Neutrophil recruitment was found to be dependent upon signaling through MyD88, an adaptor protein used in PRR signaling. Of the PRRs, several Toll-like receptors (TLR) have been shown to be dependent upon MyD88 for signaling. In vitro studies demonstrated that SR can interact with TLR2, but not TLR4, 5, or 7. Exposure of TLR2-/- mice to SR resulted in neutrophil recruitment into the lungs at similar levels as WT controls. These results suggest that signaling through TLR2 it is not necessary for neutrophil recruitment into the lung. Thus, additional receptors upstream of MyD88 interact with SR resulting in production of proinflammatory cytokines and chemokines, which leads to subsequent neutrophil recruitment.
