The macromolecules involved in translation, ribosomes, RNAs and protein factors, as well as the mechanisms of protein synthesis, are all largely conserved across the spectrum of cell types. In this thesis, a ribosome-associated ATPase, RbbA, is identified in 'Escherichia coli' extracts. RbbA can bind both 70S ribosomes and 30S ribosomal subunits. The ATPase activity of RbbA increases two-fold in the presence of purified 70S ribosomes. RbbA cross-reacts with antibody raised against the fungal-specific translation factor EF-3, and like EF-3, RbbA stimulates the synthesis of polyphenylalanine from a poly(U) template 'in vitro'. Cross-linking data show that RbbA lies near 30S ribosomal subunit protein S1 and binds to a poly(G)-rich region of the 30S subunit rRNA, 16S rRNA. This region of the ribosome is known as the decoding region of the ribosome and is important for the binding of aminoacyl-tRNAs to the ribosomal A-site. Upon binding of RbbA, 16S rRNA undergoes a specific conformational change that has been demonstrated previously to affect translational fidelity. EF-Tu, which escorts aminoacyl-tRNAs to the A-site of the ribosome, is also capable of binding with RbbA. Hygromycin, an antibiotic that interferes with protein synthesis by binding to the decoding center of the ribosome, is able specifically to inhibit the ribosomal ATPase activity by displacing RbbA from the ribosome. Together, the data suggest that RbbA, the first ATPase demonstrated to be involved in prokaryotic protein synthesis, plays a major role in protein chain elongation.

Book excerpt: The motivation for us to produce a treatise on regulation was mainly our convic tion that it would be fun, and at the same time productive, to approach the subject in a way that differs from that of other treatises. We had ourselves written reviews for various volumes over the years, most of them bringing together all possible facts relevant to a particular operon, virus, or biosynthetic system. And we were not convinced of the value of such reviews for anyone but the expert in the field reviewed. We thought it might be more interesting and more instructive-for both author and reader-to avoid reviewing topics that anyone scientist might work on, but instead to review the various parts of what many different scientists work on. Cutting across the traditional boundaries that have separated the subjects in past volumes on regulation is not an easy thing to do-not because it is difficult to think of what interesting topics should replace the old ones, but because it is difficult to find authors who possess sufficient breadth of knowledge and who are willing to write about areas outside those pursued in their own laboratories. For example, no one scientist works on suppression per se. He may study the structure of suppressor tRNAs in Escherichia coli, he may study phenotypic suppression of various characters in drosophila, he may study polarity in gene expression, and so on.

Book excerpt: The literature on recoding is scattered, so this superb book ?lls a need by prov- ing up-to-date, comprehensive, authoritative reviews of the many kinds of recoding phenomena. Between 1961 and 1966 my colleagues and I deciphered the genetic code in Escherichia coli and showed that the genetic code is the same in E. coli, Xenopus laevis, and guinea pig tissues. These results showed that the code has been c- served during evolution and strongly suggested that the code appeared very early during biological evolution, that all forms of life on earth descended from a c- mon ancestor, and thus that all forms of life on this planet are related to one another. The problem of biological time was solved by encoding information in DNA and retrieving the information for each new generation, for it is easier to make a new organism than it is to repair an aging, malfunctioning one. Subsequently, small modi?cations of the standard genetic code were found in certain organisms and in mitochondria. Mitochondrial DNA only encodes about 10–13 proteins, so some modi?cations of the genetic code are tolerated that pr- ably would be lethal if applied to the thousands of kinds of proteins encoded by genomic DNA.
