Book excerpt: In an attempt to isolate a temperature sensitive allele of ftsN, chemical random mutagenesis was performed using hydroxylamine. SMM 12 a temperature sensitive filamentous strain was successfully isolated. However, it did not carry an ftsN(Ts) allele. Transforming with different pasmids carrying cloned genes and random genomic plasmid libraries have yielded no complementation. Using P I -mediated transduction, the location of the mutation was found to be at the 88-89 minute region on the E. coli chromosome. This finding represents the discovery of an unprecedented gene at the 88- 89 minute region that blocks cell division. in addition, we report in this work the isolation of the first temperature-sensitive mutant that is suppressed by L-glutamate. Although the precise identity of the mutated gene remains illusive, all data produced by this work point out that we may have isolated a previously unknown player in the cell- division protein machinery in E. coli.

Book excerpt: The normally rod-shaped bacterium Escherichia coli K-12 divides by elongating and then forming a septum at midcell, which will result in the format ion of two daughter cells. Septation is driven by the polymerization of the essential d ivision protein FtsZ into a structure called the Z-ring, and the subsequent recruitment of other cell division proteins to form the divisome. Temperature-sensitive (Ts) ftsZ mut ants can divide as rods at 30 degreesC but filament and lyse at 42 degreesC, and are ther efore used as tools to study the role of FtsZ in cell division. The rod shape of E. coli is maintained by the pbpA gene product Penicillin-Binding Protein 2 (PBP2), and upon inactivation of pbpA the cells become spherical, fail to divide, and eventually die. Simultaneous overexpression of the essential cell division genes ftsQ, ftsA and ftsZ from the lowcopy- number plasmid pBS58 restores division in spherical cells on LB agar 30 degreesC and 37 degreesC but not 42 degreesC. Several temperature-sensitive (Ts) ftsZ mutants were tested for their ability to support division in spheres, with the mutation being either on the chromosome, o n a pBS58 derivative, or both. At the permissive temperature of 30degreesC, ftsZ84 s upported division in all combinations. On the other hand, ftsZ972, ftsZ2066, ftsZ2863, fts Z6460 and ftsZ9124 supported division and viability in spherical cells only when prese nt in combination with wild-type ftsZ. Furthermore, an ftsZ26 mutant carrying pBS58 co uld divide more efficiently as a sphere than a rod. Overexpression of ftsWin additio n to ftsQAZ recovered spherical division in ftsZ972 but had no effect on the other mu tants. Surprisingly, the ftsZ26, ftsZ84, ftsZ2863 and ftsZ6460 mutations in combination with wild-type ftsZ supported division in spheres at the non-permissive temperature. These data demonstrate that in addition to conferring temperature-sensitivity, some ft sZ(Ts) mutants impair cell division lethally at the permissive temperature in spherical but not rod-shaped E. coli, and a few of these as well as other mutants suppress the dea th of spherical cells at the non-permissive temperature. Darwin emphasized "many slight differences" as the ultimate source of variation to be acted upon by natural selection. In the early 1900’s, this view was opposed by "Mendelian geneticists", who emphasized the importance of "macromutations" in evolution. The Modern Synthesis resolved this controversy, concluding that mutations in genes of very small effect were responsible for adaptive evolution. A decade ago, Allen Orr and Jerry Coyne reexamined the evidence for this neo-Darwinian view and found that both the theoretical and empirical basis for it were weak. Orr and Coyne encouraged evolutionary biologists to reexamine this neglected question: what is the genetic basis of adaptive evolution?
