Book excerpt: The vertebrate immune system defends the organism against invading pathogens while at the same time being self-tolerant to the body’s own constituents thus preserving its integrity. Multiple mechanisms work in concert to ensure self-tolerance. Apart from purging the T cell repertoire from auto-reactive T cells via negative selection in the thymus dominant tolerance exerted by regulatory T cells plays a major role in tolerance imposition and maintenance. Among the various regulatory/suppressive cells hitherto described, CD4+CD25+ regulatory T cells (Treg) and interleukin-10 producing T regulatory 1 (Tr1) cells have been studied in most detail and are the subject of most articles in this issue. Treg, also called "natural" regulatory T cells, will be traced from their intra-thymic origin to the site of their action in peripheral lymphoid organs and tissues. The repertoire of Treg is clearly biased towards recognition of self-antigens, thereby potentially preventing autoimmune diseases such as gastritis and oophoritis. Regulatory T cells, however also control infections, allergies and tolerance to transplanted tissues and this requires their induction in the periphery under conditions which are not yet fully understood. The concept of dominant tolerance, by far not novel, will offer new insights and hopefully tools for the successful treatment of autoimmune diseases, improved cancer immunotherapy and transplant survival. The fulfillment of these high expectations will, however, require their unambiguous identification and a better understanding of their mode of action.

During this time, the abundance of fascinating literature on the immunological and functional aspects of this organ has been in contrast to the paucity of reports on its role in pathology. The causal or formal pathogenesis even of many of the well-documented pathological fea tures is so far unexplained or at least uncertain. In spite of some concep tual progress related to the work of CASTLEMAN, LEVINE, and ROSAI, we regrettably have to say that in pathology the thymus remains almost as inconspicuous as 25 years ago. Only the new techniques developed in recent years have made it possible to look as closely into the complicated structural organization of the thymus as it appears to be necessary to observe and document abnormalities. Major steps have been taken with the advent of mono clonal antibody immune histochemistry and with detailed ultrastructur al studies in embryology. At this point, pathologists and researchers with a special interest in the thymus were asked to give overviews of their respective fields of interest in light of recent findings in immunology and basic insights into the structural-functional interrelationship of the human thymus. The results of this initiative have been brought together in this volume.

Such assays are particularly important in situations where a large number of foods/bioactives need to be screened rapidly and in a cost effective manner in order to ultimately identify lead foods/bioactives that can be the subject of in vivo assays. Book excerpt: Emigration of mature naïve CD4 SP T cells from the human thymus to the periphery is a field that remains not fully understood, although elucidation of the mechanisms that govern egress of T cells is crucial to understanding both basic immunology and the immune response in disease states such as HIV infection. In this dissertation work, I have examined the expression and function of a novel requisite T cell egress receptor expressed within the human thymus, as well as characterized changes observed in the expression and function of this receptor during HIV infection, and finally examined additional markers of maturation stages of human thymocytes. In Chapter 2, ("S1P/ S1P-R1 signaling is required for migration of naïve human T cells from the thymus to the periphery") I investigated whether Sphingosine-1-phosphate receptors (S1P-Rs) are expressed on human thymocyte populations and whether they function in the egress of mature human thymocytes from the thymus to the periphery. In Chapter 3 ("HIV-1 infection results in upregulation of sphingosine-1-phosphate receptor 1 on mature human thymocytes and functional naïve T cell response to S1P"), I examined whether Human Immunodeficiency Virus 1 (HIV-1) infection affects the expression and function of S1P-R1 utilizing humanized (NSG thy/liv) mice as well as various ex vivo assays to investigate the function of S1P-R downstream signaling. Finally, in Chapter 4, ("Alternative markers for maturation stages of human thymocytes: Expression of CD31 (PECAM-1) during T cell differentiation in the human thymus"), work I performed in collaboration with others within the AIDS Institute at UCLA, I contributed to the characterization of additional markers of developing thymocytes within the human thymus. Interestingly, a novel marker characterized may play a role in modulation of TCR signaling during T cell development in the thymus and may moreover be coexpressed with S1P-R1 on mature human thymocytes. Taken together, this work should advance the fields of basic T cell Immunology as well as HIV Immunology and open new avenues for exploration into therapeutics for HIV infected individuals.
