Book excerpt: Samples of sound, untreated enamel and enamel subjected to a number of different treatment procedures were selected for this study, to determine the potential of the scanning electron micro-scope as applied to the study of dental tissue. Sound enamel specimens were treated with acetate buffer (pH 4.0) and with EDTA (pH 7.94) for varying periods of time. The results were compared to those obtained from sound, untreated enamel specimens. Surface and sub-surface enamel morphology and bacteria-like structures were examined, photographed at magnifications ranging from 20X to over 100,000X, and described in relation to the ultra-structural morphology of enamel. Structures resembling enamel prisms, the 'prism-less' zone of surface enamel, the surface covering of an 'enamel skin' and sequential patterns of acid demineralization occuring preferentially down prism cores are described and illustrated. Comparison with EDTA-treated specimens revealed peripheral dissolution of enamel rods, in contrast to the acid effects. Organic surface coverings suggestive of dental plaque and microbial forms resembling cocci and lactobacilli were also shown. (Author).

Book excerpt: Samples of sound dental enamel were treated with topical applications of a 10% solution of stannous fluoride and then demineralized using acetate buffer (pH 4.0) and EDTA (pH 8.0) for varying periods of time. Nonfluoridated enamel treated for equivalent periods with the acetate buffer and with the EDTA, as well as untreated enamel, served as controls. A scanning electron microscope comparison of the surface alterations produced within the different groups revealed that: the in-virto application of stannous fluoride solutions inhibited enamel surface alterations when challenged by an acid or chelating agent. This finding adds further support to the concept that fluoride protection involves a reduction in enamel solubility. In addition, there was suggestive evidence that fluorides possibly play a role in the remineralization or repair or early etched enamel defects by the formation of a fluoride complex which may involve the less mineralized enamel matrix components. Book excerpt: SEM studies were performed on 25 deciduous and permanent teeth from members of seven kindreds with autosomal dominant osteogenesis imperfecta (O.I.). Two families had normal teeth on clinical and radiological examination; five families had blue or brown opalescent teeth with specific radiologic findings. Enamel surfaces and prism organization were normal on all teeth. On fractured surfaces, the dentin tubules of normal teeth from patients with O.I. were evenly distributed and coursed regularly to the dentin-enamel junction. Opalescent teeth had few tubules; those tubules present were narrow, short, and tortuous. Dentin calcification fronts of normal teeth were composed of many mushroom shaped nodules, with regularly spaced openings on their surfaces. Calcification fronts of opalescent teeth were composed of irregularly spaced, small nodules, which varied greatly in size one from another; the nodules lacked tubule openings on their surfaces. The results of this study support the concept that two autosomal dominant O.I. syndromes exist, one in which all individuals with O.I. have normal teeth, and the other in which all with O.I. have blue or brown opalescent teeth.
