Book excerpt: Betalain-rich extracts (BREs) are a pigment found in food sources such as beetroot and prickly pear. They have been touted for their potent anti-oxidant and anti-inflammatory properties and these properties have been studied in both healthy and diseased models. Using a randomized, double-blind, crossover design, we tested the hypothesis that BRE supplementation would decrease markers of muscle damage, muscle soreness and fatigue and thus, improve running time trial performance compared to control in competitive, male runners (n=13). Subjects were supplemented with 50 mg of BRE or 50mg of oat [beta]-glucans (control) twice per day for 7 days prior to each trial. On the day of experimentation, cardiopulmonary and metabolic measurements and markers of muscle damage were obtained at rest, during a 30 min submaximal exercise trial, and during a 5 km TT with ~3 to 5 minutes of rest between trials. Book excerpt: Background: Betalains are phytochemical pigments of red beets that have antioxidant and anti-inflammatory properties. The purpose of this study was to determine the effects of betalain-rich concentrates (BRC) containing no sugars and nitrates on exercise performance and recovery in competitive triathletes. Methods: Twenty-two (9 men and 13 women) triathletes (38 ± 11 yrs) completed two double blind, cross-over, randomized trials (BRC and placebo) separated by 7 days. Each trial was preceded by 6 days of supplementation with 50 mg of BRC or placebo, twice per day for a total of 100mg per day. On the 7th day of supplementation, exercise trials commenced 120 min after supplementation with 50mg BRC or placebo and consisted of 40 min of cycling (75±5 %VO2max) followed by a 10 km running time trial (TT). Subjects returned 24 hours later to complete a pre-exercise blood draw and a 5km running TT to assess recovery. Heart rate (HR), rate of perceived exertion (RPE), oxygen consumption (VO2), respiratory exchange ratio (RER), blood lactate, hematocrit, serum glucose and markers of muscle damage (creatine kinase [CK] and lactate dehydrogenase [LDH]) and subjective evaluations of whole body muscle soreness and fatigue (via visual analogue scales) and a brief fatigue inventory (BFI) were assessed. Results: There were no differences between treatments during the sub-maximal cycling bout at the same exercise intensity. 10km running TT duration (49.5±8.9 versus 50.8±10.3 min, p=0.03) was faster in 15 of the 22 subjects with the BRC treatment compared to placebo. Despite running faster, average HR (161.7±11.4 versus 161.9±12.4 bpm) and RPE (5.8±1.2 versus 5.7±1.1 out of 10) were not different between BRC and placebo. 5-km running TT duration 24 hours after the 10-km TT (23.2±4.4 versus 23.9±4.7 min, p=0.003) was faster in 17 of the 22 subjects with the BRC treatment compared to placebo. Average HR during the 5-km TT (166.3±12.6 versus 162.8±14.5 bpm, p=0.004) was higher with BRC treatment, but RPE (6.5±1.4 versus 6.4±1.3 out of 10) was not different between BRC and placebo. CK, a muscle damage marker, increased less (40.5±22.5 versus 49.7±21.5 UL−1, p=0.02) from pre-exercise to after the 10-km TT and fatigue increased less from the BFI (-0.05±6.1 versus 3.23±6.1, p=0.05) 24 hours after the 10-km TT with the BRC treatment. There were no differences due to treatment for VO2, or RER. Conclusion: Betalain-rich concentrate supplementation improved 10-km TT performance in competitive male and female triathletes. Improved 5-km TT performances 24h after the 10-km TT and the attenuated increase of CK and fatigue, suggest an increase in recovery, and the ability to work harder with the same effort while taking BRC.

During the past two decades, there have been major new developments in methods which can be applied toward the isolation, separation and structure determination of complex natural products. Therefore, the topic of this symposium, "Modem Phytochemical Methods", is a very timely one. The organizers of the symposium recognized that it would not be possible to cover in detail all new advances in phytochemical methodology. It was therefore decided to emphasize general reviews on recent developments of major separation techniques such as high performance liquid chromatography as well as supercritical fluid chromato graphy. In addition, advances in commonly used structure determination methods, mainly NMR and MS, are reviewed. Other topics include methodo logies of micro-sampling for isolation and analysis of trichome constituents as well as recent breakthroughs on biosynthetic studies of monoterpenes using "enriched" basal cells of trichomes. The volume concludes with a review of quantitative structure-activity relationship (QSAR) studies of biologically active natural products. In Chapter I, K. Hostettmann and his colleagues give a general review of recent developments in the separation of natural products with major emphasis on preparative separations of biologically active plant constituents. The ultimate proof of health benefits will be deciphered by the isolation and identification of specific phytochemicals acting on specific receptors in the body exerting their biological effects. The important point being emphasized is that if the physiological effect of an ingredient is glaring, it is considered to elicit a significant response. However, when the effect of an ingredient is significant, yet the effect is not readily perceivable then the usefulness goes unnoticed and can be questioned as a myth. For example, coffee has several ingredients, but only caffeine acting on the adenosine receptors stimulates the central nervous system and promotes wakefulness. Similarly, the specific active ingredient in hot chili pepper, capsaicin, activates a member of a recently identified family of receptors called transient receptor potential (TRP) channels. TRP vanilloid 1 (TRPV1), which is involved in certain modalities of pain, promotes the release of hormones in the gastrointestinal (GI) tract. Although, the bioavailability of the active ingredients is low, it is important to realize that these ingredients can cause significant local effects in the GI tract by preventing cholesterol regeneration by subduing microbiota, inhibiting precancerous growth by promoting calcium influx and stimulating nerve endings to transmit information and to release of gut hormones that are involved in regulating a variety of functions including appetite and satiety.
