Book excerpt: Scientific Frontiers in Developmental Toxicology and Risk Assessment reviews advances made during the last 10-15 years in fields such as developmental biology, molecular biology, and genetics. It describes a novel approach for how these advances might be used in combination with existing methodologies to further the understanding of mechanisms of developmental toxicity, to improve the assessment of chemicals for their ability to cause developmental toxicity, and to improve risk assessment for developmental defects. For example, based on the recent advances, even the smallest, simplest laboratory animals such as the fruit fly, roundworm, and zebrafish might be able to serve as developmental toxicological models for human biological systems. Use of such organisms might allow for rapid and inexpensive testing of large numbers of chemicals for their potential to cause developmental toxicity; presently, there are little or no developmental toxicity data available for the majority of natural and manufactured chemicals in use. This new approach to developmental toxicology and risk assessment will require simultaneous research on several fronts by experts from multiple scientific disciplines, including developmental toxicologists, developmental biologists, geneticists, epidemiologists, and biostatisticians.

This book highlights the known associations between kynurenine pathway and various disease states, as well as examines the current status of drug development and clinical trials of compounds known to alter tryptophan metabolism. The research plays a critical role in molecular targeted therapies directed at altering the kynurenine pathway of tryptophan metabolism. The initial and rate-limiting step of tryptophan metabolism is mediated by one of two enzymes, tryptophan-2,3-dioxygenase (TDO; predominantly in the liver, but also in the brain) and indoleamine-2,3-dioxygenase (IDO; in a host of tissues in response to immune activation). Targeting the enzymes IDO and TDO, as well as other downstream effectors would therefore be likely to generate novel treatment options that would be helpful in a wide variety of clinical settings. This book provides a unique bridge between basic mechanistic understanding of the role of the kynurenine pathway with translational applications and clinical relevance. It will explore the indications that tryptophan metabolism is a potential biomarker of disease activity, can contribute to local and possibly systemic immune suppression in cancer, and is an attractive target for which a variety of inhibitors are readily available.

However, in the early 1500s, it was apparent to Paracelsus that "the dose differentiates a poison and a remedy". Clearly, the two most important tenets of toxicology were established during that time. The level of exposure (dose) and the duration of exposure (time) will determine the degree and nature of a toxicological response. Since that time the discipline of toxicology has made major advances in identifying and characterizing toxicants. The growth of toxicology as a scientific discipline has been driven to a large extent by the use of extremely powerful molecular and cell biology techniques. The overall aim of this volume is to demonstrate how these advances are being used to elucidate causal pathways (or linkages) for potential adverse health consequences of human exposure to environmental chemicals or radiation. A unique feature of this volume is its illustration of how carefully-designed studies of the molecular mechanisms of chemical action provide not only understanding of the potential toxicity of the chemical under investigation, but also new insights into the functioning of the biological system used as an experimental model. Each chapter contains a listing of major peer-reviewed articles and reviews and useful web-sites. In addition, each chapter contains a broad introductory section that outlines the subsequent sections. Bitumens are produced by distillation of crude oil during petroleum refining, and also occur naturally. Bitumens can be divided into six broad classes, according to their physical properties and specifications required for different applications. The major use (about 80%) of bitumens is for road paving; other uses include roofing, waterproofing, sealing and painting. The term "bitumen" should not be confused with "asphalt", which refers to the mixture of bitumen (4-10% by weight), small stones, sand and filler used for road paving. Bitumens are complex mixtures that contain a large number of organic chemical compounds. Application of bitumens may generate emissions (fumes and vapours) that may contain, among volatile and non-volatile compounds, a number of known or probable carcinogens. An IARC Monographs Working Group reviewed epidemiological evidence, animal bioassays, and mechanistic and other relevant data to reach conclusions as to the carcinogenic hazard to humans of various occupations that entail exposure to bitumens and bitumen emissions, including road paving, roofing, and application of mastic asphalt, and to various heterocyclic polycyclic aromatic compounds.

These include physical and chemical pollutants as well as other stressors such as parasites and environmental impact (e. g. , climate change or habitat loss). The combi- tion of stressors can result in increased risk to organisms (either additive or synergistic effects) or decreased effects (protective or antagonistic effects). The multiple stressor challenge is an international, multi-disciplinary problem requiring an international, multi-disciplinary approach. The c- rent approach to multiple stressors is to examine one stressor at a time and assume additivity. Little work has been done on combinations of stressors such that potential interactions can be determined. The problem is very complex. Multiple stressors pose a whole spectrum of challenges that range from basic science to regulation, policy and gove- ance. The challenges raise fundamental questions about our understanding of the basic biological response to stressors, as well as the implications of those uncertainties in environmental risk assessment and management. In addition to the great breadth, there is also great depth in the research ch- lenges, largely due to the complexity of the issues. From a basic science point of view, many of the mechanisms and processes under investigation are at the cutting edge of science — involving new paradigms such as genomic ins- bility and bystander effects.

Book excerpt: Early in 1985, a meeting was held to discuss the organization of a work shop in honour of Professor Reinier L. Zielhuis on the occasion of his retirement on November 30,1986. Various themes for such a workshop were considered, but it was Zielhuis himself, who ultimately selected "Health surveillance of individual workers exposed to chemical agents" as the theme of the workshop. Although this topic is of the utmost im portance for occupational health practice, it is also a very difficult one, given our currently limited knowledge on the subject. The choice of this topic is characteristic of Reinier Zielhuis, a scientist and scholar who throughout his career has been deeply concerned with the health and health risks of workers. Because he regards occupational and environmental exposure - including life style - as two aspects of the total exposure to chemical and physical factors, he emphasizes the need to assess the impact of this total exposure on workers' health. The impact of total exposure probably detennines to a large extent the in dividual variability in response. This was the main reason why he selected the above-mentioned topic for the workshop.
