Book excerpt: In biological literature, several definitions of quantitative autoradio graphy are given. The term is defined as either the determination and com parison of the density of silver grains above various structures or under varying conditions, or the determination of absolute quantities of radio activity. In both these cases, photometric measurement serves for more rapid and more exact evaluation of grain densities than would be possible by visual counting of the grains. The equipment generally used for the photometric measurement of silver grains consists of a microscope, a photocell, an electronic amplifier system and a display unit. Grains can be made accessible to photometric evaluation by various kinds of microscopic illumination: 1. Substage bright-field illumination. 2. Substage dark-field illumination. 3. Incident dark-field illumination. 4. Vertical bright-field illumination. With all these types of illumination, the relationship between the luminous flux I absorbed by the film, scattered into the objective and reflected or diffracted, and the flux 10 which is not affected by the film is used as a measure of grain density. Since these are differential measurements, the light beam I transmitted by the film is in itself a measure of grain density if the luminous flux 10 incident on the grains is kept constant. This approach has been used in a large number of measuring setups.

Book excerpt: Because of the possible errors in interpretation as a result of the occurrence of pseudoimages formed by improper preparation of plant material for autoradiography, three drying methods were compared: freeze-drying, oven-drying, and air-drying. Freeze-drying was the best method for preparing plants for autoradiography, oven-drying the least effective method. Pseudoimages were consistently produced by oven-drying, especially in bean plants; these false images were particularly characterized by peripheral movement of the label in the treated leaves. Sectioning did not prevent this movement. Air-drying was ranked second as an effective technique. Pseudo-images produced by air-dried plants appeared to be relatively minor compared with those of oven-dried plants. The apparent presence of the label in the roots appeared to be the principal anomaly in air-dried bean plants. Freeze-drying was not as essential for the woody plants as for the bean plants. Air-dried ash produced excellent autoradiographs similar to those of freeze-dried ash. However, metabolic degradation of the labeled compound may occur during air-drying, resulting in probable loss of the one-for-one relationship between the compound under study and the radioactive tracer. It also provides additional data collected in the subsequent years to show how the radioactivity levels in agricultural products and their growing environments have changed with time and the route by which radioactive materials entered agricultural products as well as their movement between different components (e.g., soil, water, and trees) within an environmental system (e.g., forests). The book covers various topics, including radioactivity testing of food products; decontamination trials for rice and livestock production; the state of contamination in, trees, mushrooms, and timber; the dynamics of radioactivity distribution in paddy fields and upland forests; damage incurred by the forestry and fishery industries; and the change in consumers’ attitudes. Chapter 19 introduces a real-time radioisotope imaging system, a pioneering technique to visualize the movement of cesium in soil and in plants. This is the only book to provide systematic data on the actual change of radioactivity, and as such is of great value to all researchers who wish to understand the effect of radioactive fallout on agriculture. In addition, it helps the general public to better understand the issues of radio-contamination in the environment. The project is ongoing; the research groups from the Graduate School of Agricultural and Life Sciences of The University of Tokyo continue their work in the field to further evaluate the long-term effects of the Fukushima accident.

By means of slow and empirical selections, it has been possible to greatly increase both quantity and quality of plant crops. This procedure has brought the most useful cereals to a state of refinement that seems to be difficult to further improve by the same methodology. Indeed, natural sexual mechanisms were always used to cross closely related sexually and genetically compatible organisms; the selection procedure consisted of isolating the most promi sing progenies. Obviously, by this way, plants could only share preexisting genetic pools. On the other hand, the last decade has seen drastic modifi cations of the experimental plant sciences, with the appearance of new technological possibilities. Because of this profound reshaping of our experimental ap proaches, other means can now be realistically envisaged in order to achieve similar or even higher goals. It is, for instance, possible to attempt completing sexual crosses (where both male and female gametes bring together the genetic informations necessary for growth and development of the offspring) by parasexual means by which novel genetic informa tions could possibly be added to the heritage •. At the limit, such genetic manipulations could enable man to create plants capable of producing new substances characteris tic of unrelated plants or, more generally, of other living or ganisms. Even if these possibilities might appear quite remote, the interest of Scientists has been awaken and, indeed, several at tempts to such genetic manipulations have already been made.
