Download or read book Modern Methods of Drug Discovery written by Alexander Hillisch and published by Springer Science & Business Media. This book was released on 2002-12-11 with total page 314 pages. Available in PDF, EPUB and Kindle. Book excerpt: Research in the pharmaceutical industry today is in many respects quite different from what it used to be only fifteen years ago. There have been dramatic changes in approaches for identifying new chemical entities with a desired biological activity. While chemical modification of existing leads was the most important approach in the 1970s and 1980s, high-throughput screening and structure-based design are now major players among a multitude of methods used in drug discov ery. Quite often, companies favor one of these relatively new approaches over the other, e.g., screening over rational design, or vice versa, but we believe that an intelligent and concerted use of several or all methods currently available to drug discovery will be more successful in the medium term. What has changed most significantly in the past few years is the time available for identifying new chemical entities. Because of the high costs of drug discovery projects, pressure for maximum success in the shortest possible time is higher than ever. In addition, the multidisciplinary character of the field is much more pronounced today than it used to be. As a consequence, researchers and project managers in the pharmaceutical industry should have a solid knowledge of the more important methods available to drug discovery, because it is the rapidly and intelligently combined use of these which will determine the success or failure of preclinical projects.

Book excerpt: Dr. G. Robert Pettit is a man who has dedicated his life to finding cures for cancer. A gifted organic chemist, his search has taken him all over the world, for it is his belief that the most promise lies in naturally occurring compounds which have evolved over billions of years as part of the profound diversity of life on this planet. His search has drawn him across deserts, into jungles, and below the surface of oceans across the planet. A prolific research scientist with over 800 peer reviewed articles and responsible for the discovery and development of numerous anticancer compounds in use today, Dr. Pettit is a man who has saved lives and made a very real difference. This is his story.

Download or read book Organic Syntheses Based on Name Reactions and Unnamed Reactions written by Alfred Hassner and published by Elsevier. This book was released on 2013-10-22 with total page 462 pages. Available in PDF, EPUB and Kindle. Book excerpt: Synthetically useful organic reactions or reagents are often referred to by the name of the discoverer(s) or developer(s). Older name reactions are described in text books, but more recently developed synthetically useful reactions that may have been associated occasionally with a name are not always well known. For neither of the above are experimental procedures or references easy to find. In this monograph approximately 500 name reactions are included, of which over 200 represent newer name reactions and modern reagents. Each of these reactions are extremely useful for the contemporary organic chemistry researcher in industry or academic institutions. This book provides the information in an easily accessible form. In addition to seminal references and reviews, one or more examples for each name reaction are provided and a complete typical experimental procedure is included, to enable the student or researcher to immediately evaluate reaction conditions. Besides an alphabetical listing of reactions and reagents, cross references permit the organic practitioner to find those name reactions or reagents that enable specific transformations, such as, conversion of amines to nitriles, stereoselective reduction, fluoroalkylation, phenol alkynylation, asymmetric syntheses, allylic alkylation, nucleoside synthesis, cyclopentanation, hydrozirconation, to name a few. Emphasis has been placed on stereoselective and regioselective transformations as well as on enantioselective processes. The listing of reactions and reagents is supported by four indexes.
Download or read book Total Synthesis of Natural Products written by Jie Jack Li and published by Springer Science & Business Media. This book was released on 2013-03-14 with total page 292 pages. Available in PDF, EPUB and Kindle. Book excerpt: 'Total Synthesis of Natural Products' is written and edited by some of today's leaders in organic chemistry. Eleven chapters cover a range of natural products, from steroids to alkaloids. Each chapter contains an introduction to the natural product in question, descriptions of its biological and pharmacological properties and outlines of total synthesis procedures already carried out. Particular emphasis is placed on novel methodologies developed by the respective authors and their research groups. This text is ideal for graduate and advanced undergraduate students, as well as organic chemists in academia and industry.
Download or read book Retrosynthetic Analysis and Synthesis of Natural Products 1 written by Olivier Piva and published by John Wiley & Sons. This book was released on 2019-12-12 with total page 314 pages. Available in PDF, EPUB and Kindle. Book excerpt: For chemists, attempting to mimic nature by synthesizing complex natural products from raw material is a challenge that is fraught with pitfalls. To tackle this unique but potentially rewarding task, researchers can rely on well-established reactions and methods of practice, or apply their own synthesis methods to verify their potential. Whatever the goal and its complexity, there are multiple ways of achieving it. We must now establish a strategic and effective plan that requires the minimum number of steps, but lends itself to widespread use. This book is structured around the study of a dozen target products (butyrolactone, macrolide, indole compound, cyclobutanic terpene, spiro- and polycyclic derivatives, etc.). For each product, the different disconnections are presented and the associated syntheses are analyzed step by step. The key reactions are described explicitly, followed by diagrams showing the range of impact of certain transformations. This set of data alone is conducive to understanding syntheses and indulging in this difficult, but worthwhile activity.

Download or read book Drug Discovery and Design Using Natural Products written by Jorddy Neves Cruz and published by Springer Nature. This book was released on 2023-08-28 with total page 541 pages. Available in PDF, EPUB and Kindle. Book excerpt: Historically, natural products have great relevance for the development of new drugs. Natural molecules, in addition to directly providing new compounds with different therapeutic functions have provided scaffolds and a great diversity of structures that serve as inspiration for the design and discovery of new drugs. The great structural complexity and the significant number of natural compounds that can be obtained and their advantages also present some historical problems that need to be overcome. Issues such as difficulty in isolation, difficulty in synthesizing compounds found in nature in the laboratory, screening to evaluate the biological properties of these compounds, and structural optimization to enhance the pharmacological properties of biomolecules; are some of the main difficulties encountered in research with natural products and in the design/discovery of new medicines. Therefore, this book addresses the great scientific advances that provide practical solutions to using natural products to develop new drugs. We address topics related to the genesis of research projects that use natural products to design new drugs. We address the discovery of new species, initial processes for identification and extraction of compounds, isolation and identification of these structures, techniques, and advances in vitro and in vivo methods that support the discovery of new natural drugs, synthesis of new compounds from natural molecules, in silico optimization processes of potentially promising compounds.
