Book excerpt: Single-molecule fluorescence spectroscopy is a compelling technique for studying the dynamics of catalytic reactions because heterogeneity in catalyst populations can be directly observed. Understanding the nature of diversity in catalyst behavior will lead to the design of better, more efficient catalysts. Synthetic challenges emerge when developing chemical systems for single-molecule spectroscopy techniques, because the molecules need to be fluorescently labeled and anchored to a surface for long-term imaging. A reliable attachment to the surface is critical for successful single-molecule spectroscopy investigations. Robust surface linkages play an important role in the stability and recyclability of surface-supported molecular catalysts as well. This thesis focuses on the design and synthesis of suitable fluorescent chemical systems for single-molecule fluorescence spectroscopy investigations of catalyst transformations and highlights the challenges related to developing and implementing surface attachment strategies for surface-supported molecular catalysts. A fluorescent Pd-PEPPSI precatalyst complex is designed and synthesized for single-molecule fluorescence spectroscopy experiments, and the mechanism of catalyst initiation is examined using both bulk NMR and single-molecule fluorescence spectroscopy. Various surface-attachment strategies are investigated for the immobilization of molecular Pd-PEPPSI complexes onto both polymers and metal oxides. Multiple characterization techniques are employed to analyze the nature of the surface linkage chemistries and gain insight into the stability and degradation mechanisms of surface-supported molecular palladium catalysts.

While the use of catalyst is typically recognized in the automobile industry, their impact is more widespread as; catalysts are used in the synthesis of 80% of the US commercial chemicals. Despite the improved selectivity provided by catalyst, process inefficiencies still threaten the sustainability of a number of synthesis methods, especially in the pharmaceutical industry. Recyclable solid supported catalysts offer a unique opportunity to address these inefficiencies. Such systems coupled with continuous synthesis techniques, have the potential to significantly reduce the waste to desired product ratio (E-factor) of the production techniques. This research focuses developing sustainable processes to synthesize organic molecules by using continuous synthesis methods. In doing so, solid supported metal catalyst systems were identified, developed, and implemented to assist in the formation of carbon-carbon bonds. Newly developed systems, which utilized metal nanoparticles, showed reactivity and recyclability, comparable to commercially available catalyst. Nanoparticles are emerging as useful materials in a wide variety of applications including catalysis. These applications include pharmaceutical processes by which complex and useful organic molecules can be prepared. As such, an effective and scalable synthesis method is required for the preparation of nanoparticle catalysts with significant control of the particle size, uniform dispersion, and even distribution of nanoparticles when deposited on the surface of a solid support. This project describes the production of palladium nanoparticles on a variety of solid supports and the evaluation of these nanoparticles for cross coupling reactions. This report highlights novel synthesis techniques used in the formation of palladium nanoparticles using traditional batch reactions. The procedures developed for the batch formation of palladium nanoparticles on different solid supports, such as graphene and carbon nanotubes, are initially described. The major drawbacks of these methods are discussed, including limited scalability, variation of nanoparticle characteristics from batch to batch, and technical challenges associated with efficient heating of samples. Furthermore, the necessary conditions and critical parameters to convert the batch synthesis of solid supported palladium nanoparticles to a continuous flow process are presented. This strategy not only alleviates the challenges associated with the robust preparation of the material and the limitations of scalability, but also showcases a new continuous reactor capable of efficient and direct heating of the reaction mixture under microwave irradiation. This strategy was further used in the synthesis of zinc oxide nanoparticles. Particles synthesized using this strategy as well as traditional synthesis methods, were evaluated in the context industrially relevant applications.

It is our hope that the following chapters and the concluding summary of recommendations for research may help to provide a definition of surface organometallic chemistry. Besides catalysis. the central theme of the Workshop, four main topics are considered: 1) Reactions of organometallics with surfaces of metal oxides, metals. and zeolites; 2) Molecular models of surfaces, metal oxides, and metals; 3) Molecular approaches to the mechanisms of surface reactions; 4) Synthesis and modification of zeolites and related microporous solids. Most surface organometallic chemistry has been carried out on amorphous high-surf ace-area metal oxides such as silica. alumina. magnesia, and titania. The first chapter. contributed by KNOZINGER. gives a short summary of the structure and reactivity of metal oxide surfaces. Most of our understanding of these surfaces is based on acid base and redox chemistry; this chemistry has developed from X-ray and spectroscopic data, and much has been inferred from the structures and reactivities of adsorbed organic probe molecules. There are major opportunities for extending this understanding by use of well-defined (single crystal) oxide surfaces and organometallic probe molecules.

