Book excerpt: Abstract: Protein phosphatase-1 and phosphatase-2A are two ubiquitously expressed enzymes known to catalyze the majority of dephosphorylation reactions on serine and threonine inside cells. They play roles in most cellular processes and are tightly regulated by regulatory subunits in holoenzymes. Their misregulation and malfunction contribute to disease development and progression, such as in cancer, diabetes, viral infections, and neurological as well as heart diseases. Therefore, targeting these phosphatases for therapeutic use would be highly desirable; however, their complex regulation and high conservation of the active site have been major hurdles for selectively targeting them in the past. In the last decade, new approaches have been developed to overcome these hurdles and have strongly revived the field. I will focus here on peptide-based approaches, which contributed to showing that these phosphatases can be targeted selectively and aided in rethinking the design of selective phosphatase modulators. Finally, I will give a perspective on www.depod.org, the human dephosphorylation database, and how it can aid phosphatase modulator design. © 2017 The Authors. Book excerpt: The importance of phosphatases in regulating intracellular signaling has recently been realized by the identification and characterization of various families of phosphatases. In particular, the protein tyrosine phosphatases occupy a prominent role in regulating protein tyrosine kinase mediated signaling events. In this regard, the SH2 domain containing cytosolic protein tyrosine phosphatase SHP-1 has been implicated in the downregulation of signaling cascades in haemopoietic cells. Herein, we report that SHP-1 can directly regulate the Src protein tyrosine kinase p60 c-Src. We have demonstrated that Src isolated from platelets and T cells is preferentially dephosphorylated at its inhibitory phosphotyrosine site by the SHP-1 tyrosine phosphatase. This represents the first biochemical evidence that SHP-1 can also function as a positive signal regulator. Furthermore, we have also tried to elucidate the role of SHP-1 in the signaling of the surface comodulators CD19 and CD5, on B and T cells respectively. With respect to SHP-1 and CD19 signaling, we demonstrate an indirect role for SHP-1 and present data revealing the capacity of SHP-1 to dephosphorylate the Lyn autophosphorylation site. This data provides strong evidence that SHP-1 functions also include the negative regulation of the Lyn kinase activation in B cells. Finally, given the recent biochemical evidence for CD5 and SHP-1 interaction, we investigated the biochemical and physiological relevance of the two signaling proteins in the context of T cell development and activation. By utilizing HY-TCR and HY-TCR CD5 over-expressing transgenic mice crossed with the SHP-1 deficient motheaten mice, we first examined the role of SHP-1 in T cell development. Herein we report that SHP-1 function also includes the down regulation of both positive and negative T cell selection. With respect to CD5, we report that although CD5 can down modulate the T cell selection process, our findings suggest that this can be achieved independent of SHP-1. Taken together, these studies provide novel insights into the mechanisms whereby SHP-1 modulates antigen receptor signaling. SHP-1, by virtue of its ability to regulate protein tyrosine kinase activity and membrane co-receptors represents a protein tyrosine phosphatase occupying a pivotal role in the translation of antigen receptor signals to such biological outcomes as proliferation, transformation, and selection.

A short account of the history of the effort led to the Phosphate Workshops is appro priate and can be of interest to the reader. The idea for Phosphate Workshops was born in the early days of November, 1974. One of us (S. G. M. ) suggested the thought to a group of scientists gathered for a luncheon in one of the attrac tive small restaurants in Weisbaden, Germany. The purpose of the workshop was to bring together interested scientists to discuss the newer developments and the recent advances in the field of phosphate metabolism and the other related minerals. An Organizing Committee made of Shaul G. Massry (USA), Louis V. Avioli (USA), Philippe Bordier (France), Herbert Fleisch (Switzerland), and Eduardo Slatopolsky (USA) was formed. The First Workshop was held in Paris during June 5-6, 1975 and was hosted by Dr. Philippe Bordier. Its proceeding was already published. The Second Workshop took place in Heidelberg during June 28-30, 1976 and was hosted by Dr. Eberhard Ritz. Both of these workshops were extremely successful scientific endeavors, and the need for them was demonstrated by the great interest they generated among the scientific community. The Or ganizing Committee, therefore, decided to continue with the tradi tion to hold additional Workshops annually or every other year.
