Book excerpt: The B. subtilis catabolic acetolactate synthase enzyme (EC4.1.3.18) catalyzes the condensation reaction between 2 pyruvate molecules to form a molecule of acetolactate and CO2. It has a pH optimum of 6.0 and is primarily involved in acetoin production. It requires thiamine pyrophosphate (TPP) and Mg2+ for its reaction. The activity of this enzyme can only be detected during stationary phase in Bacillus subtilis. The acetolactate that it produce is decarboxylated to form acetoin. This enzyme is coded by a1sS gene. A plasmid bearing the wild-type alsS gene was able to allow an E. coli strain deficient in all three AHAS homologs to grow in the absence of valine. This transformat still required isoleucine. In order to study how alsS is regulated, we decided to sequence the 3.7 kb of B. subtilis DNA that contains the gene. [Authors' abstract].
