Book excerpt: In order to fulfill the growing need for agility and flexibility and to handle increasing production complexity, pharmaceutical manufacturing's strategic importance has risen. However, many companies are just starting to discover the potential and benefits of Operational Excellence initiatives. This thesis has two main parts: a theoretical base, which analyzes the development of integrated production systems and explores their origins in post-war Japan, as well as discussion of OPEX initiative elements across the whole industry scale. The second section is comprised of an exhaustive operational excellence questionnaire focusing on OPEX initiatives within six globally active pharmaceutical companies. Every company surveyed was different; research and literature has not yet been able to provide an answer about the correct structure and the number of people within a company needed to sustainably implement an OPEX initiative. Making a fundamental effort to change company philosophy and empowering employees to realize their own and the company's full potential should be the long-term goal of a meaningful OPEX effort. This vision must then be translated into reality through structures within the organization: through middle managers, who serve as a crucial link between corporate and site level, project teams who are evaluated according to project targets and employees at all levels who are granted sufficient time and resources to fully support and sponsor the initiative in all of its stages. Finally, promising organizational success factors are visualized in an "OPEX implementation house" model.

With the increase in the global demand of drugs through demographic change and the medicine demand of emerging countries, the required quantity of pharmaceuticals will increase significantly in upcoming years. The industry struggles with the dual problem of drug shortages - which have quadrupled in recent years - and an increasing number of drug recalls. In general, regulatory pressure and current regulatory changes are forcing the pharmaceutical industry to develop a stable quality oriented production system. Thus, in the increasingly complex environment of the pharmaceutical industry, companies need to take a closer look at their quality organisation in order to stabilise their internal production system and increase their flexibility and overall performance. Due to this need, the research focuses on the investigation of the interdependencies of quality and excellence to leverage the advantages of both. Operational Excellence (OPEX), as an initiative and lived as a philosophy, leads to a better performance, more efficient use of resources and continuous improvement. Quality management approaches have a positive impact on quality, innovation and the overall performance. Even though well-known excellence models include quality as basic element, the examples from practice show that the pharmaceutical industry discusses these aspects self-sufficiently. The divide of quality and excellence is not a conceptual problem but an industry-made challenge. Based on an extensive literature review, a statistical evaluation, a benchmarking and a qualitative case study analysis, the thesis designs a descriptive model that builds the basis to structure an internal quality management system for pharmaceutical companies, using a systemic approach combining quality and excellence. Despite the importance of an integrated approach to drive business performance, scholars and practitioners have primarily focused on OPEX in manufacturing to create sustainable success. However, in the pharmaceutical industry the quality control (QC) lab constitutes a major bottleneck in the value creation. In recent time OPEX in QC labs has gained increased interest as a driver of the performance of the organization that has been overlooked to date. Literature contributes with generic excellence models. However, scholars have neglected to show their application in the specific context of QC labs of this research. The diverse performance measurement literature emphasizes that the approach to operational performance depends on the unit of analysis and cannot be transferred one-to-one from other areas. Today, the empirical work from academia and practice that discusses OPEX performance in pharmaceutical QC labs from an integrated perspective is scarce. Accordingly, this thesis addresses the outlined gap of knowledge with a scientifically conceptualized, practically operationalized, unified, and integrated approach to measure OPEX performance in pharmaceutical QC labs. The research employs a mixed-methods approach combining quantitative and qualitative research. The resource-based view (RBV) constitutes the scientific basis framing the research at hand. The thesis comprises three research stages. First, OPEX performance in pharmaceutical QC labs is conceptualized and operationalized. This stage concludes in an operationalized performance measurement model (PMM). Additionally, this stage yields in research propositions and hypotheses for the subsequent research stage. Second, a quantitative analysis provides new insight into the relation between the model dimensions and OPEX performance in QC labs. In particular, it focuses on the relation.

Book excerpt: The management of shop floor behavior plays a critical role for the success of operational improvement programs, which rely on the workers that perform the direct value creation tasks. But what precisely should managers do to influence that behavior constructively? In the corresponding stream of Operational Excellence (OPEX) literature, this question has recently experienced an increase in research interest. For pharmaceutical manufacturers, addressing this question can be considered mission critical for two main reasons. First, OPEX programs are viewed as an antidote for low quality levels in pharmaceutical manufacturing, which are significantly lower compared to other industries. Second, procedural regulation can affect what behaviors are deemed acceptable. This should be accounted for, as OPEX and regulatory compliance can have differing behavioral priorities. The purpose of this dissertation is to provide theoretical and practical guidance for impacting shop floor behavior with pharmaceutical OPEX programs to facilitate balanced and high levels of operational performance. For this purpose, this research draws on literature on OPEX, Management Control (MC) and structuration theory to develop a research model in a first step. A mixed-method approach combining qualitative analysis and quantitative hierarchical linear regression analysis is then applied to empirically study the determinants of shop floor behavior in the context of pharmaceutical OPEX programs. The research findings suggest that the interplay between structural and behavioral determinants of shop floor behavior can lead to unintended consequences that are context dependent. They further suggest that this interplay is not comprehensively accounted for as part of pharmaceutical OPEX programs, since the underlying determinants are typically managed in isolation. Finally, the regression results underline the importance of taking unintended consequences that result from said interplay seriously, suggesting that MC has significant positive and negative moderation effects, which can increase respectively decrease the performance returns from OPEX programs.
