Book excerpt: Coordination compounds have been well-known for their wide variety of applications for over a century, as well as enhancing the researcher’s interest and concern in evaluating their action mechanism. It is certainly one of the most intensely discussed research topics. Coordination compounds involve different metal-ion-ligand phenomenon. The involved metal ions play a significant role in structural association and functioning of several processes in the genetic and metabolism system. In recent years, Schiff base ligands have gained significant interest and received a keen interest of many researchers. Schiff’s base ligands have been recognized to hold a wide variety of biological and medicinal activities due to the presence of donor atoms. They have proved exceptional pharmalogical actions such as antimicrobial, anti-tuberclosis, antiplatelet, antidiabetic, antiarthritis, antioxidant, anti-inflammatory, anticancer, antiviral, antimalarial, and analgesic. These biologically active Schiff base ligands have also been shown to inhibit enzyme mobilization and, when bound to a metal ion, exhibit enhanced biological activity, making them useful in a number of fields. As a result, metal complexes of Schiff base ligands are gaining popularity due to their unique properties and functionalities. Schiff base complex-based research for educational and industrial purposes is booming, and the number of publications is gradually increasing. Despite these interests, there is currently no detailed book on Schiff base metal complexes that covers the structures, biological activities, and other non-biological perspectives. This book delves into the structures of Schiff base metal complexes, which are critical in assessing the biological viability of any complex. It also highlights their biological significance in pharma and drug discovery like antibacterial, antifungal, anticancer, anti-inflammatory, anti-arthritis, anti-diabetic, antioxidants, anti-proliferative, antitumor, anticancer, antiviral. Book excerpt: Transition metal complexes of nickel(II) and zinc(II) with 5-bromo- N -(8-quinolyl)salicylaldimine (HqsalBr, HL 1 ) and 3,5-dibromo- N -(8-quinolyl)salicylaldimine (HqsalBr 2 , HL 2 ) have been synthesized and successfully characterized using several techniques: IR, 1 H-NMR, mass spectrometry and thermogravimetric analysis (TGA). The molecular structures of nickel(II) and zinc(II) complexes with HL 2 were determined by single crystal X-Ray crystallography. Ligands HL 1 and HL 2 both act as a tridentate ligand and coordinate to metal ion center via deprotonated hydroxyl oxygen atom, nitrogen of azomethine functional group and nitrogen of quinoline substituent. The interaction of these complexes with DNA was investigated using calf thymus DNA (CT-DNA) in which the studies by electronic absorption and luminescence titrations reveal that DNA binding mode of the complexes is intercalation. All the synthesized complexes provide great values of binding constant (K b ) and these results are consistent with the quenching capability defined by quenching constant values (K q ). Amongst the synthesized complexes, zinc(II) with ligand HL 2 complex, [Zn(qsalBr) 2 ], exhibits the most superior ability in the interaction with CT-DNA and the verification mode of binding by the competitive reaction with ethidium bromide (EB) also indicates the strongest intercalation by such complex. Additionally, ligands and complexes were also examined in anti-lung cancer activity against A549 cell line and compared with standard compounds, Etoposide and Cisplatin.

The reaction of bis(salicylaldehyde) complexes of nickel(II) and copper(II) with m-xylylenebis-(2-(1,3-propanediamine)) (3) result in the formation of 5 and 7 respectively, while the reaction of managanese(II) acetate with salicylaldehyde and 3 results in the formation of 6a. The nickel complex, 5, undergoes a quasireversible two-electron transfer at -1.47 V vs SCE attributable to the nickel(II)-nickel(I) couple. The binuclear nickel(I) complex can be generated from 5 both electrochemically and chemically, and has been shown by infrared analysis to form adducts of CO and MeNC. Magnetic susceptibility measurements by the Faraday method show the manganese and copper complexes (6a and 7) have solid state room temperature moments of 5.47 and 1.92 B.M. per metal center, respectively, while the solution magnetic moment of the binuclear copper complex (7) is found by the Evans method to be 1.49 B.M. per copper center. This reduced magnetic moment relative to the moment for mononuclear Cusalprn(1.79 B.M) suggests a weak magnetic interaction between the two copper centers in the binuclear copper complex 7.
