Available in PDF, EPUB and Kindle. Book excerpt: The biological activation of dioxygen is a key reaction in biological systems. Enzymes involved in direct oxygen activation are oxidases and oxygenases. Multi-copper oxidases are an important class of oxidases reducing dioxygen in a four-electron reduction to water with concomitant one-electron oxidation of the reducing substrate. The progress in the characterization and understanding of the structure and function of these enzymes has advanced so tremendously over the last ten years that the publication of a book documenting these achievements has been overdue.Especially the recent discovery of a key role of the FET3 protein of Saccharomyces cerevisae, a multi-copper oxidase, in iron metabolism of this eukaryote has underpinned the function of the plasma multi-copper oxidase ceruloplasmin in vetebrate iron transport. The lately determined x-ray structure of human ceruloplasmin confirms its close structural relatedness to the plant multi-copper oxidases ascorbate oxidase and laccase and due to strong amino-acid sequence similarities has allowed to construct a useful model of the more distantly related blood-clotting factor VIII.This book contains review articles from experts in the field, dealing with modern spectroscopy, enzyme kinetics, bioinorganic chemistry, x-ray crystallography, electron transfer reactions, molecular biology, medical aspects and potential industrial applications of the three main members of multi-copper oxidases, i.e., laccase, ascorbate oxidase and ceruloplasmin.

Book Oxidases and Related Redox Systems

Download or read book Oxidases and Related Redox Systems written by Tsso E. King and published by Elsevier. This book was released on 2013-10-22 with total page 1295 pages. Available in PDF, EPUB and Kindle. Book excerpt: Oxidases and Related Redox Systems is a collection of papers from the Third International Symposium on Oxidases and Related Reduction Systems held in Albany, New York on July 3-7, 1979. This book deals with the oxygen and peroxide activating enzymes field. The book addresses electron transfer related to oxygen biochemistry by comparing quantum, semiclassical, and classical methods of electron transfer reactions. Several papers then discuss the active and toxic states of oxygen and superoxide as the discovery of superoxide dismutase activity of erythrocuprein can provide a means to studying oxygen reaction in biological systems. One paper then compares the active sites of molluskan and arthropodan hemocyanins, which are known as reversible oxygen-carriers. The result of this study is presented in a table. Other papers discuss the flavin catalyzed reactions of molecular oxygen and the implications of the physiological function of D-amino acid oxidase from inhibition studies. The book then explains the role of carbon monoxide in the reaction mechanism of oxygen with cytochrome oxidase. This collection will prove beneficial for research students and professors in the field of biochemistry and chemical physics.

Book Polyphenol Oxidases PPOs in Plants

Download or read book Polyphenol Oxidases PPOs in Plants written by Aravind Jukanti and published by Springer. This book was released on 2017-08-18 with total page 137 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book is first of its kind exclusively dedicated to plant polyphenol oxidases (PPOs), highlighting their importance in the food processing industry. By reviewing the scientific developments of the past several decades, it offers a comprehensive overview of various aspects of plant PPOs, including chemistry, structure, functions, regulation, genetics/genomics and molecular aspects. PPOs are copper-containing proteins found in several plant species that catalyze the hydroxylation of o-monophenols to o-diphenols and oxidation of the o-dihydroxyphenols to o-quinones. Further, the quonines undergo self-polymerization or react with amines/thiol groups to produce brown/dark coloration of products. All the PPOs contain two Cu-binding sites (CuA and CuB) as their central domain, these interact with phenolic substrates and molecular oxygen. Several of the plant PPOs contain an N-terminal transit peptide (~80-100 amino acids ) necessary for plastid import. The PPOs occur in latent form that are activated by various treatments including acid and base shock, exposure to detergents or proteolytic degradation. The pH optimum of PPOs varies widely depending upon different plant species but is usually ~4.0 – 8.0. Similarly, the optimum temperature also varies as per the source and substrate involved ranging from 30 to 45 °C. Multiple PPO isoforms have been reported in several plant species, and the chromosomal location of PPOs has also been studied in some species. The physiological role (s) of PPOs is not entirely understood, but they could be involved in defense-related functions in plants. From an applied perspective, PPOs are implicated in enzymatic browning/darkening of cereal products, vegetables and fruits. Interestingly, browning is preferred in some instances like the processing of black tea, cocoa, and coffee as it enhances their quality by forming flavorful products. There have b een initiatives to specifically breed and develop cultivars with reasonably low PPO levels in the mature grain or fruit. Further, several types of inhibitors that reduce the PPO activity have also been identified. Despite their commercial/economic importance and the availability of literature on different aspects of PPOs in different plant species, this is the first book to provide basic information regarding PPOs. It is a valuable resource for researchers involved in quality-related research specifically in crops, vegetables and fruits. Further, as PPOs are also implicated in defense- or stress-related functions, the book is also useful to breeders, pathologists, molecular biologists, physiologists and entomologists.

Book Structure and Functions of Amine Oxidases

Download or read book Structure and Functions of Amine Oxidases written by Mondovi and published by CRC Press. This book was released on 2018-01-10 with total page 299 pages. Available in PDF, EPUB and Kindle. Book excerpt: A good portion of this book has been devoted to the copper-dependent enzymes, these being the more numerous. The chapter dealing with serum amine oxidases also focuses attention on their catalytic mechanism, as these enzymes have been studied in greater depth. As the presentation of topics whose experimental basis is rapidly developing is likely to stimulate the reader‘s interest, many bibliographic references have been included.Readers could find this book poor, as far as many topics are dealt with in a relatively little space, but we believe it essential to trace the background of our present knowledge in the field of amine oxidases, stressing the future outlook of research on these enzymes, for they are becoming more and more important in general and medical biochemistry.

Book excerpt: Excerpt from The Oxidases: And Other Oxygen-Catalysts Concerned in Biological Oxidations During the past eight years I have devoted a considerable part of my time to a study of the oxidases and of certain phases of biological oxidation, with the result that I have come to appreciate as never before the need of a somewhat more comprehensive and complete treatment of this subject than is ordinarily to be found in most treatises on fermentation. Furthermore, a study of the earlier literature of the subject has revealed certain rather curious mistakes, which probably found their way into the literature originally as the result of typographical errors, but which pervade the whole subject with an annoying persistency. During the past four years, as the result of easy access to the departmental libraries in this city, I have had unusual facilities for familiarizing myself with the literature of this subject, and it was partly with the view of giving to other workers in this field the benefits of my opportunities along this line, that this monograph has been written. No special claims are made for originality either in the mode of treatment of the subject, or in some cases even with regard to phraseology, my only object being to present the subject as accurately and comprehensively as possible within the time at my disposal.
