The study of the molecular mechanism responsible for the crown gall phenomenon (induced by Agrobacterium tumefaciens) illustrates the point that the fundamental study of the cause(s) and mechanism(s) of abnormal growth might be one of the most efficient ways to understand cellular differentiation and the molecular basis of gene expression. The book is organized into three parts that contain research on abnormal plant growth, crown gall tumors, and potential vectors for genetic engineering in agriculture. The genetic structure responsible for the neoplasmic transformation of plant cells in crown galls is a bacterial plasmid (called Ti for tumor-inducing). Research described in this volume demonstrates that these Ti plasmids were designed by evolution as natural gene vectors with which some bacteria can introduce active genes into plants. These transferred genes are maintained by integration in the plant genome and their expression is directly or indirectly responsible for the tumorous growth pattern.

The purpose of the workshop was to bring together experts in symbiosis, plant pathology and plant molecular biology in order to discuss recent progress in the field of microbe -plant re cognition at the molecular level, to promote integration of various disciplines, and to define recommendations for future research and applications. Plants have developed a variety of sophisticated defence mechanisms to cope with an environment in which many different microbes live. Most microbes which colonize plant tissues are harmless. Some microbes have developed ways to attack plants successfully, resulting in enormous losses of crop yields. Other microbes have reached an agreement with the host plant which is beneficial for both: these microbes live in symbiosis with the plant and provide their host plant for example with substantial amounts of atmospheric nitrogen. Chemical protection of crops is a necessity in modern crop management but this treatment has some negative effects as well. Therefore scientists are looking for alternative, biological, ways to control crop pests. Against this background specialists from eleven countries discussed the results of their most recent work on the molecular background of microbe -plant interactions. It appeared that, in order to capitalize the recent rapid progress made in the mole cular genetical studies on Rhizobium-legume and pathogen-host plant interactions, a multidisciplinary approach is required.

This book gives a comprehensive overview on our present molecular biological knowledge about the Rhizobiaceae, which currently can be called the best-studied family of soil bacteria. Some of these associations are beneficial for the plant, as is the case for the Rhizobiaceae subgroups collectively called rhizobia, which are able to fix nitrogen in a symbiosis with the plant hosts. The molecular machinery, needed for T-DNA generation and transport into the host cell and encoded by a series of chromosomal (chv) and Ti-plasmid virulence (vir) genes, has been the subject of numerous studies over the past several decades. Today, Agrobacterium is the tool of choice for plant genetic engineering with an ever expanding host range that includes many commercially important crops, flowers, and tree species. Furthermore, its recent application for the genetic transformation of non-plant species, from yeast to cultivated mushrooms and even to human cells, promises this bacterium a unique place in the future of biotechnological applications. Book excerpt: The broad host range pathogenic bacterium Agrobacterium tumefaciens has been widely studied as a model system to understand horizontal gene flow, secretion of effector proteins into host cells, and plant-pathogen interactions. Agrobacterium-mediated plant transformation also is the major method for generating transgenic plants for research and biotechnology purposes. Agrobacterium species have the natural ability to conduct interkingdom genetic transfer from bacteria to eukaryotes, including most plant species, yeast, fungi, and even animal cells. In nature, A. tumefaciens causes crown gall disease resulting from expression in plants of auxin and cytokinin biosynthesis genes encoded by the transferred (T-) DNA. Gene transfer from A. tumefaciens to host cells requires virulence (vir) genes that reside on the resident tumor-inducing (Ti) plasmid. In addition to T-DNA, several Virulence (Vir) effector proteins are also translocated to host cells through a bacterial type IV secretion system. These proteins aid in T-DNA trafficking through the host cell cytoplasm, nuclear targeting, and T-DNA integration. Genes within native T-DNAs can be replaced by any gene of interest, making Agrobacterium species important tools for plant research and genetic engineering. In this research topic, we provided updated information on several important areas of Agrobacterium biology and its use for biotechnology purposes.
