Available in PDF, EPUB and Kindle. Book excerpt: Jean BERNARD * I should like to begin with an assumption and a paradox. The assumption is that leukemia is a disease of a stem cell characterized by pathologie alterations of that cell and its progeny. All present research and discussions are centered around the leukemic cell. So is this symposium, which would not take place except for our primary interest in the leukemic cell. This does not preclude, of course, consideration of other definitions and other approaches to the prOblem. By definition, then, the leukemic cells are abnormal cells and their metabolism and functions are presumed to be abnormal. Yet, the classification of the different types of leukemias is based upon the characteristics of normal cells. We talk of "lymphoblasts" and "myeloblasts" as predominant cell types in leukemia. This leads to a double paradox. In the first pi ace it is clearly illogical to classify abnormal cells by their resemblance to normal cells, since their very abnormality consists in not being normal. Yet, as a second paradox, the classifica ti on has had the happy consequence of ai ding us in the treatment and prognosis of leukemia for the past 25 years. A more detailed analysis shows that the consequence of this paradox are complex: while there exists a useful correlation between cellular types, treatment and prognosis, numerous problems and difficulties persist. The most serious of them concems the "unclassified leukemias" which are the reason for this reunion.

Book Dynamic Morphology of Leukemia Cells

Download or read book Dynamic Morphology of Leukemia Cells written by H. Felix and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 197 pages. Available in PDF, EPUB and Kindle. Book excerpt: Dynamic Morphology is the attempt to correlate surface architec ture and shape of fixed cells, as visualized by scanning electron microscopy (SEM), with the behavior of living cells, recorded by microcinematography (MCM). If SEM and MCM are used concurrently for the analysis of cell populations, a dynamic inter pretation of SEM photographs is only valid if the experimental conditions are identical for the two techniques. This is achieved by allowing the cells to settle on a glass surface where they remain long enough to perform their various activities under conditions identical for both techniques (for technical details see Methodology). The analysis of a population necessitates the study of a large number of cells. This prerequisite is met by operating the scanning electron micro scope at low levels of magnification, and by using culture chambers for cinematography. It can be argued that the examina tion of attached cells excludes a complete SEM survey of a population, as cells not adhering from the outset or becoming detached during the different preparatory steps are lost. For this, cinematography proved to be a reliable control: All cell types recognized in time-lapse films were also seen in scanning electron (SE) micrographs. Another, and more general, objection to a dynamic interpretation concerns the artificiality of cellular behavior on glass. This is true, but does not invalidate compara tive studies making use of this substrate.

Available in PDF, EPUB and Kindle. Book excerpt: In spite of old vintage and 200 years of clinical use, digitalis remains an interesting therapeutic agent, to clinicians as well as to the pharmacologist, the biochemist, and colleagues in other diciplines of theoretic medicine. When a drug, however, has so many attractive facets, it seems proper and aJvis able for the success of a scientific meeting to focus on a number of well-defined aspects. This symposium was devoted to pharmacokinetics, drug metabolism, analytic procedures, blood level determinations, and their interpretation both for thera peutic and toxic situations. Considerable progress has been made during the last years in this area of digitalis research. The time was suitable for a critical reap praisal of facts and theories and for future planning. Our main intention was to relate analytic data and biochemical findings to clinical problems and questions. Despite the undoubtedly basic character of clinical pharmacology, it is neverthe less an applied science which should help to develop the rational basis of thera peutics. We are particularly grateful to the active participants who bore the burden of preparing presentations and - even worse - manuscripts. At the same time we are well-aware that many other active research groups would have been able to contribute in this way, but our program was limited because of the short time available. Their knowledge is included in the discussion parts of the meeting, so we hope a well-balanced description of the present state of affairs emerged in this volume.

Available in PDF, EPUB and Kindle. Book excerpt: Hemolysis during filtration through micropores studied by Chien et al. [I] showed a dependence on pressure gradient and pore diameter that, at the time of publication, did not permit an easy interpretation of the hemolytic mechanism. Acting on the assumption that thresholds of hemolysis are easier to correlate with physical forces than extents of hemolysis, we performed a series of experi ments repeating some of the conditions reported in [I] and then focusing on low L1P in order to define better the thresholds of hemolysis for several pore sizes. Employing a model of a deformed red cell shape at the pore entrance (based on micropipette observations) we related the force field in the fluid to a biaxial tension in the membrane. The threshold for lysis correlated with a membrane tension of 30 dynes/cm. This quantity is in agreement with lysis data from a number of other investigators employing a variety of mechanisms for introduc ing membrane tension. The sequence of events represented here is: a. Fluid forces and pressure gradients deform the cell into a new, elongated shape. b. Extent of deformation becomes limited by the resistance of the cell mem brane to undergo an increase in area. c. Fluid forces and pressure gradients acting on the deformed cell membrane cause an increase in biaxial tension in the membrane. d. When the strain caused by this tension causes pores to open in the membrane, the threshold for hemolysis has been reached [2].

Book Chemoreception in the Carotid Body

Download or read book Chemoreception in the Carotid Body written by H. Acker and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 302 pages. Available in PDF, EPUB and Kindle. Book excerpt: It is becoming traditional that periodically those of us interested in the carotid body hold an international meeting to discuss their results. In 1966 a meeting was organized by R. W. Torrance in Oxford and in 1973 by M. J. Purves in Bristol; in 1974 A. S. Paintal organized a satellite symposium of the Physiological Congress in Kashmir. The organizers of these meetings are to be commended for their efforts in publishing both the papers and discussions. At these meetings it has become apparent that the direction of research is becoming more sharpely focused on the cellular mechanisms of chemoreception. During the meeting in Dortmund the papers dealt mostly with the different cell types in the carotid body and their environment, i. e. , local P0 and local flow. These included 2 light and electron microscopic studies of the morphometric and histochemical pro perties of the different cells, microelectrode studies of the glomoid tissue to understand the conversion of a chemical stimulation into nervous activity, as well as strictly bio chemical and physiologic investigations concerning the dependence of the chemo receptive process on O consumption and the turnover of catecholamines. In spite 2 of the variety in methodology, it was apparent that all contributors had a common interest: to understand the mechanisms of chemoreception. Although at the meeting itself there was ample time to fully discuss the various papers, it has become necessary here to shorten the papers and discussions; otherwise the cost of publication would have been prohibitive.
