Book excerpt: This volume is the fourteenth classified bibliography of organic, organometallic and metal complex crystal structures prepared by the Cambridge Crystallo graphic Data Centre and published jointly with the International Union of Crystallography. The previous thirteen volumes covered the years 1935-81; the majority of references in the present volume pertain to structure analyses reported in the literature during 1981 and 1982. A few structures reported prior to 1981 and omitted in earlier volumes are also included. Volume 14 contains 4094 references to 4001 distinct chemical compounds with 2162 cross-references. Some 90% of these references were obtained by direct in-house scanning of 51 major journals; the remaining material was located by searching Chemical Abstracts and Bulletin Signa/etique. The table below summarizes the cut-off dates for the 25 direct-scan journals yielding most entries in Volume 14. The fully classified Chemical Diagram Index, first presented in Volume 13, is continued here. Volume 14 contains 3912 chemical structural diagrams, repre senting 98% of the compounds. The reasons for the omission of some diagrams are noted in the Introduction. This new index provides a rapid visual overview of compounds studied in 1981-82 and will, we hope, add greatly to the usefulness of the annual Bibliographies. The textual indexes in Volume 14 continue the system established in Volumes 9-13 and in the special volume Guide to the Literature 1935-76. The latter volume presents a set of cumulative indexes to the contents of Bibliographic Volumes 1-8.

The main focus is on reactions in solution, although solid-state and gas-phase studies are included where they provide relevant mechanistic insight. Each volume covers an eighteen-month literature period, and this, the eighth volume in the series, includes papers published during January 1990 through June 1991. Where appropriate, references to earlier reports and to specific sections in previous volumes are given. Coverage spans the whole area as comprehensively as possible in each volume, and while it is impossible to be absolutely exhaustive, every effort is made to include all of the important published work that is relevant to the elucidation of reaction mechanisms. Numerical data are reported in the units used by the original authors, and they are converted to common units only when comparisons are being made. The successful format of earlier volumes is retained to facilitate tracing progress over several years in a particular topic, and the series now permits this to be done for a twelve-year period. The introduction three volumes ago of computerized techniques to improve cross-referencing in the Index brought positive reader comments, and their use is being continued.

This book is intended to reminisce this event, to take inventory, and to point out new lines of development in this field. Divided in 6 sections and 22 chapters, the book provides an up-to-date account on topics such as - the chemistry and biochemistry of cisplatin, - the clinical status of Pt anticancer drugs, - the impact of cisplatin on inorganic and coordination chemistry, - new developments in drug design, testing and delivery. It also includes a chapter describing the historical development of the discovery of cisplatin. The ultimate question - How does cisplatin kill a cell? - is yet to be answered, but there are now new links suggesting how Pt binding to DNA may trigger a cascade of cellular reactions that eventually result in apoptosis. p53 and a series of damage recognition proteins of the HMG-domain family appear to be involved. The book addresses the problem of mutagenicity of Pt drugs and raises the question of the possible relevance of the minor DNA adducts, e.g. of interstrand cross-links, and the possible use of trans-(NH3)2Pt(II)-modified oligonucleotides in antisense and antigene strategies. Our present understanding of reactions of cisplatin with DNA is based upon numerous model studies (from isolated model nucleobases to short DNA fragments) and application of a large body of spectroscopic and other physico-chemical techniques. Thanks to these efforts there is presently no other metal ion whose reactions with nucleic acids are better understood than Pt. In a series of chapters, basic studies on the interactions of Pt electrophiles with nucleobases, oligonucleotides, DNA, amino acids, peptides and proteins are reported, which use, among others, sophisticated NMR techniques or X-ray crystallography, to get remarkable understanding of details on such reactions. Reactivity of cisplatin, once bound to DNA and formerly believed to be inert enough to stay, is an emerging phenomenon. It has (not yet) widely been studied but is potentially extremely important. Medicinal bioinorganic chemistry - the role of metal compounds in medicine - has received an enormous boost from cisplatin, and so has bioinorganic chemistry as a whole. There is hardly a better example than cisplatin to demonstrate what bioinorganic chemistry is all about: The marriage between classic inorganic (coordination) chemistry and the other life sciences - medicine, pharmacy, biology, biochemistry. Cisplatin has left its mark also on areas that are generally considered largely inorganic. The subject of mixed-valance Pt compounds is an example: From the sleeping beauty it made its way to the headlines of scientific journals, thanks to a class of novel Pt antitumor agents, the so-called "platinum pyrimidine blues". In the aftermath diplatinum (III) compounds were recognized and studies in large numbers, and now an organometalic chemistry of these diplatinum (III) species is beginning to emerge. The final section of the book is concerned with new developments such as novel di- and trinuclear Pt(II) drugs with DNA binding properties different from those of cisplatin, with orally active Pt(IV) drugs which are presently in clinical studies, and with attempts to modify combinatorial chemistry in such a way that it may become applicable to fast screening of Pt antitumor drugs. The potential of including computational methods in solving questions of Pt-DNA interactions is critically dealt with in the concluding chapter.

The main objective of the Centre is to assemble a computer-based File containing both bibliographic information and numerical data abstracted from the Iiterature and relevant to molecular and crystal structures, as obtained by diffraction methods. The File is designed to permit the checking of published numerical values, particularly interatomic distances and bond angles and to present published results in a variety of new ways. Contents of the File will be made available through the series "Molecular Structures and Dimensions" and also by the provision of data on magnetic . tapes or discs. At present the numerical data relating to molecular dimensions are being checked and will be published, at a later date, as further volumes in this series and also as magnetic tapes. The File is being extended to cover electron diffraction in the gas phase.
