Wird aber die Phenolhydroxylgruppe des Morphins in der Weise geschlossen, dass eine Absprengung der dem Hydroxyl substituierten Gruppe im Organismus nicht zu stande kommt, so kann die aus dem Morphin dargestellte Substanz auch vollig jeder Wirkung entbehren. So ist auch die Morphinschwefelsaure, selbst in Dosen von 5 g, unwirksam oder bei einzelnen Tieren im Sinne der Kodeingruppe sehr schwach wirksam, so ist auch das Oxydimorphin (der Morphinather des Morphins), das sogenannte Pseudomorphin (C17 Hlg NOa)2l unwirksam, dessen Bau man sich durch Substitution des Hydroxylwasserstoffes im Morphin durch ein Morphinradikal vergegenwartigen kann; so ist auch Athylenmorphin unwirksam, eine Verbindung, in welcher zwei Molekule Morphin durch Athylenbindung der beiden Phenolhydroxyle miteinander verbunden sind (v. Meringj. Tatsachen dieser Art ') mussen zu der Annahme fuhren, dass zum Zustandekommen einer Wirkung, ausser der Gegenwart einer wirksamen Gruppe, noch eine zweite Gruppe notwendig ist, welche der Substanz J) Man findet diese Ableitungen, sowie viele Beispiele solcher Art naher ausgefuhrt in meinem Buche: Arzneimittelsynthese auf Grundlage der Beziehungen zwischen Konstitution und Wirkung, Berlin bei Springer 1901. solche Eigenschaften verleiht, dass sie mit dem reaktionsfahigen Gewebe sich verbinden, von diesem festgehalten werden kann und dann die wirksame Gruppierung zur physiologischen Wirkung gelangt. Wir deuten dabei das Zustandekommen der Wirkung chemischer Substanzen auf bestimmte Zellgruppen, also die selektive Wirkung neben physikalischen (Losungsverhaltnissen, welche sicherlich mit eine Rolle spielen) so, dass gewisse endstandige Gruppen im Molekul in chemische Beziehungen zu Zellsubstanzen treten, von ihnen festgehalten werden; erst dann kann der ganze Molekularkomplex, einmal im bestimmten Gewebe oder in einer bestimmten Zellgruppe festgehalten (verankert), zur Wirkung gelangen, indem nun nach dem Festhalten ihr partieller Abbau stattfindet. Andern wir nun den Charakter einer solchen endstandigen Gruppe, oder fuhren wir eine solche chemisch reaktionsfahigere als die schon vorhandene ein, so wird die Bindung der eingefuhrten chemischen Substanz an den bestimmten Zellkomplex nicht stattfinden. Die Substanz wird von der betreffenden Zellgruppe nicht mehr festgehalten und kann daher auch nicht mehr zur Wirkung gelangen, wenn auch die eigentlich wirksame Gruppe vollig intakt geblieben. Wir werden dem Verstandnis des Zustandekommens der Wirkung am besten naherkommen, wenn wir annehmen, dass zum Zustandekommen der Wirkung mehrere Faktoren beitragen und an dieser beteiligt sind. Vorerst ein wirksamer Korper, welcher aber an sich noch keine Wirkung zu entfalten braucht, aber sie schon an und fur sich entfalten kann.
