For some time however, there has been a need for the collection in one volume of information from individual experts actively engaged in different aspects of prostaglandin research. This book is an attempt to fulfil that need. In view of the expanding interest and ever-growing accumulation of data of the various effects of prostaglandins it is hoped to revise the book and bring it up-to-date periodically. The largest section is that on reproduction, in which field progress in research and clinical applications thereof have been, and are, of great significance particularly in the area of popula tion control. This is not to decry the skill and efforts of investi gators in other aspects of prostaglandin activity nor the potential biological or therapeutic importance of their findings. In this context, what may appear to be omissions are individual sections on prostaglandin activity in the central nervous system, in the eye and as mediators of the inflammatory response. These are considered in the chapter on General Pharmacology of the Prostaglandins not because they might be considered to be of lesser impact and significance but in order to achieve a rapid publication of the book. Book excerpt: There is much evidence to suggest that prostaglandins may have a physiological role to play in the control of thyroid gland metabolism, although the precise nature of this role remains to be defined. Prostaglandins have been found in normal thyroid tissue, and there is evidence of increased levels in some, but not all, cases of medullary carcinoma of the thyroid. which may account for the associated diarrhea. Exogenous prostaglandins, like TSH, bind to thyroid plasma mem branes, stimulate the adenylate cyclase-cyclic AMP system, thyroid hormone secretion, iodide organification, colloid droplet formation, glucose oxidation, and 32p incorporation into phospholipids. Prosta glandins of the E series are the most potent, although their maximal effects are generally less than the maximal effects of TSH. The interrelationships existing between TSH and prostaglandins have been the subject of intensive investigation, but as yet no one uni fying concept has emerged. TSH and prostaglandins have been shown to occupy different receptors on the thyroid plasma membrane. Al though TSH can increase intracellular prostaglandin levels, probably as a result of increased synthesis, prostaglandins do not appear to be essential intermediates in TSH action on adenylate cyclase. The pos sibility that prostaglandins are part of a negative feedback system con trolling TSH action is, as yet, a tentative hypothesis, and further work will be required to unravel the interrelationships between these sub stances that now appear to be far more complex than has been envisaged thus far.

In fact small meetings where in depth discussion is confined to a limited number of scientists, are now generally considered successful vehicles of scientific information. With this in mind, we made an effort in collecting a number of experts in both experimental and clinical use of these important arachidonate metabolites. We have been very fortunate in bringing together a faculty of international authorities who contributed to the course with brilliant presentations and lively discussion. Thus, this book represents an up-to-date overview of the physiology and pharmacology of different products derived from arachidonic acid metabolism, associated with outstanding suggestions of their clinical potential. It is our hope that from this Advanced Course, and its resulting publication, new ideas concerning the physio pathological role of these compounds will arise together with new directions for future research. Most of the information on practical applications of prostaglandins is scattered in different journals. Although a number of books dealing with prostaglandin research have been published (including some on practical applications in specific areas, e.g. reproduction) there is no single volume dealing with all areas of practical applications of these substances. This book aims at filling this gap and should be of interest to clinicians as well as to medical and basic scientists in academic institutions and in the pharmaceutical industry. In some areas, exciting potential applications of prostaglandins have been identified from basic research but clinical trials have so far not been carried out. Such areas of practical applications are covered in a separate intro ductory chapter. All contributors to the volume are scientists and clinicians actively engaged in prostaglandin research and most are recognized authorities in specific areas. The need for rapid publication in a fast-expanding field is obvious.

Book excerpt: The International Symposium on Prostaglandins and Related Compounds, first held in Vienna 1972, revisited the city after 24 years for the 10TH Symposium. For the many re searchers working in this multi-disciplinary field it was an opportunity to exchange their ex periences and share new data with colleagues from all around the world. This scientific exchange was largely encouraged by the unseasonably cold and rainy weather. For the first time, there was quite a large attendance from the former Communist countries. Eugene Garfield prepared a key note address delivered during the meeting (The Sci entist 1996, 12) reviewing the contribution of the Nobel Laureates U.S. von Euler, l.R. Vane, S.K. Bergstrom, and B.I. Samuelsson, discussing the relevance of the more than 40,000 pa pers in this area published since 1991. Overall, there is still a rapidly growing interest, and in particular a great variety of clinical applications of this family of compounds which were dis cussed in detail during the meeting. Beside the lectures there were 19 workshops covering nearly all the topics of key in terest. All the speakers were invited to prepare a manuscript which has resulted in the volume now in your hands. Special thanks to Dr. Patrick Wong and the new publisher of this series who helped to publish the proceedings in the usual quality and reasonable period of time. Looking forward to seeing all of you again in Florence in 1999, hopefully with much more sun.

Williams first reported that elevated prostaglandin levels are present in human medullary car­ cinoma. Since that time, the concept that arachidonic acid metabolites may be in­ volved in cancer has expanded to include every aspect of the disease from cell transformation through metastasis. Prostaglandins and leukotrienes are generic terms used to describe a family of bioactive lipids produced from unsaturated fatty acids (principally from arachidonic acid) via the cyclooxygenase and lipoxygenase pathways, respec­ tively. Cyclooxygenase products consist of diverse products such as prosta­ glandin E2 (PGE2), prostacyclin (PGI2) and thromboxane A2 (TXA2), whereas lipoxygenase products consist of hydroperoxy fatty acids and mono-, di-and tri-hydroxy acids including leukotrienes. The precursor fatty acids for the cyclooxygenase and lipoxygenase pathways are present in cellular phospholipids. This finding established an important control point in their biosynthesis-the release of substrate. This occurs in response to numerous stimuli that act at the cell surface. Dr. Bengt Samuelsson's extensive study of the metabolism of pros­ taglandins indicated that they are rapidly inactivated on a single pass through pulmonary circulation. Thus, they cannot act as circulating hormones and appear to be made on demand in or in the vicinity of target tissues leading to the concept that prostaglandins are local hormones or autocoids.

In the last few years there has been an extraordinary evolution of data on prostaglandins (and related compounds) and cancer. This vol ume is based on papers presented at the 1986 International Confer ence on Prostaglandins and Cancer organized by the Italian National Research Council and the II University of Rome, and held in Rome, Italy, in June, 1986. This Conference brought together oncologists and specialists in the areas of prostaglandin chemistry, biochemistry, pharmacology, physiology, cellular and molecular biology to overview the actual state of knowledge on the role of eicosanoids in cancer and to focus on the key questions that need to be answered. The picture that comes out of this book describes a very complicated network of interactions between arachidonic acid metabolites and different as pects of the complex phenomenon "cancer". Eicosanoids participate in carcinogenesis initiation and promotion, and their relationship with tumor promoters and growth factors is well established. During cancer growth, different prostaglandins can have different roles in the regulation of cell proliferation and differentiation and in metastasis formation; meanwhile evidence is accumulating for a pos sible use of some of these compounds as antineoplastic agents.
