Book excerpt: Growing interest in understanding the fundamental processes of bacterial cellulose degradation for the generation of cellulosic ethanol has resulted in the study of the Cellulomonadaceae, a family of cellulolytic bacteria that includes the only reported bacteria capable of degrading and utilizing cellulose both aerobically and anaerobically. This could thus represent a novel strategy for cellulolytic activity, as the most common strategies for aerobic and anaerobic cellulose degradation utilize free or attached cellulases, respectively. These strategies are functionally distinct with any given bacterium employing only one approach. Therefore, bacteria capable of both aerobic and anaerobic cellulose degradation provide a unique opportunity for understanding bacterial strategies for cellulose degradation. This thesis investigates the strategy that cellulomonads use for cellulose degradation by first determining that all 19 species of Cellulomonas tested in this work are capable of degrading cellulose and untreated switchgrass, a more complex lignocellulosic substrate, under both aerobic and anaerobic conditions. Furthermore, these bacteria may be capable of producing ethanol as a byproduct of this metabolism. Investigation into how these bacteria degrade cellulose showed that overall, fewer cellulases were excreted under anaerobic conditions as compared to aerobic conditions, leading to the hypothesis that Cellulomonas switch their cellulolytic strategy in response to oxygen availability. This hypothesis was further examined through a combination of genomic analyses and physiologic assays on Cellulomonas gilvus, a model cellulomonad. Experiments were performed on the motility, chemotactic, cellulase localization, and fiber adherence capabilities of C. gilvus during cellulose degradation, the results of which suggest that C. gilvus degrades cellulose using a novel hybrid of the canonical aerobic and anaerobic strategies and provides further insights into the metabolism of this organism. These insights into the cellulose degradation capabilities of the genus Cellulomonas were then used to help identify and characterize a novel cellulose degrading bacterium isolated by freshmen at the University of Wisconsin-Madison as part of an introductory course. After characterization, this bacterium was named Cellulomonas wisconsinensis as a novel species of the genus Cellulomonas isolated in Wisconsin.
