Book excerpt: Rodent behavioral testing has been used to study brain functions since the 1890s and has become a gold-standard model in modern neuroscience. Up to the 1950s, most behavioral tests on laboratory rodent models were based on punishments and rewards. Both approaches can lead to a certain degree of animal pain or suffering. Punishments involved the employment of painful stimuli, typically electric shocks. Passive avoidance and fear conditioning tests, among the most widely used behavioral paradigms used to evaluate learning and memory in rodents, can be performed using only a single brief shock. Other tests, such as the active avoidance, might require up to tens or hundreds of shocks, strongly challenging the psychological welfare of the model animals. On the other hand, tests based on rewards, which apparently may seem more ethical, actually still induce suffering in the animals, as food rewards are almost always associated with a food restriction protocol, in order to motivate food-seeking behavior. Rodents are starved for days before starting the test and kept under food restriction for the whole duration of the test. The distress during the testing session is only a minimal part compared to the stress lived outside of the testing session, which is prolonged and continuous. Analogously, liquid rewards commonly rely on a previous water restriction protocol to use thirst as motivation. Animal stress is not only an ethical issue per se, but also an important factor potentially impacting on the reliability and reproducibility of experimental results.

The analysis of known major animal allergens has shown that most belong to single protein families: lipocalins and serum albumins for inhalant allergens, EF-hand proteins, tropomyosins and caseins for the digestive allergens. The finding that allergens are often clustered in large families may be related to the fact that common structural, biochemical or functional features contribute to their allergenicity, in addition to external adjuvant factors. Currently, there is no curative treatment for animal allergy available. In order to lower allergic reactions to respiratory allergens in daily life and to food allergens upon accidental exposure, it is important to desensitize concerned patients. Tolerance induction by allergen-specific immunotherapy is in the current focus of an ambitious research. Book excerpt: Image-guided radiotherapy (IGRT) is a new radiotherapy technology that combines the rapid dose fall off associated with intensity-modulated radiotherapy (IMRT) and daily tumor imaging allowing for high precision tumor dose delivery and effective sparing of surrounding normal organs. The new radiation technology requires close collaboration between radiologists, nuclear medicine specialists, and radiation oncologists to avoid marginal miss. Modern diagnostic imaging such as positron emission tomography (PET) scans, positron emission tomography with Computed Tomograpgy (PET-CT), and magnetic resonance imaging (MRI) allows the radiation oncologist to target the positive tumor with high accuracy. As the tumor is well visualized during radiation treatment, the margins required to avoid geographic miss can be safely reduced , thus sparing the normal organs from excessive radiation. When the tumor is located close to critical radiosensitive structures such as the spinal cord, IGRT can deliver a high dose of radiation to the tumor and simultaneously decreasing treatment toxicity, thus potentially improving cure rates and patient quality of life. During radiotherapy, tumor shrinkage and changes of normal tissues/volumes can be detected daily with IGRT. The volume changes in the target volumes and organs at risk often lead to increased radiation dose to the normal tissues and if left uncorrected may result in late complications. Adaptive radiotherapy with re-planning during the course of radiotherapy is therefore another advantage of IGRT over the conventional radiotherapy techniques. This new technology of radiotherapy delivery provides the radiation oncologist an effective tool to improve patient quality of life. In the future, radiation dose-escalation to the residual tumor may potentially improve survival rates. Because the treatment complexity, a great deal of work is required from the dosimetry staff and physicists to ensure quality of care. Preliminary clinical results with IGRT are encouraging but more prospective studies should be performed in the future to assess the effectiveness of IGRT in improving patient quality of life and local control. In this Frontiers Research Topic, we encourage submission of original papers and reviews dealing with imaging for radiotherapy planning, the physics and dosimetry associated with IGRT, as well as the clinical outcomes for cancer treatment with IGRT for all tumor sites.

Thus, there is substantial evidence showing that chronic stress can increase the likelihood of major depressive disorder and anxiety disorders, as well as cardiovascular diseases, irritable bowel syndrome and pain syndromes, to name but a few, in vulnerable individuals. Although a number of pharmacological agents are available to treat such stress-related disorders, many patients do not respond to them, and those who do often report a number of side effects. Therefore, a major emphasis in modern basic research is to uncover the underlying aetiology of these disorders, and to develop novel efficacious treatment strategies. This has led to a resurgence in developing, and using, appropriate animal models to study a wide variety of stress-related disorders. Thus, the aim of this research topic “Using stress-based animal models to understand the mechanisms underlying psychiatric and somatic disorders” was to bring together novel research articles and comprehensive review articles from prominent stress researchers. In addition to describing the insights such models have provided relating to the aetiology of psychiatric and somatic disorders, these articles also encompass mechanisms that are believed to underlie stress resilience and stress-protection. Finally, given the current prominence on the role of the brain-gut axis in health and disease, the research topic covers the emerging evidence showing how the gut, particularly the microbiota, influences affective behaviour and physiology.

