Book excerpt: The goal of this work is to develop novel functionalized block copolymers to promote adhesion at inorganic substrate/polymer interfaces. We envision several potential advantages of functionalized block copolymers over small molecule coupling agents. Greater control over the structure of the interphase region should result through careful design of the backbone of the copolymer. The number of chains per area, the degree of entanglement between the copolymer and the polymer matrix, the number of sites per chain able to attach to the substrate, and the hydrophobicity of the interphase region can all be strongly affected by the choice of block lengths and the monomer sequence. In addition, entanglement between the copolymer and the polymer matrix, if achieved, should contribute significantly to adhesive strength. Our program involves four key elements: the synthesis of suitable functionalized block copolymers, characterization of the conformation of the copolymers at the interface by neutron reflectivity and atomic force microscopy, characterization of the degree of bonding by spectroscopy, and measurement of the mechanical properties of the interface. In this paper we discuss block copolymers designed as adhesion promoters for the copper/epoxy interface. We have synthesized a diblock with one block containing imidazole groups to bond to copper and a second block containing secondary amines to react with the epoxy matrix. We have also prepared a triblock copolymer containing a hydrophobic middle block. Below we describe the synthesis of the block copolymers by living, ring-opening metathesis polymerization (ROMP) and the first characterization data obtained by neutron reflectivity.

Fiberglass-reinforced polymer com posites, similarly, have two (apparently) insurmountable obstacles to per formance: 1) Water can hydrolyze any conceivable bond between organic and inorganic phase, and 2) Stresses across the interface during temperature cycling (resulting from a mismatch in thermal expansion coefficients) may exceed the strength of one of the phases. Organofunctional silanes are hybrid organic-inorganic compounds that are used as coupling agents across the organic-inorganic interface to help overcome these two obstacles to composite performance. One of their functions is to use the hydrolytic action of water under equilibrium condi tions to relieve thermally induced stresses across the interface. If equilib rium conditions can be maintained, the two problems act to cancel each other out. Coupling agents are defined primarily as materials that improve the practical adhesive bond of polymer to mineral. This may involve an increase in true adhesion, but it may also involve improved wetting, rheology, and other handling properties. The coupling agent may also modify the inter phase region to strengthen the organic and inorganic boundary layers.

Book excerpt: The biophysical characteristics of cell adhesion from single protein to cell length scales have primarily been studied using purely elastic substrates. However, natural extracellular matrix (ECM) is viscoelastic and contains mobile components. In this work, we combined chemistry and cell biology tools to design and characterize laterally mobile viscoelastic polymer films that promote receptor-specific cell adhesion. Moreover, we used amphiphilic block copolymers that are end-labeled with RGD peptide ligands to allow for integrin-mediated cell adhesion. The addition of a trace hydrophobic homopolymer in the supported bilayer block-copolymer films is used to tune the lateral mobility of the films. NIH 3T3 fibroblasts demonstrate a non-linear spreading response against the mobility of the RGD-displaying polymer films. Employing immunostaining and adhesion strength assays, we decoupled the partial contributions of focal adhesions (FA) and integrin-RGD complexes on cell adhesion. Furthermore, we employed these biomimetic polymer platforms to investigate the importance of viscous dissipation within the extracellular substrate and its connection to cell-surface receptors. Our results suggest that cells preferentially use avb3 and a5b1 integrins to control spreading and polarization in response to mechanical properties of their substrate. In order to further control the spatial presentation of biochemical molecules on mechanically-tunable polymer substrates, we successfully transferred fibronectin patterns on bilayer polymer films. We showed that NIH 3T3 fibroblasts spreading and adhesion features depend on the mechanical properties of these hybrid materials even in the presence of spatially and chemically identical biochemical signals. Overall, the present work demonstrates the potential of amphiphilic block copolymers to form artificial substrates that can capture a key feature of cell-ECM interactions: specifically, the ability of cells to induce changes in the substrate over time. Furthermore, it highlights the need for future studies on cell-substrate interactions that simultaneously consider the time-dependent mechanical properties of the ECM, the spatial characteristics of ligand presentation, and the receptor-mediated intracellular signaling.
