In this work, SERS applications in the realms of art conservation and biosensing are reviewed. First, the theory of this unambiguous and ultrasensitive technique is explained. Next, a review of the pedagogical journey in art conservation research through the lenses of undergraduates is offered in Chapter 2, a chapter recently submitted to the American Chemical Society. Finally, an investigation in biosensing SERS is explored with developing a SERS pH sensitive probe while offering new insight into SERS capabilities and the relationship between analyte and nanoparticle.

Book excerpt: In art conservation, color is essential to understanding a society's culture and history-as an indicator of beauty, status, religion, and more-but has a tendency to fade and diminish over time. Analytical techniques, particularly that of pigment identification, can reveal the artifact's original color and appearance and give new insights to an artist's intentions, techniques, date of creation, and more. However, most identification procedures are invasive and destroy the samples in the process. Surface-enhanced Raman spectroscopy (SERS) has recently been identified as a technique that is minimally invasive and also solves the issue of fluorescence that is found in many other techniques. In this paper, a specific SERS procedure has been developed for the identification of yellow organic dyes from 18th century Japanese Woodblock prints. Several SERS spectra of nine dyes both in solution and applied on artist paper have also been documented in hopes of assisting with pigment identification in the future.

These themes are naturally wider than what at first might be considered as artworks and archaeological artefacts and the topics include pigments, paintings, ceramics, glass, sculpture and patination / corrosion, textiles, industrial archaeology, the degradation and preservation of biomaterials, mummies and human skeletal remains. An interesting feature is the inclusion of modern case studies which describe specific problems and approaches to the Raman spectral analysis of items important to our cultural heritage. The text is prefaced with an introduction to the important parameters used in nondestructive Raman measurements and also highlights some future applications based upon novel miniaturised instrumentation for in-field studies and potential screening work which will identify specimens which would repay further studies in the laboratory. An important aside from this work is the means of spectral identification of ongoing biodeterioration and biological colonisation in specimens in storage and the effects of environmental deterioration such as humidity and temperature upon their integrity.

Book excerpt: Herein analysis of dye molecules has been carried out by means of surface enhanced Raman spectroscopy (SERS) and/or surface enhanced resonance Raman spectroscopy (SERRS) using citrate- and/or borohydride-reduced silver colloids employing laser exciting wavelengths equal to 514.5 and/or 632.8 nm. SERS and/or SERRS spectra are reported using, as model system probes, eight dye molecules which belong to several distinct chemical structural classes. Experimental protocols were developed and subsequently modified, as required, for each dye molecule examined. Vibrational spectroscopic profiles were obtained, where possible, with respect to concentration and pH dependence. SERS and/or SERRS vibrational bands provided unique fingerprint spectra for each dye molecule. In an attempt to develop novel applications of SERRS the technique has been used, in a kinetic investigation, to monitor and analyse the synthesis of the dye indigo carmine from indigo using a silver sol as the SERRS substrate/medium. In another study it was possible to differentiate between two structurally similar anthraquinones, alizarin and purpurin, using SERRS. It was also possible to demonstrate the existence of multiple molecular species of certain dye molecules, as a function of pH e.g. nuclear fast red, metanil yellow, purpurin and alizarin. For some dye molecules e.g. alcian blue it was possible to combine the linear regions of normal (non-resonance/non-enhanced) Raman and SERS/SERRS plots, thereby extending the dynamic range available for semi-quantitative analysis. The sensitivity of the SERS/SERRS technique for semi-quantitative trace analysis of eight dye molecules has been successfully demonstrated.

The first application aimed to identify red colorants on Japanese woodblock prints by novel sampling and analytical methods. The inception of this project was in partnership with the Portland Art Museum (PAM) and the 2018 exhibition entitled Suzuki Harunobu and the Culture of Color. The work herein developed a soft mechanical sampling technique for the acquisition of micrometer-sized colorant particles from the surface of 18th century woodblock prints. In Chapters 3 and 4, the sampling method is introduced and validated, and the collected samples were analyzed by surface-enhanced Raman spectroscopy (SERS). The analysis was aided by the implementation of a genetic algorithm (GA) which matches the spectral contributions of the samples to identify the components present. In Chapter 5, the results of a study of artworks attributed to Suzuki Harunobu in the PAM's permanent collection are presented. Through a multi-analytical approach, the palette of red colorants in the collection was established. This work provides technical information and historical context to the practice of commercial printmaking in late-18th century Japan. The second application, presented in Chapter 7, aimed to develop a sensor for the detection of active corrosion products on the surface of metal sculpture. This work provides the preliminary synthesis and characterization of a carbon-quantum dot functionalized polymeric hydrogel (bCQD-hydrogel). The bCQD-hydrogels harness the fluorescence properties of carbon quantum dots by incorporating them into a flexible polymer network. This soft material is then demonstrated to be sensitive to soluble metal ions, the first indicators of active metal corrosion. Finally, preliminary data is presented demonstrating the use of these materials on metal surfaces. These two projects, while seemingly unrelated, represent a breadth of research focus and illustrate the expansive applications of conservation science.

In current times, Raman spectroscopy has emerged as a versatile and powerful tool in a diverse set of scientific fields. Its implementation has grown markedly in the past 20 years due to technological advances and affordability of instrumentation. As such, more and more undergraduate institutions have acquired Raman instrumentation, and faculty from a variety of disciplines have begun to utilize the technique. This has resulted in an increased number of students gaining hands-on experience with Raman spectroscopy. As its use has grown, curricular pedagogies that utilize Raman spectroscopy to investigate interesting scientific problems have continually been developed, implemented, and publicized. Given the recent developments in the field and inspired by similar symposia on nuclear magnetic resonance and x-ray crystallography at recent ACS meetings, the editors developed a symposium titled "Engaging Undergraduates with Raman Spectroscopy." This symposium was held at the National ACS meeting held in Washington, D.C., in 2017. It generated strong interest, and the quality of presentation and breadth of knowledge displayed by the presenters was indicative of the continual pedagogical innovation of Raman spectroscopy in the undergraduate curriculum. The collection of chapters herein is based on the symposium, and several contributors to this book were its invited speakers. One of the main objectives of this volume is to convey the ideas discussed at the symposium to the broader scientific community. Our hope is that readers not only learn a great deal about the uses of Raman spectroscopy but also are stimulated to innovate new ways to incorporate Raman spectroscopy into the undergraduate curriculum.
