Book excerpt: Lipase is an extracellular enzyme in food industry and is produced by different microbial and fungal sources by submerged and solid state fermentation process. In the present study, Bacillus subtilis was employed for the production of lipase in peptone broth with olive oil as inducer. Maximum lipase production was observed at 96 hours of incubation and NH4Cl (40.75 U/ml) as salt and peptone (24U/ml) as nitrogen source and olive oil (24 U/ml) as inducer. The results of the present study demonstrate that Bacillus subtilis is ideal for extracellular production at industrial level.

A lipase produced from thermophilic bacteria is revised to have the advantage of thermal stability at 65°C-70°C, which makes them good candidates for lipid modifications which is most effective at high temperature. Thus, main focus of this research is to find the best lipase producing thermotolerant bacteria on its optimum condition of fermentation. The first step on finding lipase producer is to screen the isolated thermotolerant bacterial strains on tributyrin agar plate to check for zone of hydrolysis. Then, followed by screening under submerged fermentation. Next, lipase assay was carried out to find out the best lipase producer among the strains and finally the optimum condition for producing lipase investigated.

Book excerpt: Lipases comprise a group of hydrolytic enzymes which catalyze the hydrolysis and synthesis of triacylglycerides in the oil water interface. Screening of microorganism for extracellular lipase production from coconut dregs was conducted by using Aspergillus niger and Rhizopus oligosporus. The result shows that Rhizopus oligosporus able to produce higher lipase, 0.1527 U/mL compared to Aspergillus niger and thus it was chosen for further analysis. Process conditions such as pH, temperature and moisture content which influence the growth and production of lipase were studied. Central-Composite Design (CCD) of Response Surface approach was used to analyze the results and the optimum conditions for lipase production was determined. According to CCD, the optimum process conditions for Rhizopus oligosporus to produce lipase were as follow: 34.8 C, pH 5.24 and 62 % of moisture content. Validation process was carried out for 10 days under these conditions and it gives the maximum lipase activity at day 6 (0.2281 U/mL) . The standard error between optimization and validation was 2.23 %.

Available in PDF, EPUB and Kindle. Book excerpt: Production of extracellular protease refers to proteases produced from microorganisms and functions out of the cell in which it was originated. In this term, the microorganism was sought to be normal mesophilic bacteria. The experiment was conducted to evaluate the effects of temperature and agitation speed of the incubator shaker towards the production of extracellular protease. Optimization of the enzyme production condition involving the two parameters was also made using statistical software. Typical methods conducted in previous researches were utilized in the experiments using different temperature and agitation speed. However, in previous researches, experiments were conducted through one-factor-at-a-time method (OF AT) but in this experiment, that method was overrun and designed beforehand using Minitab. The standard procedures of protease assay were carried out when experimenting. The bacteria were re-cultured to enable its functionality to serve its purpose. The results obtained proved that both parameters had significant effects if stood alone towards the production, however had to interact with each other due to randomization of the values implemented in the experiments. Simple analysis was not an option therefore the results were opted to be analyzed by Minitab and also the condition of protease production was optimized.

In fact, this process is very old. In older industries such the food and agricultural, this technique has been extensively used. An example of this is the production of pork sausages and Roquefort cheese. Pharmaceutical industry could make extensive use of SSF in the production of secondary metabolites of many kinds and development in this direction is soon expected.
