This book gives an overview of biomedical optical imaging with contributions from leading international research groups who have pioneered many of these techniques and applications. A unique research field spanning the microscopic to the macroscopic, biomedical optical imaging allows both structural and functional imaging. Techniques such as confocal and multiphoton microscopy provide cellular level resolution imaging in biological systems. The integration of this technology with exogenous chromophores can selectively enhance contrast for molecular targets as well as supply functional information on processes such as nerve transduction. Novel techniques integrate microscopy with state-of-the-art optics technology, and these include spectral imaging, two photon fluorescence correlation, nonlinear nanoscopy; optical coherence tomography techniques allow functional, dynamic, nanoscale, and cross-sectional visualization. Moving to the macroscopic scale, spectroscopic assessment and imaging methods such as fluorescence and light scattering can provide diagnostics of tissue pathology including neoplastic changes. Techniques using light diffusion and photon migration are a means to explore processes which occur deep inside biological tissues and organs. The integration of these techniques with exogenous probes enables molecular specific sensitivity.

Book excerpt: Chemotaxis and cell migration are important processes for life, involved in organism development and homeostasis and implicated in a number of disease states. Dictyostelium discoideum, an amoeba, is a useful model for investigation of chemotaxis and development, due to its ability to undergo chemotactic aggregation and development upon starvation. Although cell migration has been well described on planar transparent surfaces, it is uncertain how well these conditions replicate the natural environment of a cell. However, attempts to better replicate these environments generally make use of opaque substrates and 3D matrices, in which it is more challenging to image cell migration. Protocols were developed to enable optical coherence tomography, a 3D structural imaging technique which requires no sample processing or staining, to be successfully employed in imaging Dictyostelium cell migration in time-lapse on non-transparent substrata and within an agarose gel. I compared the effects of two substrates, a nitrocellulose filter and a polystyrene Petri dish on aggregating cells and found differences in speed but not persistence. Extension of this to include cells within agarose revealed that these cells exhibited less directed migration, but their velocity was unaffected. I showed that cells lacking myosin II failed to complete development within an agarose gel and had significantly reduced velocity and directional migration when compared to their parent strain. Furthermore, the velocities of cells migrating within agarose gel were bimodally distributed, potentially indicating two distinct cell populations, fast and slow, and fast movement was shown to be largely myosin II dependent. Great potential therefore exists for cell-substrate and cell-matrix interactions to affect the migration character of cells, even those, such as Dictyostelium, which do not form strong focal adhesions. Moreover a properly ordered cytoskeleton is implicated in enabling cells to effectively utilise different modes of cell motility.

First, for nearly 20 years no Symposium devoted to optical methods in solids had been organized. Second, recent advances in digital image processing provided many new applications which are described in the following. We have the honour to present here the proceedings of this Symposium. st th The Symposium took place from august 31 to September 4 at the Institut International de la Prospective in Futuroscope near Poitiers. A significant number of internationally renowned specialists had expressed their wish to participate in this meeting. The Scientific Committee proposed 16 general conferences and selected 33 regular lectures and 17 poster presentations. Papers corresponding to posters are not differentiated in the proceedings from those that were presented orally. It is worth noting that a total of 80 participants, representing 16 countries, registered for this symposium.. The Scientific Committee deserves praise for attracting a significant number of young scientists, both as authors and as participants. Let us add our warm acknowledgements to Professor J.W. Dally and to Professor A.S. Kobayashi who, throughout the symposium preparation time, brought us valuable help.

Book excerpt: Optical coherence tomography (OCT) has been widely used in biomedical imaging ever since its introduction. However, the application of OCT for biometrics is relatively new. This thesis demonstrates the feasibility of applying a spectral domain optical coherence tomography (SD-OCT) system to fingertip biometric information retrieval. The results show that SD-OCT is capable of retrieving Level 3 fingertip information. Combined with traditional Level 1 and 2 fingerprint information, which can also be easily acquired by SD-OCT, the proposed application shows potential for ultra-secure scenarios. Photoacoustic microscopy (PAM) is another imaging modality gaining considerable interest in the past decade. Different than OCT, it employs ultrasound detection for optically induced thermal expansion in tissue. Bovine articulate cartilage samples are imaged with a single wavelength PAM system using India ink as a contrast agent. The same samples were also imaged with SD-OCT. Image analysis shows that PAM produces excellent contrast between intact and fibrillated regions in cartilage while SD-OCT visualizes topological changes with fine spatial resolution. A spectroscopic PAM system is also developed using a tunable source based on supercontinuum generation in a photonic crystal fiber. Imaging experiments of tissue phantoms demonstrate the ability to differentiate various chromophores with unique absorption characteristics. Another spectroscopic PAM system is currently being developed, where the tunable source is based on stimulated Raman scattering. This new system is being studied and some preliminary results are given to demonstrate its advantages over the supercontinuum approach.