Book excerpt: The development of methodologies for C-N bond formation reactions is an important scientific challenge because of many academic and industrial applications. This work will focus particularly on palladium-catalyzed cross-couplings of amine-containing compounds with aryl halides. The scope of the BrettPhos precatalyst for the cross-coupling of ortho-substituted aryl iodides with amides is studied using substrates with a variety of functional groups. Due to potential metal-chelating issues with some of the substrates used in this study, a proposed ligand synthesis is discussed in which one of the methoxy groups of BrettPhos is replaced with a morpholine capable of occupying palladium's open coordination site during its catalytic cycle. A final C-N bond formation study focuses on the cross-coupling of aryl halides with amidine salts. For this cross-coupling, a methodology has been developed that can be applied to various electron-rich, electron-poor, and electron-neutral substrates. Furthermore, the products of this cross-coupling can be used for a subsequent electrocyclization through a reaction with aldehyde, demonstrating that a relatively simple two-pot methodology can be used to make relatively complex substrates with pharmaceutical applications. Both amides and amidines are common moieties in drug-like molecules because of the various biological activities of these functional groups. Potential medicinal applications of the developed cross-coupling of amidine salts with aryl halides methodology are described. Thus, methodologies for various palladium-catalyzed, C-N cross-couplings as well as a potential ligand synthesis to be used for palladium catalysis are herein discussed.

Transition metal catalysis is at the forefront of synthetic organic chemistry since it offers new and powerful methods to forge carbon–carbon bonds in high atom- and step-economy. In Chapter 1, the author describes a Pd- and Ni-catalyzed cycloisomerization of aryl iodides to alkyl iodides, known as carboiodination. In the context of the Pd-catalyzed variant, the chapter explores the production of enantioenriched carboxamides through diastereoselective Pd-catalyzed carboiodination. It then discusses Ni-catalyzed reactions to generate oxindoles and an enantioselective variant employing a dual ligand system. Chapter 2 introduces readers to a Pd-catalyzed diastereoselective anion-capture cascade. It also examines diastereoselective Pd-catalyzed aryl cyanation to synthesize alkyl nitriles, a method that generates high yields of borylated chromans as a single diastereomer, and highlights its synthetic utility. Lastly, Chapter 3 presents a Pd-catalyzed domino process harnessing carbopalladation, C–H activation and π-system insertion (benzynes and alkynes) to generate spirocycles. It also describes the mechanistic studies performed on these reactions.

Current research interests involve the development of new catalyst systems for various palladium catalyzed reactions. Previous work has suggested the active species for palladium-catalyzed cross-coupling reactions to be a mono-ligated palladium(0) species when sterically demanding ligands are used. Therefore, the my work focused on the synthesis of precatalysts with an established 1:1 L:Pd. Specifically, we were focused on the synthesis of mono-ligated palladium(II) precatalysts of general formula [(R_3P)PdCl_2]_2, using di-tert-butylneopentylphosphine (DTBNpP), tert-butyldineopentylphosphine (TBDNpP), and trineopentylphosphine (TNpP) as ligands. Under optimized conditions, both [(DTBNpP)PdCl_2]_2 and [(TNpP)PdCl_2]_2 were effective precatalysts for the Suzuki cross-coupling of a wide range of aryl bromides. Comparison studies with the air-stable precatalysts versus the in-situ generated catalyst showed the precatalysts to have improved conversions and higher rates of reaction under both inert and ambient atmospheres. The precatalyst activation pathways were investigated by 31P NMR spectroscopy. The spectra obtained from 31P NMR experiments revealed a side reaction resulting in a catalytically inactive palladacycle species. The results obtained from the mechanistic investigations led to further optimization of reaction conditions to decrease the amounts of catalytically inactive side products formed.
