, several genes have been identified some of which represent previously unreported, novel genes. Studies are underway to discern the exact epitope being recognized. Studies are also underway to assess the level of gene expression in normal vs. tumor tissue from representative regions of the body.

Book excerpt: The purpose of the research is to clone, isolate and characterize protein markers that are specifically expressed by advanced stage, androgen-independent prostate cancers. Two types of markers with this specificity are expected to be identified: (1) proteins secreted specifically by advanced stage prostate cancer cells, potentially useful in serum-based assays designed to specifically detect this class of tumors; and (2) markers expressed specifically on the surface of these cells, potentially useful as specific targets for clinical treatment of androgen-independent prostate cancer. Progress to date has involved: (1) preparation of membrane fraction mRNA's from four human cell lines representing four stages of prostate cancer, and use of these mRNA's to prepare cDNA libraries; (2) isolation of proteins from membrane fractions of two of these cell lines, and initiation of a collaboration to employ the techniques of Proteomics to differentially analyze these protein populations. We can now apply the new DNA microarray technology to the cDNA libraries described in (1) to identify clones possessing the characteristics described above. In addition, we are now employing the proteins described in (2) above for differential screening via two-dimensional gel analysis, to directly identify cell surface proteins specifically expressed by advanced stage prostate cancer cells.

Book excerpt: The major goal of the proposed research is to develop a novel methodology for improving T cell epitopes. The underlying hypothesis is that T cells capable of recognizing tumor- associated antigens (TAA) are present but often difficult to activate. However, once activated such T cells might be effective against tumors due to the less stringent triggering requirements of mature effectors. We proposed to develop a novel bacterial expression system for modifying and screening the epitopes of PSA, a known TAA. In the current year, we have employed a saturation mutagenesis technique to a PSA peptide epitope we have identified. Expression libraries were constructed corresponding to each position in the peptide. These libraries were screened functionally, The clones showing enhanced activity were sequenced. From these screens, we were able to identify a number of enhanced peptide epitopes. Peptides were synthesized and tested in a functional assay. These peptides showed enhanced activity with the T cell hybridoma. The findings show that altered peptide ligands can be discovered using the novel methodology proposed. These approaches should be applicable to other tumor antigens.

These proteins enhance the growth of cancer cells and protect tumors from treatments such as drugs or surgery. However, new drugs have recently been developed particularly those targeting heat shock protein 90. As heat shock protein 90 functions to stabilize many of the oncogenes and growth promoting proteins in cancer cells, such drugs have broad specificity in many types of cancer cell and offer the possibility of evading the development of resistance through point mutation or use of compensatory pathways. Heat shock proteins have a further property that makes them tempting targets in cancer immunotherapy. These proteins have the ability to induce an inflammatory response when released in tumors and to carry tumor antigens to antigen presenting cells. They have thus become important components of anticancer vaccines. Overall, heat shock proteins are important new targets in molecular cancer therapy and can be approached in a number of contrasting approaches to therapy.