Book excerpt: The history of livestock started with the domestication of their wild ancestors: a restricted number of species allowed to be tamed and entered a symbiotic relationship with humans. In exchange for food, shelter and protection, they provided us with meat, eggs, hides, wool and draught power, thus contributing considerably to our economic and cultural development. Depending on the species, domestication took place in different areas and periods. After domestication, livestock spread over all inhabited regions of the earth, accompanying human migrations and becoming also trade objects. This required an adaptation to different climates and varying styles of husbandry and resulted in an enormous phenotypic diversity. Approximately 200 years ago, the situation started to change with the rise of the concept of breed. Animals were selected for the same visible characteristics, and crossing with different phenotypes was reduced. This resulted in the formation of different breeds, mostly genetically isolated from other populations. A few decades ago, selection pressure was increased again with intensive production focusing on a limited range of types and a subsequent loss of genetic diversity. For short-term economic reasons, farmers have abandoned traditional breeds. As a consequence, during the 20th century, at least 28% of farm animal breeds became extinct, rare or endangered. The situation is alarming in developing countries, where native breeds adapted to local environments and diseases are being replaced by industrial breeds. In the most marginal areas, farm animals are considered to be essential for viable land use and, in the developing world, a major pathway out of poverty. Historic documentation from the period before the breed formation is scarce. Thus, reconstruction of the history of livestock populations depends on archaeological, archeo-zoological and DNA analysis of extant populations. Scientific research into genetic diversity takes advantage of the rapid advances in molecular genetics. Studies of mitochondrial DNA, microsatellite DNA profiling and Y-chromosomes have revealed details on the process of domestication, on the diversity retained by breeds and on relationships between breeds. However, we only see a small part of the genetic information and the advent of new technologies is most timely in order to answer many essential questions. High-throughput single-nucleotide polymorphism genotyping is about to be available for all major farm animal species. The recent development of sequencing techniques calls for new methods of data management and analysis and for new ideas for the extraction of information. To make sense of this information in practical conditions, integration of geo-environmental and socio-economic data are key elements. The study and management of farm animal genomic resources (FAnGR) is indeed a major multidisciplinary issue. The goal of the present Research Topic was to collect contributions of high scientific quality relevant to biodiversity management, and applying new methods to either new genomic and bioinformatics approaches for characterization of FAnGR, to the development of FAnGR conservation methods applied ex-situ and in-situ, to socio-economic aspects of FAnGR conservation, to transfer of lessons between wildlife and livestock biodiversity conservation, and to the contribution of FAnGR to a transition in agriculture (FAnGR and agro-ecology).

The problem of antimicrobial resistance (AMR), caused by excessive and inappropriate use of antibiotics, is a public health issue that concerns us all. The introduction of penicillin in the 1940s, the start of the antibiotics era, has been recognized as one of the greatest advances in therapeutic medicine. However, according to the World Health Organization (WHO), AMR infections are now an increasing worldwide public health threat and a post-antibiotic era is imminent, where common infections and minor injuries could be fatal. AMR is a typical ‘One Health’ problem, in which livestock animals and the environment constitute AMR reservoirs and transmission routes to and from the human population. Without effective antimicrobials to counter and prevent infections, other major achievements in modern medicine, such as organ transplantation, cancer chemotherapy and major surgery, risk being compromised. AMR infections in animals have negative outcomes on animal health, welfare, biosecurity and production. In 2006, the ban of growth promoting antibiotics highlighted antibiotic use in animal production as a risk factor in the development of antibiotic resistant bacteria. Bacteria can be transferred to humans via several routes; consumption of animal products, exposure through contact with animals, and the contamination of ground and surface waters by animal waste products. Therefore, it is of utmost importance that antimicrobial use in animals is reduced to a minimum, without compromising animal health and welfare. Mechanisms of bacterial antibiotic resistance are classified according to the types of antibiotic molecules or their targets in the cell. Environmental antibiotic-resistance genes are spread then acquired by clinically relevant microorganisms. Many resistance genes are conveyed into pathogen genomes via mobile genetic elements such as plasmids, transposons or integrons, increasing the propagation of potential resistant pathogens. Substantial progress has already been made in elucidating the basic regulatory networks that endow bacteria with their extraordinary capacity to adapt to a diversity of lifestyles and external stress factors. So how will we face bacteria in the future?
